    Sequential SEM imaging of microbial calcite precipitation consolidation treatment

    Cultural heritage built from limestone is prone to deterioration by chemical weathering, a natural process, that is enhanced by pollution. There are many historic monuments built from calcareous rocks that suffer from deterioration, and thus there have been a number of approaches over the last few decades to consolidate these types of rocks and surfaces. Using natural biological processes by fostering the activity of calcite-producing bacteria, also referred to as biomineralization, is one strategy that has also been commercialized. The base of proving the effectiveness of any surface treatment is the observation of the surface at sequential stages before and after treatment, as well as after exposure to weathering. Due to the heterogeneity of natural materials and processes, our aim was to observe identical test areas at the micron scale throughout the observation period. In order to achieve this on a tungsten SEM, we employed a beam deceleration accessory that allowed low kV imaging on non-conductive surfaces at a sufficiently high image resolution with a modified sample holder accommodating drill cores of 25 mm diameter and up to 15 mm height. The presented method is capable of producing time-sequenced images on the same test area on natural rock surface samples without manipulation for imaging purposes. This offers interesting perspectives for effective documentation of such processes in various fields

    Time resolved XANES illustrates a substrate-mediated redox process in Prussian blue cultural heritage materials

    The pigment Prussian blue is studied in heritage science because of its capricious fading behavior under light exposure. We show here that XANES can be used to study the photosensitivity of Prussian blue heritage materials despite X-ray radiation damage. We used an original approach based on X-ray photochemistry to investigate in depth the redox process of Prussian blue when it is associated with a cellulosic substrate, as in cyanotypes and watercolors. By modifying cation and proton contents of the paper substrate, we could tune both rate and extent of Prussian blue reduction. These results demonstrate that the photoreduction and fading of Prussian blue is principally mediated by the substrate and its interaction with the oxygen of the environment

    Comparison of determinants of research knowledge utilization by practitioners and administrators in the field of child and family social services

    <p>Abstract</p> <p>Background</p> <p>An important gap exists between research production and its utilization. Few studies have examined the factors affecting knowledge utilization in the field of child and family social services.</p> <p>Methods</p> <p>The objectives of the study are to describe knowledge utilization by child protection administrators and practitioners (N = 477) and to compare factors related to knowledge utilization by these two occupational groups. The study was conducted with an adapted version of the <it>Questionnaire sur l'utilisation des connaissances </it>(Knowledge Utilization Questionnaire). Factor analysis was used to collapse data collected on the questionnaire items. Factor score for each respondent served as independent variables in three separate multivariate regression analyses to explore variables likely to predict research-based knowledge utilization.</p> <p>Results</p> <p>A minority of respondents (18%) report using on a frequent basis research-based knowledge in their practice. Relational capital between researchers and users and perceived usefulness of research based knowledge were the two factors most strongly related to utilization. There was a specificity in the factors associated with knowledge utilization according to occupational groups in child protection organizations. Use of active knowledge transfer strategies was associated with knowledge utilization by practitioners, while knowledge dissemination efforts played a more significant role for administrators.</p> <p>Conclusion</p> <p>These results encourage both the use of strategies differentiated according to users and the intensification of interactions between users and researchers to foster research knowledge utilization.</p

    Light and anoxia fading of Prussian blue dyed textiles.

    Although Prussian blue is a popular pigment, its stability has been questioned since its discovery in 1704. Its stability upon exposure to light and anoxia remains difficult to apprehend. The present paper focuses on the relative influences of light, anoxia and type of substrate on the discoloration of Prussian blue dyed textiles. Spectrophotometry and X-ray absorption spectroscopy measurements of samples artificially aged by light in air or anoxia show that both the extent of the reduction process at the origin of Prussian blue discoloration and the aging of the textile substrate are linked and strongly differ with the environment. The complex inter-relationship existing between Prussian blue discoloration and textile degradation and the final impact it may have on the conservation of the entire system is discussed

    "Live” Prussian blue fading by time-resolved X-ray absorption spectroscopy

    Prussian blue (PB) is an artists' pigment that has been frequently used in many artworks but poses several problems of conservation because of its fading under light and anoxia treatment. PB fading is due to the reduction of iron(III) into iron(II) and depends a lot on the object investigated. Due to the complexity of the structure, the precise physico-chemical mechanisms behind the redox process remain obscure. In this paper, we present a procedure to investigate light- and anoxia-induced fading of PB-paper samples by means of time resolved X-ray absorption spectroscopy performed at the Fe K-edge. A system composed of a visible light source and a flux-controlled environmental cell allowed light, gas and humidity to be modified in situ. The synchrotron X-ray beam was evidenced to induce a reduction of PB and to play a major role in the kinetics. The analysis of the PB fading kinetics of a sample submitted to various gas and light environments showed that both synchrotron beam and anoxia were influencing PB reduction in a correlated way. In comparison, light was found to play a minor role. Finally, we have demonstrated that the type of paper substrate could influence significantly the kinetics of reduction.     Building Capacity and Infrastructure at Hospitals Implementing Minimally Invasive Tissue Sampling: Experience and Lessons Learned From Nepal, Rwanda, and Tanzania.

    BACKGROUND: Minimally invasive tissue sampling (MITS) is a useful tool to determine cause of death in low- and middle-income countries (LMICs). In 2019 the MITS Surveillance Alliance supported the implementation of small-scale postmortem studies using MITS in several LMICs. METHODS: In this article we describe the preparations, challenges, and lessons learned as part of implementing MITS across 4 study sites in 3 countries: Nepal, Rwanda, and Tanzania. We describe the process for building capacity to conduct MITS, which consisted of training in MITS sample collection, individual site assessment to determine readiness and gaps prior to implementation, site visits as sites began implementation of MITS, and feedback based on remote evaluation of histology slides via an online portal. RESULTS: The 4 study sites each conducted 100 MITS, for a total of 400. All 4 sites lacked sufficient infrastructure and facilities to conduct MITS, and upgrades were required. Common challenges faced by sites included that clinical autopsies were neither routinely conducted nor widely accepted. Limited clinical records made cause of death determination more difficult. Lessons learned included the importance of sensitization of the community and medical staff to MITS to enhance understanding and increase consent. CONCLUSIONS: The study sites accomplished MITS and utilized the available support systems to overcome the challenges. The quality of the procedures was satisfactory and was facilitated through the organized capacity-building programs

    Cytotoxic stilbenes and canthinone alkaloids from Brucea antidysenterica (Simaroubaceae)

    Phytochemical study of the root and bark of Brucea antidysenterica J. F. Mill. (Simaroubaceae) afforded three new compounds including a stilbene glycoside bruceanoside A (1), and two canthinone alkaloids bruceacanthinones A (3) and B (4), as well as ten known secondary metabolites, rhaponticin (2), 1,11-dimethoxycanthin-6-one (5), canthin-6-one (6), 1-methoxycanthin-6-one (7), 2-methoxycanthin-6-one (8), 2-hydroxy-1,11-dimethoxycanthin-6-one (9), β-carboline-1-propionic acid (10), cleomiscosin C (11), cleomiscosin A (12) and hydnocarpin (13). The structures of all the compounds were determined using spectrometric and spectroscopic methods including 1D and 2D NMR, and HRSEIMS. The identity of the known compounds was further confirmed by comparison of their data with those reported in the literature. The root and bark methanolic extracts, the dichloromethane and ethyl acetate soluble fractions, as well as the isolated compounds (3–13), were assessed for their cytotoxicity against the cancer cell lines A-549, MCF-7 and PC-3. The results suggested that compounds in the extracts might possess a synergic action in their cytotoxicity

    Gaining Greater Insight into HCV Emergence in HIV-Infected Men Who Have Sex with Men: The HEPAIG Study

    OBJECTIVES: The HEPAIG study was conducted to better understand Hepatitis C virus (HCV) transmission among human immuno-deficiency (HIV)-infected men who have sex with men (MSM) and assess incidence of HCV infection among this population in France. METHODS AND RESULTS: Acute HCV infection defined by anti-HCV or HCV ribonucleic acid (RNA) positivity within one year of documented anti-HCV negativity was notified among HIV-infected MSM followed up in HIV/AIDS clinics from a nationwide sampling frame. HIV and HCV infection characteristics, HCV potential exposures and sexual behaviour were collected by the physicians and via self-administered questionnaires. Phylogenetic analysis of the HCV-NS5B region was conducted. HCV incidence was 48/10 000 [95% Confidence Interval (CI):43-54] and 36/10 000 [95% CI: 30-42] in 2006 and 2007, respectively. Among the 80 men enrolled (median age: 40 years), 55% were HIV-diagnosed before 2000, 56% had at least one sexually transmitted infection in the year before HCV diagnosis; 55% were HCV-infected with genotype 4 (15 men in one 4d-cluster), 32.5% with genotype 1 (three 1a-clusters); five men were HCV re-infected; in the six-month preceding HCV diagnosis, 92% reported having casual sexual partners sought online (75.5%) and at sex venues (79%), unprotected anal sex (90%) and fisting (65%); using recreational drugs (62%) and bleeding during sex (55%). CONCLUSIONS: This study emphasizes the role of multiple unprotected sexual practices and recreational drugs use during sex in the HCV emergence in HIV-infected MSM. It becomes essential to adapt prevention strategies and inform HIV-infected MSM with recent acute HCV infection on risk of re-infection and on risk-reduction strategies
