Coincidência de florescimento entre linhagens de couve-flor na produtividade e qualidade de sementes híbridas
The missing of flowering synchronization between the self-incompatible lines in a crop field of cauliflower hybrid seeds besides making the seed production smaller can compromise the genetic purity of them. The coincidence of the flowering time between two cauliflower lines was examined to study its effect on the productivity and quality of hybrid seeds. The treatments consisted of six different sowing dates, every fifteen days, using a self-incompatible tropical line pollinated by a winter line which does not present self-incompatibility. The following characteristics were evaluated: leaf average area and number of flowers per plant, number of siliques per plant, number and weight of seeds per plant, weight of thousand seeds and average number of seeds per silique. The germination standard test and genetic seed purity were determined for each treatment. The coincident flowering season between cauliflower lines affects directly the productivity and the genetic quality of the produced hybrid seeds. The closer the flowering time coincidence between the lines, the greater the number of seeds per silique and the smaller the percentage of non-hybrid seedlings. However, the coincidence of the flowering season between lines was found to influence physiological seed quality.A falta de sincronismo de florescimento entre as linhagens auto incompatíveis em um campo de produção de sementes híbridas de couve flor pode além de reduzir a produção de sementes comprometer a pureza genética das mesmas. Com o objetivo de estudar o efeito da coincidência de florescimento entre linhagens de couve-flor na produtividade e qualidade de sementes híbridas, foi realizado o presente experimento. Os tratamentos consistiram em seis diferentes épocas de semeadura, espaçadas a cada quinze dias, de uma linhagem de verão auto-incompatível que foi polinizada por uma linhagem de inverno que não apresenta auto-incompatibilidade. Observou-se a coincidência do florescimento das diferentes épocas de semeadura com a linhagem polinizadora. Foram avaliadas as seguintes características: área foliar média, número de flores por planta, número de síliqüas por planta, número de sementes por planta (peso e número), peso médio de 1000 sementes e foi determinado o número de sementes por síliqüa. Foi realizado ainda, o teste padrão de germinação e determinada a pureza genética das sementes para cada tratamento. A coincidência da época de florescimento entre as linhagens de couve-flor afetou diretamente a produtividade e a qualidade genética das sementes híbridas produzidas, sendo que, quanto maior foi o nível de coincidência, maior foi o número de sementes formadas por síliqüa e menor a percentagem de sementes contaminantes. Entretanto, não teve influência na qualidade fisiológica das mesmas
Effects of culture filtrates of endophytic fungi obtained from Piper aduncum L. on the growth of mycobacterium tuberculosis
Substances that inhibit the growth of Mycobacterium tuberculosis could potentially be used as antibiotics. These substances could also be added to test culture media to improve the speed of tuberculosis diagnosis. The aim of this work was to investigate the influence of culture filtrates of endophytic fungi isolated from P. aduncum L. on the growth of M. tuberculosis. To achieve this objective, the following methodology was used: a) endophytic fungi were isolated from the leaves and stems of P. aduncum L.; b) the isolated fungi were submitted to submerged bioprocessing; c) culture filtrates from the bioprocess were assayed to evaluate their effect on the growth of M. tuberculosis. We isolated 315 fungal types, which represented 85 morphologies, from different parts of P. aduncum L. The bioassays were performed on 82 culture filtrates and 6 plant extracts and resulted in the detection of 1 culture filtrate that stimulated the growth of M. tuberculosis and 15 that inhibited microbial growth. None of the phytochemical extracts had an effect on the growth of M. tuberculosis. In conclusion, we observed that the endophytic fungi isolated from P. aduncum L. (Piperaceae) produced extracellular metabolites (present in the culture filtrate) that affect the growth of M. tuberculosis. Molecular genetic diversity and mating system in sacha inchi progenies
Sacha inchi (Plukenetia volubilis L.) is a species with nutraceutical benefits traditionally consumed by Amazonic indigenous and urban communities. Studies on the genetic diversity and mating system are required to preserve and make the best use of the genetic resources for this species. This study aimed to estimate the genetic diversity and mating system parameters of sacha inchi progenies using Amplified Fragment Length Polymorphism (AFLP) markers. A total of 360 progenies from 30 accessions of sacha inchi were analyzed using three AFLP primers combinations. The percentage variation between and within families and the crossing rate, among other parameters, were estimated. The AFLP markers were efficient in genetically differentiating progenies, revealing 251 (98.82 %) polymorphic fragments. The analysis of molecular variance showed that the strongest variation occurs between progenies (57.16 %). However, the genetic differentiation within progenies was considerable (42.84 %), and could be exploited in breeding programs. The estimated population outcrossing rate was high (0.957), indicating it is a predominantly allogamous species. On the other hand, 33.7 % of the crosses occurred between related individuals. The estimate for biparental crosses revealed that the progenies consisted mainly of half-siblings (66.9 %) and full-siblings (28.8 %). For purposes of breeding and ex situ genetic conservation, sacha inchi seeds should be collected from a large number of parent plants
Óleo essencial de hortelã-pimenta como anestésico e sua toxicidade para juvenis de jundiá
The objective of this work was to evaluate peppermint (Mentha piperita) essential oil toxicity to and potential as an anesthetic for juvenile silver catfish (Rhamdia quelen). To determine the lethal concentration at 4 hours (LC50-4h), 210 fish (3.08±0.8 g and 7.59±0.67 cm) were exposed to 0, 20, 50, 80, 110, and 140 mg L-1 essential oil. To evaluate the anesthetic potential, nine fish were individually exposed to each oil concentration (50, 80, 110, and 140 mg L-1) used. Water quality parameters were controlled. The mortality rate and the severity and extent of the gill injuries of silver catfish juveniles at 4 hours of exposure increased with increasing peppermint essential oil concentrations, with LC50-4h estimated to be 75.06 mg L-1. The main gill injuries were: congestion of the venous sinus of the primary lamella and at the base of the secondary lamella; interlamellar hyperplasia with fusion of lamellae; epithelial detachment; dilation of the venous sinus; edema adjacent to the lamella; and aneurysm. However, this oil is an effective anesthetic for silver catfish juveniles at the concentration of 80 mg L-1, with a short time of anesthesia (<4 min) and recovery (<10 min), with no mortality.O objetivo deste trabalho foi avaliar a toxicidade e o potencial como anestésico do óleo essencial de hortelã-pimenta (Mentha piperita) para juvenis de jundiá (Rhamdia quelen). Para determinar a concentração letal em 4 horas (LC50-4h), 210 peixes (3,08±0,8 g e 7,59±0,67 cm) foram expostos a 0, 20, 50, 80, 110 e 140 mg L-1 do óleo essencial. Para avaliar o potencial anestésico, nove peixes foram individualmente expostos à cada concentração (50, 80, 110 e 140 mg L-1) de óleo utilizada. Os parâmetros de qualidade da água foram controlados. A taxa de mortalidade e a severidade e a abrangência das lesões branquiais de juvenis de jundiá em 4 horas de exposição cresceu com o aumento da concentração do óleo essencial de hortelã-pimenta, com LC50-4h estimada em 75,06 mg L-1. As principais lesões branquiais foram: congestão do seio venoso da lamela primária e na base da lamela secundária; hiperplasia interlamelar e com fusão das lamelas; descolamento do epitélio; dilatação do seio venoso; edema justalamelar; e aneurisma. Entretanto, esse óleo é eficiente anestésico para juvenis de jundiá na concentração de 80 mg L-1, com reduzido tempo de anestesia (<4 min) e de recuperação (<10 min), sem mortalidade
Induced aerocystitis and hemato-immunological parameters in Nile tilapia fed supplemented diet with essential oil of Lippia alba
O presente estudo avaliou a suplementação dietária com óleo essencial de Lippia alba sobre os parâmetros hemato-imunológicos em tilápias do Nilo (Oreochromis niloticus) submetidas à inflamação aguda induzida por carragenina na bexiga natatória. Pelo período de 45 dias, 96 peixes foram divididos em quarto tratamentos em triplicata: a) peixes suplementados com óleo esencial de L. alba (4 mL kg-1 de ração) injetados com carragenina; b) peixes suplementados com álcool de cereais injetados com carragenina; peixes não suplementados com óleo essencial injetados com carragenina; c) peixes não suplementados não injetados. Os níveis de cortisol, eritrograma, leucograma e o infiltrado inflamatório foram analisados seis horas após o estímulo inflamatório. Peixes injetados com carragenina apresentaram reação inflamatória aguda na bexiga natatória caracterizada por maior infiltrado de neutrófilos e monócitos. O número de neutrófilos circulantes foi significativamente maior nos peixes suplementados com L. alba quando comparado aos outros tratamentos. Não houve diferença nos níveis de cortisol. Para a dose, o tempo e a forma de administração testada, a suplementação com óleo essencil de L. alba não apresentou atividade anti-inflamatória. Por outro lado, foi constatada influência da suplementação dietária no número de neutrófilos após a aerocistite enfatizando a sua característica imunomoduladora.The present study evaluated the dietary supplementation with essential oil of Lippia alba on the hemato-immunological parameters of Nile tilapia (Oreochromis niloticus) submitted to acute inflammation induced by carrageenin injection in the swim bladder. For a period of 45 days, 96 fish were divided in four treatments in triplicate, as follows: fish fed supplemented diet with essential oil of L. alba (4 mL kg-1 dry ration) injected with carrageenin; fish fed supplemented diet with cereal alcohol injected with carrageenin; fish fed unsupplemented diet with essential oil injected with carrageenin; fish fed unsupplemented diet and noninjected. Cortisol levels, erythrogram, leukogram and the inflammatory infiltrate were analyzed 6 h after inflammatory stimulus. Carrageenin-injected fish showed acute inflammatory reaction in the swim bladder characterized by higher infiltrate of neutrophils and monocytes. The circulating neutrophils number was significantly higher in fish fed L. alba when compared to other treatments. No difference in cortisol levels was found. For dose, time and administration form tested, supplementation with essential oil of L. alba did not present anti-inflammatory activity. On the other hand, influence of dietary supplementation was observed on the neutrophils number after induced aerocystitis highlighting its immunomodulatory characteristic
In vitro susceptibility of Plasmodium falciparum Welch field isolates to infusions prepared from Artemisia annua L. cultivated in the Brazilian Amazon
Artemisinin is the active antimalarial compound obtained from the leaves of Artemisia annua L. Artemisinin, and its semi-synthetic derivatives, are the main drugs used to treat multi-drug-resistant Plasmodium falciparum (one of the human malaria parasite species). The in vitro susceptibility of P. falciparum K1 and 3d7 strains and field isolates from the state of Amazonas, Brazil, to A. annua infusions (5 g dry leaves in 1 L of boiling water) and the drug standards chloroquine, quinine and artemisinin were evaluated. The A. annua used was cultivated in three Amazon ecosystems (várzea, terra preta de índio and terra firme) and in the city of Paulínia, state of São Paulo, Brazil. Artemisinin levels in the A. annua leaves used were 0.90-1.13% (m/m). The concentration of artemisinin in the infusions was 40-46 mg/L. Field P. falciparum isolates were resistant to chloroquine and sensitive to quinine and artemisinin. The average 50% inhibition concentration values for A. annua infusions against field isolates were 0.11-0.14 μL/mL (these infusions exhibited artemisinin concentrations of 4.7-5.6 ng/mL) and were active in vitro against P. falciparum due to their artemisinin concentration. No synergistic effect was observed for artemisinin in the infusions

In vivo antimalarial activity and mechanisms of action of 4-nerolidylcatechol derivatives
4-Nerolidylcatechol (1) is an abundant antiplasmodial metabolite that is isolated from Piper peltatum roots. O-Acylation or O-alkylation of compound 1 provides derivatives exhibiting improved stability and significant in vitro antiplasmodial activity. The aim of this work was to study the in vitro inhibition of hemozoin formation, inhibition of isoprenoid biosynthesis in Plasmodium falciparum cultures, and in vivo antimalarial activity of several 4-nerolidylcatechol derivatives. 1,2-O,O-Diacetyl-4-nerolidylcatechol (2) inhibited in vitro hemozoin formation by up to 50%. In metabolic labeling studies using [1-(n)-3H]geranylgeranyl pyrophosphate, diester 2 significantly inhibited the biosynthesis of isoprenoid metabolites ubiquinone 8, menaquinone 4, and dolichol 12 in cultures of P. falciparum 3D7. Similarly, 2-O-benzyl-4-nerolidylcatechol (3) significantly inhibited the biosynthesis of dolichol 12. P. falciparum in vitro protein synthesis was not affected by compounds 2 or 3. At oral doses of 50 mg per kg of body weight per day, compound 2 suppressed Plasmodium berghei NK65 in infected BALB/c mice by 44%. This in vivo result for derivative 2 represents marked improvement over that obtained previously for natural product 1. Compound 2 was not detected in mouse blood 1 h after oral ingestion or in mixtures with mouse blood/blood plasma in vitro. However, it was detected after in vitro contact with human blood or blood plasma. Derivatives of 4-nerolidylcatechol exhibit parasite-specific modes of action, such as inhibition of isoprenoid biosynthesis and inhibition of hemozoin formation, and they therefore merit further investigation for their antimalarial potential. In vitro inhibition of Plasmodium falciparum by substances isolated from Amazonian antimalarial plants
In the present study, a quassinoid, neosergeolide, isolated from the roots and stems of Picrolemma sprucei (Simaroubaceae), the indole alkaloids ellipticine and aspidocarpine, isolated from the bark of Aspidosperma vargasii and A. desmanthum (Apocynaceae), respectively, and 4-nerolidylcatechol, isolated from the roots of Pothomorphe peltata (Piperaceae), all presented significant in vitro inhibition (more active than quinine and chloroquine) of the multi-drug resistant K1 strain of Plasmodium falciparum. Neosergeolide presented activity in the nanomolar range. This is the first report on the antimalarial activity of these known, natural compounds. This is also the first report on the isolation of aspidocarpine from A. desmanthum. These compounds are good candidates for pre-clinical tests as novel lead structures with the aim of finding new antimalarial prototypes and lend support to the traditional use of the plants from which these compounds are derived
