    Genetic Dissimilarity between Mates, but Not Male Heterozygosity, Influences Divorce in Schistosomes

    Background: Correlational studies strongly suggest that both genetic similarity and heterozygosity can influence female mate choice. However, the influence of each variable has usually been tested independently, although similarity and heterozygosity might be correlated. We experimentally determined the relative influence of genetic similarity and heterozygosity in divorce and re-mating in the monogamous endoparasite Schistosoma mansoni. Methodology/Principal Findings: We performed sequential infections of vertebrate hosts with controlled larval populations of parasites, where sex and individual genetic diversity and similarity were predetermined before infection. Divorce rate increased significantly when females were given the opportunity to increase genetic dissimilarity through re-mating with a new partner, independently of the intensity of male-male competition. We found however no evidence for females attempting to maximize the level of heterozygosity of their reproductive partner through divorce. Conclusions/Significance: Female preference for genetically dissimilar males should result in more heterozygous offspring.     12 pagesInternational audienceSeveral parasites with complex life-cycles induce phenotypic alterations in their intermediate hosts. According to the host manipulation hypothesis, such phenotypic alterations are supposed to increase the fitness of the parasite at the expense of that of its intermediate hosts through increasing the probability of transmission to next hosts. Although the phenomenon has received a large attention, the proximate factors modulating the occurrence and intensity of host manipulation remain poorly known. It has however, been suggested that the amount of energy reserves in the intermediate host might be a key parameter, although its precise influence on the intensity of manipulation remains unclear. Dietary depletion in the host may also lead to compromise with other parasite traits, such as probability of establishing or growth or virulence. Here, we address the question through performing experimental infections of the freshwater amphipod Gammarus pulex with two different populations of the acanthocephalan fish parasite Pomphorhynchus laevis, and manipulation of host nutritional condition. Following exposure, gammarids were given either a "standard" diet (consisting of elm leaves and chironomid larvae) or a "deprived" food treatment (deprived in proteins), and infection parameters were recorded. Once parasites reached the stage at which they become infective to their definitive host, refuge use (a behavioural trait presumably implied in trophic transmission) was assessed, and metabolic rate was measured. Infected gammarids exposed to the deprived food treatment showed a lower metabolic rate, indicative of a lower body condition, compared to those exposed to the standard food treatment. Parasite size was smaller, and, depending on the population of origin of the parasites, intensity of infection was lower or mortality was higher in deprived hosts. However, food treatment had no effect on either the timing or intensity of behavioural modifications. Overall, while our results suggest that acanthocephalan parasites develop better in hosts in good condition, no evidence was found for an influence of host nutritional condition on host manipulation by parasites

    Multidimensionality in host manipulation mimicked by serotonin injection.

    8 pagesInternational audienceManipulative parasites often alter the phenotype of their hosts along multiple dimensions. 'Multidimensionality' in host manipulation could consist in the simultaneous alteration of several physiological pathways independently of one another, or proceed from the disruption of some key physiological parameter, followed by a cascade of effects. We compared multidimensionality in 'host manipulation' between two closely related amphipods, Gammarus fossarum and Gammarus pulex, naturally and experimentally infected with Pomphorhynchus laevis (Acanthocephala), respectively. To that end, we calculated in each host-parasite association the effect size of the difference between infected and uninfected individuals for six different traits (activity, phototaxis, geotaxis, attraction to conspecifics, refuge use and metabolic rate). The effects sizes were highly correlated between host-parasite associations, providing evidence for a relatively constant 'infection syndrome'. Using the same methodology, we compared the extent of phenotypic alterations induced by an experimental injection of serotonin (5-HT) in uninfected G. pulex to that induced by experimental or natural infection with P. laevis. We observed a significant correlation between effect sizes across the six traits, indicating that injection with 5-HT can faithfully mimic the 'infection syndrome'. This is, to our knowledge, the first experimental evidence that multidimensionality in host manipulation can proceed, at least partly, from the disruption of some major physiological mechanism

    Offspring Microbiomes Differ Across Breeding Sites in a Panmictic Species

    High dispersal rates are known to homogenize host’s population genetic structure in panmictic species and to disrupt host local adaptation to the environment. Long-distance dispersal might also spread micro-organisms across large geographical areas. However, so far, to which extent selection mechanisms that shape host’s population genetics are mirrored in the population structure of the enteric microbiome remains unclear. High dispersal rates and horizontal parental transfer may homogenize bacterial communities between breeding sites (homogeneous hypothesis). Alternatively, strong selection from the local environment may differentiate bacterial communities between breeding sites (heterogeneous hypothesis). Furthermore, selection from age-specific environmental or physiological factors may differentiate the microbiome between juveniles and adults. Here, we analyzed the cloacal bacterial 16S rRNA gene of fledgling greater flamingos, Phoenicopterus roseus, across nine western Mediterranean breeding sites and four breeding seasons (n = 731) and adult birds (n = 27) from a single site. We found that fledgling cloacal microbiome, as measured by alpha diversity, beta diversity, the relative abundance of assigned sequence variants (ASVs) belonging to a phylum and genus composition within phylum, varied significantly between sampling sites and across time within site despite high adult dispersal rates. The spatio-temporal effects were stronger on individual ASV absence/presence than on ASV abundance (i.e., than on core microbiome composition). Spatial effects had a stronger effect than temporal effects, particularly on ASV abundance. Our study supports the heterogeneous hypothesis whereby local environmental conditions select and differentiate bacterial communities, thus countering the homogenizing effects of high-dispersing host species. In addition, differences in core microbiome between adult vs. fledgling samples suggests that differences in age-specific environmental and/or physiological factors result in differential selection pressure of core enteric microbiome between age classes, even within the same environment. In particular, the genus Corynebacterium, associated with both seasonal fat uptake and migration in previous studies, was much more abundant in high-dispersing fledglings than in more resident adults. To conclude, selection mechanisms that shape the host’s genetic structure cannot be extended to the genetic structure of the enteric microbiome, which has important implications regarding our understanding of both host local adaptation mechanisms and enteric microbiome population genetics

    Personality May Confound Common Measures of Mate-Choice

    The measurement of female mating preferences is central to the study of the evolution of male ornaments. Although several different methods have been developed to assess sexual preference in some standardized way, the most commonly used procedure consists of recording female spatial association with different males presented simultaneously. Sexual preference is then inferred from time spent in front of each male. However, the extent to which the measurement of female mate-choice is related to exploration tendencies has not been addressed so far. In the present study we assessed the influence of variation in exploration tendencies, a trait closely associated to global personality, on the measurement of female mating preference in the zebra finch (Taeniopygia guttata) using the widely used four-chamber choice-apparatus. The number of movements performed within both exploration and mate-choice apparatus was consistent within and across the two contexts. In addition, personality explained variation in selectivity, preference strength and consistency. High-exploratory females showed lower selectivity, lower preference scores and displayed more consistent preference scores. Our results suggest that variation in personality may affect the measurement of female mating preference and may contribute to explain existing inconsistencies across studies
