    Integral transform solutions of dynamic response of a clamped–clamped pipe conveying fluid

    Analysis of dynamic response of pipe conveying fluid is an important aspect in nuclear power plant design. In the present paper, dynamic response of a clamped–clamped pipe conveying fluid was solved by the generalized integral transform technique (GITT). The governing partial differential equation was transformed into a set of second-order ordinary differential equations which is then numerically solved by making use of the subroutine DIVPAG from IMSL Library. A thorough convergence analysis was performed to yield sets of reference results of the transverse deflection at different time and spanwise position. We found good agreement between the computed natural frequencies at mode 1–3 and those obtained by previous theoretical study. Besides, modal separation analysis was carried out and the influence of mass ratio on deflection and natural frequencies was qualitatively and quantitatively assessed.Indisponível

    TIMCC: On Data Freshness in Privacy-Preserving Incentive Mechanism Design for Continuous Crowdsensing Using Reverse Auction

    © 2013 IEEE. As an emerging paradigm that leverages the wisdom and efforts of the crowd, mobile crowdsensing has shown its great potential to collect distributed data. The crowd may incur such costs and risks as energy consumption, memory consumption, and privacy leakage when performing various tasks, so they may not be willing to participate in crowdsensing tasks unless they are well-paid. Hence, a proper privacy-preserving incentive mechanism is of great significance to motivate users to join, which has attracted a lot of research efforts. Most of the existing works regard tasks as one-shot tasks, which may not work very well for the type of tasks that requires continuous monitoring, e.g., WIFI signal sensing, where the WiFi signal may vary over time, and users are required to contribute continuous efforts. The incentive mechanism for continuous crowdsensing has yet to be investigated, where the corresponding tasks need continuous efforts of users, and the freshness of the sensed data is very important. In this paper, we design TIMCC, a privacy-preserving incentive mechanism for continuous crowdsensing. In contrast to most existing studies that treat tasks as one-shot tasks, we consider the tasks that require users to contribute continuous efforts, where the freshness of data is a key factor impacting the value of data, which further determines the rewards. We introduce a metric named age of data that is defined as the amount of time elapsed since the generation of the data to capture the freshness of data. We adopt the reverse auction framework to model the connection between the platform and the users. We prove that the proposed mechanism satisfies individual rationality, computational efficiency, and truthfulness. Simulation results further validate our theoretical analysis and the effectiveness of the proposed mechanism

    Study of the correlation between vitamin D level and liver function in children with infectious mononucleosis

    Objective To investigate the correlation between the serum level of 25-hydroxyvitamin D (25(OH)D) and liver function in children with infectious mononucleosis (IM). Methods Ninety children with acute IM were enrolled into the IM group, and 40 healthy children who underwent physical examination during the same period were allocated into the control group. Serum level of 25(OH)D was determined by electrochemiluminescence and the viral load of Epstein-Barr virus DNA (EBV-DNA) in plasma was determined by quantitative fluorescent RT-PCR. Clinical data and serum 25(OH) D levels were compared between two groups. The correlation between 25(OH)D level and atypical lymphocytes, liver function parameters and plasma EBV-DNA in children with IM was analyzed. Results Serum 25(OH)D level in the IM group was significantly lower, whereas the 25(OH)D inadequacy rate was significantly higher than those in the control group (both P < 0.05). Serum 25(OH)D level was negatively correlated with atypical lymphocytes in the IM group (P < 0.05). The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), glutamyl transpeptidase (GGT), and adenosine deaminase (ADA) in IM children with serum 25(OH)D inadequacy were higher than those in their counterparts with adequate serum 25(OH)D (all P < 0.05). No significant differences were found in total bilirubin (TBIL), direct bilirubin (DBIL) and plasma EBV-DNA load between the adequate and inadequate subgroups (all P > 0.05). In the IM group, serum 25(OH)D level was negatively correlated with ALT, GGT, and ADA (all P < 0.05), whereas positively correlated with CHE (P < 0.05) and had no correlation with AST, TBIL or DBIL (all P > 0.05). Conclusions Serum vitamin D insufficiency occurs in children with IM. Vitamin D may be involved in the incidence and development of the course of IM, which is probably associated with liver function impairment

    Dichotomy in the Impact of Elevated Maternal Glucose Levels on Neonatal Epigenome

    Context Antenatal hyperglycemia is associated with increased risk of future adverse health outcomes in both mother and child. Variations in offspring's epigenome can reflect the impact and response to in utero glycemic exposure, and may have different consequences for the child. Objective We examined possible differences in associations of basal glucose status and glucose handling during pregnancy with both clinical covariates and offspring cord tissue DNA methylation. Research Design and Methods This study included 830 mother-offspring dyads from the Growing Up in Singapore Towards Healthy Outcomes cohort. The fetal epigenome of umbilical cord tissue was profiled using Illumina HumanMethylation450 arrays. Associations of maternal mid-pregnancy fasting (fasting plasma glucose [FPG]) and 2-hour plasma glucose (2hPG) after a 75-g oral glucose challenge with both maternal clinical phenotypes and offspring epigenome at delivery were investigated separately. Results Maternal age, prepregnancy body mass index, and blood pressure measures were associated with both FPG and 2hPG, whereas Chinese ethnicity (P = 1.9 x 10(-4)), maternal height (P = 1.1 x 10(-4)), pregnancy weight gain (P = 2.2 x 10(-3)), prepregnancy alcohol consumption (P = 4.6 x 10(-4)), and tobacco exposure (P = 1.9 x 10(-3)) showed significantly opposite associations between the 2 glucose measures. Most importantly, we observed a dichotomy in the effects of these glycemic indices on the offspring epigenome. Offspring born to mothers with elevated 2hPG showed global hypomethylation. CpGs most associated with the 2 measures also reflected differences in gene ontologies and had different associations with offspring birthweight. Conclusions Our findings suggest that 2 traditionally used glycemic indices for diagnosing gestational diabetes may reflect distinctive pathophysiologies in pregnancy, and have differential impacts on the offspring's DNA methylome.Peer reviewe

    Increased Drp1 promotes autophagy and ESCC progression by mtDNA stress mediated cGAS-STING pathway.

    Background: Mitochondrial dynamics homeostasis is important for cell metabolism, growth, proliferation, and immune responses. The critical GTPase for mitochondrial fission, Drp1 is frequently upregulated in many cancers and is closely implicated in tumorigenesis. However, the mechanism underling Drp1 to influence tumor progression is largely unknown, especially in esophageal squamous cell carcinoma (ESCC). Methods: Immunohistochemistry was used to examine Drp1 and LC3B expression in tissues of ESCC patients. Autophagic vesicles were investigated by transmission electron microscopy. Fluorescent LC3B puncta and mitochondrial nucleoid were observed by fluorescent and confocal microscopy. Mitochondrial function was evaluated by mitochondrial membrane potential, ROS and ATP levels. Xenograft tumor model was performed in BALB/c nude mice to analyze the role of Drp1 on ESCC progression. Results: We found that Drp1 high expression is correlated with poor overall survival of ESCC patients. Drp1 overexpression promotes cell proliferation and xenograft ESCC tumor growth by triggering autophagy. Furthermore, we demonstrated that Drp1 overexpression disturbs mitochondrial function and subsequent induces mitochondrial DNA (mtDNA) released into the cytosol thereby inducing cytosolic mtDNA stress. Mechanistically, cytosolic mtDNA activates the cGAS-STING pathway and facilitates autophagy, which promotes ESCC cancer growth. Moreover, mtDNA digestion with DNase I and autophagy inhibition with chloroquine attenuates the cGAS-STING pathway activation and ESCC cancer growth. Conclusions: Our finding reveals that Drp1 overexpression induces mitochondrial dysfunction and cytosolic mtDNA stress, which subsequently activates the cGAS-STING pathway, triggers autophagy and promotes ESCC progression

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data
