    Operationally Responsive Space (ORS): An Architecture and Enterprise Model for Adaptive Integration, Test and Logistics

    The capability to rapidly deploy tactical satellites to meet a Joint Force Commander\u27s immediate battlespace requirements is a well-documented joint capability need. Key U.S. strategic documentation cites the need for the capability to maintain persistent surveillance or an unblinking eye over battlespace and to rapidly reconstitute critical space capabilities to preserve situational awareness. The warfighter requires a tactical space-based deployment capability which employs a request to launch and operational deployment window of 90 to 120 days. This master\u27s thesis executed two (2) major areas of work: apply, and reinforce the Operationally Responsive Space (ORS) mission tasks using the Joint Capabilities Integration Development System (JCIDS) process; then based on capability gap data generated from the process, analyze and define the capability gap of an ORS Adaptive Integration, Test and Logistics (IT&L) process for payload to bus deployment to meet the identified time scales. This document recommends engineering solutions and processes for the ORS IT&L to-be state for this warfighter capability. The ORS adaptive IT&L CONOPS developed as part of this work focuses on the Tactical Satellite Rapid Deployment System (TSRDS), which is an adaptive integration, test and logistics capability that enables rapid and effective payload to bus integration to meet a 90- to 120-day warfighter window

    Retention of function without normal disc morphogenesis occurs in cone but not rod photoreceptors

    It is commonly assumed that photoreceptor (PR) outer segment (OS) morphogenesis is reliant upon the presence of peripherin/rds, hereafter termed Rds. In this study, we demonstrate a differential requirement of Rds during rod and cone OS morphogenesis. In the absence of this PR-specific protein, rods do not form OSs and enter apoptosis, whereas cone PRs develop atypical OSs and are viable. Such OSs consist of dysmorphic membranous structures devoid of lamellae. These tubular OSs lack any stacked lamellae and have reduced phototransduction efficiency. The loss of Rds only appears to affect the shape of the OS, as the inner segment and connecting cilium remain intact. Furthermore, these structures fail to associate with the specialized extracellular matrix that surrounds cones, suggesting that Rds itself or normal OS formation is required for this interaction. This study provides novel insight into the distinct role of Rds in the OS development of rods and cones

    Analysis of Immune Checkpoint Drug Targets and Tumor Proteotypes in Non-Small Cell Lung Cancer

    New therapeutics targeting immune checkpoint proteins have significantly advanced treatment of non-small cell lung cancer (NSCLC), but protein level quantitation of drug targets presents a critical problem. We used multiplexed, targeted mass spectrometry (MS) to quantify immunotherapy target proteins PD-1, PD-L1, PD-L2, IDO1, LAG3, TIM3, ICOSLG, VISTA, GITR, and CD40 in formalin-fixed, paraffin-embedded (FFPE) NSCLC specimens. Immunohistochemistry (IHC) and MS measurements for PD-L1 were weakly correlated, but IHC did not distinguish protein abundance differences detected by MS. PD-L2 abundance exceeded PD-L1 in over half the specimens and the drug target proteins all displayed different abundance patterns. mRNA correlated with protein abundance only for PD-1, PD-L1, and IDO1 and tumor mutation burden did not predict abundance of any protein targets. Global proteome analyses identified distinct proteotypes associated with high PD-L1-expressing and high IDO1-expressing NSCLC. MS quantification of multiple drug targets and tissue proteotypes can improve clinical evaluation of immunotherapies for NSCLC

    Examining ozone susceptibility in the genus Musa (bananas)

    Tropospheric ozone (O3) is a global air pollutant that adversely affects plant growth. Whereas the impacts of O3 have previously been examined for some tropical commodity crops, no information is available for the pantropical crop, banana (Musa spp.). To address this, we exposed Australia’s major banana cultivar, Williams, to a range of [O3] in open top chambers. In addition, we examined 46 diverse Musa lines growing in a common garden for variation in three traits that are hypothesised to shape responses to O3: (1) leaf mass per area; (2) intrinsic water use efficiency; and (3) total antioxidant capacity. We show that O3 exposure had a significant effect on the biomass of cv. Williams, with significant reductions in both pseudostem and sucker biomass with increasing [O3]. This was accompanied by a significant increase in total antioxidant capacity and phenolic concentrations in older, but not younger, leaves, indicating the importance of cumulative O3 exposure. Using the observed trait diversity, we projected O3 tolerance among the 46 Musa lines growing in the common garden. Of these, cv. Williams ranked as one of the most O3-tolerant cultivars. This suggests that other genetic lines could be even more susceptible, with implications for banana production and food security throughout the tropics

    The Atacama Cosmology Telescope: Two-Season ACTPol Lensing Power Spectrum

    We report a measurement of the power spectrum of cosmic microwave background (CMB) lensing from two seasons of Atacama Cosmology Telescope Polarimeter (ACTPol) CMB data. The CMB lensing power spectrum is extracted from both temperature and polarization data using quadratic estimators. We obtain results that are consistent with the expectation from the best-fit Planck LCDM model over a range of multipoles L=80-2100, with an amplitude of lensing A_lens = 1.06 +/- 0.15 (stat.) +/- 0.06 (sys.) relative to Planck. Our measurement of the CMB lensing power spectrum gives sigma_8 Omega_m^0.25 = 0.643 +/- 0.054; including baryon acoustic oscillation scale data, we constrain the amplitude of density fluctuations to be sigma_8 = 0.831 +/- 0.053. We also update constraints on the neutrino mass sum. We verify our lensing measurement with a number of null tests and systematic checks, finding no evidence of significant systematic errors. This measurement relies on a small fraction of the ACTPol data already taken; more precise lensing results can therefore be expected from the full ACTPol dataset.Comment: 17 pages, 11 figures, to be submitted to Physical Review

    Mapping the Circumstellar Environment of T Tauri with Fluorescent H_2 Emission

    We have obtained three long-slit, far UV spectra of the pre-main sequence system T Tauri. These HST/STIS spectra show a strong and variable on-source spectrum composed of both fluoresced H_2 and stellar chromospheric lines. Extended H_2 emission is seen up to 10" from the T Tau system. The on-source and extended H_2 are both pumped by H I Lyman alpha. The on-source H_2 is pumped by the red wing of a broad, self-absorbed Ly-alpha line, while the progressions seen in the extended gas are pumped from near line center. This suggests that the extended H_2 is pumped locally, and not by the stellar Ly-alpha line. The H_2 to the north and west coincides with the evacuated cavity bounded by the optical reflection nebulosity; to the south the extended H_2 coincides with the HH 255 outflow from the embedded infrared companion T Tau S. The spatial profile of the extended gas shows a prominent dip coincident with the position of T Tau S. This may be absorption by a disk associated with T Tau S. There is no evidence for absorption by a disk surrounding T Tau N large enough to obscure T Tau S.Comment: 46 pages, including 12 figures and 5 tables.
