    O presente Trabalho de Conclusão de Curso trata do lúdico e da ludopedagogia enquanto ações que promovem a saúde mental de jovens e adultos com transtornos mentais usuários dos serviços de saúde mental. Nesses espaços, que também são de atuação do pedagogo, existem diversos jovens e adultos em sofrimento, com transtornos psicossociais graves, indivíduos marcados pela exclusão e pressão da sociedade. Sendo assim, esta pesquisa buscou responder o seguinte questionamento: “qual o papel que o lúdico possui na promoção da saúde mental de jovens e adultos?’’. Partindo dele, se objetivou identificar o papel que o lúdico possui na promoção da saúde mental desse público, a partir da narrativa de trabalhadores dos serviços de saúde mental ou trabalhadores que já tiveram experiência nesses serviços e educadores. É uma pesquisa qualitativa e descritiva, assumindo um método cartográfico. As narrativas, analisadas partindo do meu olhar como futura pedagoga e da entrevista com os profissionais brasileiros e educadores uruguaios que estudam e apostam no lúdico e na ludopedagogia, demonstram que o lúdico promove saúde mental dos jovens e adultos usuários de saúde mental, ao evidenciar suas potências e diferentes formas de expressão, através de um tratamento humanizado e um diferente olhar sobre produtividade. O brincar, enquanto uma prática pedagógica, pode ser utilizado para promover aprendizagens, momentos de socialização e interação. O lúdico pode promover a saúde mental dos usuários dos serviços que compõem o Sistema Único de Saúde (SUS), tanto nos serviços de saúde mental, como o Centro de Atenção Psicossocial (CAPS) ou os atendimentos complementares, como o Acompanhamento Terapêutico (AT)

    Cereblon versus VHL:Hijacking E3 ligases against each other using PROTACs

    <div> <div> <div> <p>The von Hippel-Lindau (VHL) and cereblon (CRBN) proteins are substrate recognition subunits of two ubiquitously expressed and biologically important Cullin RING E3 ubiquitin ligase complexes. VHL and CRBN are also the two most popular E3 ligases being recruited by bifunctional Proteolysis-targeting chimeras (PROTACs) to induce ubiquitination and subsequent proteasomal degradation of a target protein. Using homo-PROTACs, VHL and CRBN have been independently dimerized to induce their own degradation. Here we report the design, synthesis and cellular activity of VHL-CRBN hetero-dimerizing PROTACs featuring diverse conjugation patterns. We found that the most active compound 14a induced potent, rapid and profound preferential degradation of CRBN over VHL in cancer cell lines. At lower concentrations, weaker degradation of VHL was instead observed. This work demonstrates proof of concept of designing PROTACs to hijack different E3 ligases against each other, and highlights a powerful and generalizable proximity-induced strategy to achieve E3 ligase knockdown. </p> </div> </div> </div

    Mouse Panx1 Is Dispensable for Hearing Acquisition and Auditory Function

    Panx1 forms plasma membrane channels in brain and several other organs, including the inner ear. Biophysical properties, activation mechanisms and modulators of Panx1 channels have been characterized in detail, however the impact of Panx1 on auditory function is unclear due to conflicts in published results. To address this issue, hearing performance and cochlear function of the Panx1−/− mouse strain, the first with a reported global ablation of Panx1, were scrutinized. Male and female homozygous (Panx1−/−), hemizygous (Panx1+/−) and their wild type (WT) siblings (Panx1+/+) were used for this study. Successful ablation of Panx1 was confirmed by RT-PCR and Western immunoblotting in the cochlea and brain of Panx1−/− mice. Furthermore, a previously validated Panx1-selective antibody revealed strong immunoreactivity in WT but not in Panx1−/− cochleae. Hearing sensitivity, outer hair cell-based “cochlear amplifier” and cochlear nerve function, analyzed by auditory brainstem response (ABR) and distortion product otoacoustic emission (DPOAE) recordings, were normal in Panx1+/− and Panx1−/− mice. In addition, we determined that global deletion of Panx1 impacts neither on connexin expression, nor on gap-junction coupling in the developing organ of Corti. Finally, spontaneous intercellular Ca2+ signal (ICS) activity in organotypic cochlear cultures, which is key to postnatal development of the organ of Corti and essential for hearing acquisition, was not affected by Panx1 ablation. Therefore, our results provide strong evidence that, in mice, Panx1 is dispensable for hearing acquisition and auditory function

    Helicobacter pylori and Epstein-Barr virus infection in gastric diseases: Correlation with IL-10 and IL1RN polymorphism.

    Introduction: Helicobacter pylori and Epstein-Barr virus (EBV) infection have recently 23 been shown to be associated with gastric diseases. Polymorphisms in genes encoding 24 cytokines such as interleukin 10 (IL-10) and interleukin 1 Receptor (IL-1RN) influence 25 cytokine secretion levels and appear to contribute to the risk of developing gastroduodenal 26 diseases. 27 To our knowledge, this is the first preliminary study to address the association of 28 coinfection with H. pylori and EBV and their correlation with genetic predisposition in the 29 development of gastric diseases. 30 Methods: Gastric biopsy samples of 96 patients with different gastric diseases were used. 31 Results: Our results showed that the rate of co-infection was higher in patients with 32 gastric cancer than in patients with normal gastric mucosa, active chronic gastritis and 33 MALT lymphoma. As regards the characterization of H. pilory strains, the 34 polymorphism s1m1i1 of vacA gene was more frequent in patients with MALT 35 Lymphoma in comparison to others, while the polymorphism s2m2i2 was most 36 frequent in patients with normal gastric mucosa. In addition, patients who tested 37 positivefor the cagA gene were more frequently those affected with gastric cancer than 38 those with inactive chronic gastritis. Similarly, the patients with oipA gene ON were more 39 frequently those with gastric cancer than those with inactive chronic gastritis. 40 Conclusion: According to our analysis, there was no correlation between coinfection 41 and polymorphisms in genes encoding IL-10 and IL-1RN. We conclude that various 42 factors can be involved in the development of gastric diseases
