Conductivity in Jurkat cell suspension after ultrashort electric pulsing
Ultrashort electric pulses applied to similar cell lines such as Jurkat and HL-60 cells can produce markedly different results , which have been documented extensively over the last few years. We now report changes in electrical conductivity of Jurkat cells subjected to traditional electroporation pulses (50 ms pulse length) and ultrashort pulses (10 ns pulse length) using time domain dielectric spectroscopy (TDS). A single 10 ns, 150 kV/cm pulse did not noticeably alter suspension conductivity while a 50 ms, 2.12 kV/cm pulse with the same energy caused an appreciable conductivity rise. These results support the hypothesis that electroporation pulses primarily interact with the cell membrane and cause conductivity rises due to ion transport from the cell to the external media, while pulses with nanosecond duration primarily interact with the membranes of intracellular organelles. However, multiple ultrashort pulses have a cumulative effect on the plasma membrane, with five pulses causing a gradual rise in conductivity up to ten minutes post-pulsing
Nanosecond electric pulses penetrate the nucleus and enhance speckle formation
Nanosecond electric pulses generate nanopores in the interior membranes of cells and modulate cellular functions. Here, we used confocal microscopy and flow cytometry to observe Smith antigen antibody (Y12) binding to nuclear speckles, known as small nuclear ribonucleoprotein particles (snRNPs) or intrachromatin granule clusters (IGCs), in Jurkat cells following one or five 10 ns, 150 kV/cm pulses. Using confocal microscopy and flow cytometry, we observed changes in nuclear speckle labeling that suggested a disruption of pre-messenger RNA splicing mechanisms. Pulse exposure increased the nuclear speckled substructures by 2.5-fold above basal levels while the propidium iodide (PI) uptake in pulsed cells was unchanged. The resulting nuclear speckle changes were also cell cycle dependent. These findings suggest that 10 ns pulses directly influenced nuclear processes, such as the changes in the nuclear RNA–protein complexes
Conductance statistics from a large array of sub-10 nm molecular junctions
Devices made of few molecules constitute the miniaturization limit that both
inorganic and organic-based electronics aspire to reach. However, integration
of millions of molecular junctions with less than 100 molecules each has been a
long technological challenge requiring well controlled nanometric electrodes.
Here we report molecular junctions fabricated on a large array of sub-10 nm
single crystal Au nanodots electrodes, a new approach that allows us to measure
the conductance of up to a million of junctions in a single conducting Atomic
Force Microscope (C-AFM) image. We observe two peaks of conductance for
alkylthiol molecules. Tunneling decay constant (beta) for alkanethiols, is in
the same range as previous studies. Energy position of molecular orbitals,
obtained by transient voltage spectroscopy, varies from peak to peak, in

Efficiency of Energy Conversion in Thermoelectric Nanojunctions
Using first-principles approaches, this study investigated the efficiency of
energy conversion in nanojunctions, described by the thermoelectric figure of

materials or creating poor thermal contacts in nanojunctions. The results of

The PDZ domain of the SpoIVB serine peptidase facilitates multiple functions
During spore formation in Bacillus subtilis, the SpoIVB protein is a critical component of the sigma (K) regulatory checkpoint. SpoIVB has been shown to be a serine peptidase that is synthesized in the spore chamber and which self-cleaves, releasing active forms. These forms can signal proteolytic processing of the transcription factor sigma (K) in the outer mother cell chamber of the sporulating cell. This forms the basis of the sigma (K) checkpoint and ensures accurate sigma (K)-controlled gene expression. SpoIVB has also been shown to activate a second distinct process, termed the second function, which is essential for the formation of heat-resistant spores. In addition to the serine peptidase domain, SpoIVB contains a PDZ domain. We have altered a number of conserved residues in the PDZ domain by site-directed mutagenesis and assayed the sporulation phenotype and signaling properties of mutant SpoIVB proteins. Our work has revealed that the SpoIVB PDZ domain could be used for up to four distinct processes, (i) targeting of itself for trans proteolysis, (11) binding to the protease inhibitor BofC, (iii) signaling of pro-sigma (K) processing, and (iv) signaling of the second function of SpoIVB
The ethos of physical activity delivery in mental health: a narrative study of service user experiences.
Our research into the physical activity experiences of people with severe mental illness has led us to take seriously the social and cultural environment in which physical activity is delivered. In this study, through narrative methodology, we examine service user accounts of physical activity to illuminate the characteristics of physical activity groups that are experienced as positive, helpful, or beneficial. We present several qualities and show how effective leadership and coaching is central to these qualities being present. We conclude that it is not so much what activity is delivered, but how it is delivered that is critical for sustained participation and positive outcomes
Simultano UV-spektrofotometrijsko određivanje ramiprila, acetilsalicilne kiseline i atorvastatin kalcija u kapsulama primjenom kemometrijskih metoda
In the present work, three different spectrophotometric methods for simultaneous estimation of ramipril, aspirin and atorvastatin calcium in raw materials and in formulations are described. Overlapped data was quantitatively resolved by using chemometric methods, viz. inverse least squares (ILS), principal component regression (PCR) and partial least squares (PLS). Calibrations were constructed using the absorption data matrix corresponding to the concentration data matrix. The linearity range was found to be 1-5, 10-50 and 2-10 µg mL–1 for ramipril, aspirin and atorvastatin calcium, respectively. The absorbance matrix was obtained by measuring the zero-order absorbance in the wavelength range between 210 and 320 nm. A training set design of the concentration data corresponding to the ramipril, aspirin and atorvastatin calcium mixtures was organized statistically to maximize the information content from the spectra and to minimize the error of multivariate calibrations. By applying the respective algorithms for PLS 1, PCR and ILS to the measured spectra of the calibration set, a suitable model was obtained. This model was selected on the basis of RMSECV and RMSEP values. The same was applied to the prediction set and capsule formulation. Mean recoveries of the commercial formulation set together with other figures of merit (calibration sensitivity, selectivity, limit of detection, limit of quantification and analytical sensitivity) were estimated. Validity of the proposed approaches was successfully assessed for analyses of drugs in the various prepared physical mixtures and formulations.U radu su opisane tri različite spektrofotometrijske metode za određivanje ramiprila, acetilsalicilne kiseline i atorvastatin kalcija u sirovinama i formulacijama. Preklapanje podataka kvantitativno je riješeno pomoću kemometrijskih metoda, tj. metodama inverznih najmanjih kvadrata (ILS), regresije glavnog sastojka (PCR) i djelomičnih najmanjih kvadrata (PLS). Kalibracije su postavljene pomoću matrice podataka za apsorpciju koja odgovara matrici pripadajućih koncentracija. Područje linearnosti za ramipril iznosilo je 1–5, za acetilsalicilnu kiselinu 10–50, a za atorvastatin kalcij 2–10 µg mL–1. Matrica s apsorbancijama dobivena je mjerenjem apsorbancije nultog reda na valnim duljinama između 210 i 320 nm. Set podataka za koncentracije ramiprila, acetilsalicilne kiseline i atorvastatin kalcija u smjesi statistički je tako organiziran da osigura maksimalnu količinu informacije u spektrima i minimalizira grešku multivarijantnih kalibracija. Primjenom odgovarajućih algoritama za PLS, PCR i ILS na snimljene spektre kalibracijskog seta dobiven je dobar model, koji je odabran na temelju RMSECV i RMSEP vrijednosti. Isti model je primijenjen i na set s predviđenim vrijednostima i na kapsule sa smjesom ove tri ljekovite tvari. Određena je srednja vrijednost povrata za komercijalnu formulaciju te ostale analitičke izvedbene značajke (kalibracijska osjetljivost, selektivnost, granica dokazivanja, granica određivanja i analitička osjetljivost). Potvrđena je primjenjljivost predloženih metoda u analizama lijekova u fizičkim smjesama i u gotovim ljekovitim oblicima
Transition Between Ground State and Metastable States in Classical 2D Atoms
Structural and static properties of a classical two-dimensional (2D) system
consisting of a finite number of charged particles which are laterally confined
by a parabolic potential are investigated by Monte Carlo (MC) simulations and
the Newton optimization technique. This system is the classical analog of the
well-known quantum dot problem. The energies and configurations of the ground
and all metastable states are obtained. In order to investigate the barriers
and the transitions between the ground and all metastable states we first
locate the saddle points between them, then by walking downhill from the saddle
point to the different minima, we find the path in configurational space from
the ground state to the metastable states, from which the geometric properties
of the energy landscape are obtained. The sensitivity of the ground-state
configuration on the functional form of the inter-particle interaction and on
the confinement potential is also investigated
Comparative Network Analysis of Preterm vs. Full-Term Infant-Mother Interactions
Several studies have reported that interactions of mothers with preterm infants show differential characteristics compared to that of mothers with full-term infants. Interaction of preterm dyads is often reported as less harmonious. However, observations and explanations concerning the underlying mechanisms are inconsistent. In this work 30 preterm and 42 full-term mother-infant dyads were observed at one year of age. Free play interactions were videotaped and coded using a micro-analytic coding system. The video records were coded at one second resolution and studied by a novel approach using network analysis tools. The advantage of our approach is that it reveals the patterns of behavioral transitions in the interactions. We found that the most frequent behavioral transitions are the same in the two groups. However, we have identified several high and lower frequency transitions which occur significantly more often in the preterm or full-term group. Our analysis also suggests that the variability of behavioral transitions is significantly higher in the preterm group. This higher variability is mostly resulted from the diversity of transitions involving non-harmonious behaviors. We have identified a maladaptive pattern in the maternal behavior in the preterm group, involving intrusiveness and disengagement. Application of the approach reported in this paper to longitudinal data could elucidate whether these maladaptive maternal behavioral changes place the infant at risk for later emotional, cognitive and behavioral disturbance
