    Get PDF

We herein report a rare case of a 25-year-old immunocompetent male patient with disseminated tuberculosis of central nervous system (CNS), first presenting as multiple cerebral lesions with no meningeal involvement. Subsequent diagnostic workup disclosed extensive peritoneal involvement. A broad differential diagnosis was considered, including neoplastic and infectious diseases. The diagnosis was confirmed with positive PCR result for Mycobacterium tuberculosis in the biopsied mesenteric tissue. The patient was started on tuberculostatic regimen with favorable outcome. No acquired or hereditary immunodeficiency was documented. Disseminated tuberculosis in immunocompetent individuals is extremely rare. Genetic susceptibility factors have been reported in individuals with extensive forms of the disease and a high index of suspicion is required, as observed in our case.info:eu-repo/semantics/publishedVersio

Robust Identification of Target Genes and Outliers in Triple-negative Breast Cancer Data

Correct classification of breast cancer sub-types is of high importance as it directly affects the therapeutic options. We focus on triple-negative breast cancer (TNBC) which has the worst prognosis among breast cancer types. Using cutting edge methods from the field of robust statistics, we analyze Breast Invasive Carcinoma (BRCA) transcriptomic data publicly available from The Cancer Genome Atlas (TCGA) data portal. Our analysis identifies statistical outliers that may correspond to misdiagnosed patients. Furthermore, it is illustrated that classical statistical methods may fail in the presence of these outliers, prompting the need for robust statistics. Using robust sparse logistic regression we obtain 36 relevant genes, of which ca. 60\% have been previously reported as biologically relevant to TNBC, reinforcing the validity of the method. The remaining 14 genes identified are new potential biomarkers for TNBC. Out of these, JAM3, SFT2D2 and PAPSS1 were previously associated to breast tumors or other types of cancer. The relevance of these genes is confirmed by the new DetectDeviatingCells (DDC) outlier detection technique. A comparison of gene networks on the selected genes showed significant differences between TNBC and non-TNBC data. The individual role of FOXA1 in TNBC and non-TNBC, and the strong FOXA1-AGR2 connection in TNBC stand out. Not only will our results contribute to the breast cancer/TNBC understanding and ultimately its management, they also show that robust regression and outlier detection constitute key strategies to cope with high-dimensional clinical data such as omics data

Discrete Formulation for the dynamics of rods deforming in space

We describe the main ingredients needed to create, from the smooth lagrangian density, a variational principle for discrete motions of a discrete rod, with corresponding conserved Noether currents. We describe all geometrical objects in terms of elements on the linear Atiyah bundle, using a reduced forward difference operator. We show how this introduces a discrete lagrangian density that models the discrete dynamics of a discrete rod. The presented tools are general enough to represent a discretization of any variational theory in principal bundles, and its simplicity allows to perform an iterative integration algorithm to compute the discrete rod evolution in time, starting from any predefined configurations of all discrete rod elements at initial times

Homocysteine treatment alters redox capacity of both endothelial and tumor cells

    Get PDF
Homocysteine is a non-proteinogenic amino acid playing key roles in two interconnected metabolic pathways, namely, the activated methyl cycle and the linear trans-sulfuration pathway that allows the conversion of methionine to cysteine. A dysregulation of intracellular homocysteine metabolism could yield an increased export of this amino acid, leading to hyperhomocysteinemia, which has been associated with an increased risk of cardiovascular diseases. In spite of decades of experimental effort, there is no definitive consensus on what could be the molecular mechanisms whereby hyperhomocysteinemia could contribute to cardiovascular disease. The redox active nature of homocysteine has favored the idea of an induction of oxidative stress as the underlying mechanism of homocysteine toxicity. In contrast, homocysteine can also behave as an anti-oxidant. The present work is aimed to further analyze the capacity of homocysteine to modulate the redox capacity of both endothelial and tumor cells. Stability of Affine G-varieties and Irreducibility in Reductive Groups

Let GG be a reductive affine algebraic group, and let XX be an affine algebraic GG-variety. We establish a (poly)stability criterion for points xXx\in X in terms of intrinsically defined closed subgroups HxH_{x} of GG, and relate it with the numerical criterion of Mumford, and with Richardson and Bate-Martin-R\"ohrle criteria, in the case X=GNX=G^{N}. Our criterion builds on a close analogue of a theorem of Mundet and Schmitt on polystability and allows the generalization to the algebraic group setting of results of Johnson-Millson and Sikora about complex representation varieties of finitely presented groups. By well established results, it also provides a restatement of the non-abelian Hodge theorem in terms of stability notions.Comment: 29 pages. To appear in Int. J. Math. Alternate cyclin D1 mRNA splicing modulates P27\u3csup\u3eKlP1\u3c/sup\u3e binding and cell migration

Cyclin D1 is an important cell cycle regulator but in cancer its overexpression also increases cellular migration mediated by p27KlP1 stabilization and RhoA inhibition. Recently, a common polymorphism at the exon 4-intron 4 boundary of the human cyclin D1 gene within a splice donor region was associated with an altered risk of developing cancer. Altered RNA splicing caused by this polymorphism gives rise to a variant cyclin D1 isoform termed cyclin D1b, which has the same N-terminus as the canonical cyclin D1a isoform but a distinct C-terminus. In this study we show that these different isoforms have unique properties with regard to the cellular migration function of cyclin D1. Whereas they displayed little difference in transcriptional co-repression assays on idealized reporter genes, microarray cDNA expression analysis revealed differential regulation of genes including those that influence cellular migration. Additionally, while cyclin D1a stabilized p27KIP1 and inhibited RhoA-induced ROCK kinase activity, promoting cellular migration, cyclin D1b failed to stabilize p27KIP1 or inhibit ROCK kinase activity and had no effect on migration. Our findings argue that alternate splicing is an important determinant of the function of cyclin D1 in cellular migration

The Safe-Port project: an approach to port surveillance and protection

    Get PDF
SAFE-PORT is a recently started project addressing the complex issue of determining the best configurations of resources for harbour and port surveillance and protection. More specifically, the main goal is to find, for any given scenario, an adequate set of configuration solutions — i.e., number and type of sensors and equipments, their locations and operating modes, the corresponding personnel and other support resources — that maximize protection over a specific area. The project includes research and development of sensors models, novel algorithms for optimization and decision support, and a computer-based decision support system (DSS) to assist decision makers in that task. It includes also the development of a simulation environment for modelling relevant aspects of the scenario (including sensors used for surveillance, platforms, threats and the environment), capable to incorporate data from field-trials, used to test and validate solutions proposed by the DSS. Test cases will consider the use of intelligent agents to model the behaviour of threats and of NATO forces in a realistic way, following experts’ definitions and parameters
