Cuestiones éticas de la edición genética mediante la tecnología CRISPR-Cas9

Immersos ja en la 4a revolució industrial, clarament l'era digital ha envaït les nostres vides. La gent tendeix a celebrar cada avanç tecnològic i científic sense pensar adequadament en els possibles efectes secundaris no desitjats o les conseqüències inesperades. Un dels assoliments més destacats dels últims anys ha estat la tècnica CRISPR/*Cas9. Sembla ser la vareta màgica per a resoldre el que sigui. En aquest article em proposo pensar críticament sobre les qüestions ètiques relacionades amb aquesta increïble tècnica. Centraré aquest treball en 3 aspectes ètics de l'edició de gens: 1. El vincle entre l'edició de gens i l'antic concepte d'eugenèsia; 2. Aspectes relacionats amb la idea de modificar la genètica humana per a conquistar altres planetes i 3. Les implicancias ètiques de pujar totes aquestes dades a la web. Finalment, suggereixo enfocar-se en la discussió ètica interdisciplinària, el diàleg entre professionals, institucions i govern que ha d'assegurar la protecció de l'autonomia dels pacients i la salvaguarda dels seus drets. Per a concloure, es planteja el suggeriment de considerar el paper de les Universitats en l'educació ètica dels futurs professionals amb un enfocament més pràctic que teòric, amb especial atenció a la integració de la cibernètica en la medicina.Immersed already in the 4th industrial revolution, clearly the digital age has invaded our lives. People tend to celebrate every technological and scientific advance without thinking properly about the possible undesired side effects or unexpected consequences. One of the most outstanding achievements of the last years has been the CRISPR/Cas9 technique. It seems to be the magic wand to solve whatever. In this article I propose to think critically about the ethical issues related to this incredible technique. I will focus this work on 3 ethical aspects of gene editing: 1. The link between gene editing and the old concept of eugenics; 2. Aspects related to the idea of modifying human genetics in order to conquer other planets and 3. The ethical implications of uploading all these data to the web. Finally, I suggest focusing on interdisciplinary ethical discussion, the dialogue between professionals, institutions and government which must ensure the protection of the autonomy of patients and the safeguarding of their rights. To conclude, the suggestion of considering the role of Universities in ethical education of the future professionals with a practical rather than a theoretical approach is considered, with special attention to the integration of cybernetics in medicine.  Inmersos ya en la 4ª revolución industrial, claramente la era digital ha invadido nuestras vidas. La gente tiende a celebrar cada avance tecnológico y científico sin pensar adecuadamente en los posibles efectos secundarios no deseados o las consecuencias inesperadas. Uno de los logros más destacados de los últimos años ha sido la técnica CRISPR/Cas9. Parece ser la varita mágica para resolver lo que sea. En este artículo me propongo pensar críticamente sobre las cuestiones éticas relacionadas con esta increíble técnica. Centraré este trabajo en 3 aspectos éticos de la edición de genes: 1. El vínculo entre la edición de genes y el antiguo concepto de eugenesia; 2. Aspectos relacionados con la idea de modificar la genética humana para conquistar otros planetas y 3. Las implicancias éticas de subir todos estos datos a la web. Finalmente, sugiero enfocarse en la discusión ética interdisciplinaria, el diálogo entre profesionales, instituciones y gobierno que debe asegurar la protección de la autonomía de los pacientes y la salvaguarda de sus derechos. Para concluir, se plantea la sugerencia de considerar el papel de las Universidades en la educación ética de los futuros profesionales con un enfoque más práctico que teórico, con especial atención a la integración de la cibernética en la medicina

Bioética consolidada: abordaje histórico a más de 40 años de surgimiento

En este trabajo realizamos un abordaje histórico sobre algunos aspectos relacionados con el surgimiento, evolución y consolidación disciplinaria de la bioética. Nos centramos en analizar el número de publicaciones que contuvieran el vocablo bioética desde 1971 hasta 2011, analizamos brevemente la temática preponderante en cada año en ese período y el registro de revistas indexadas que contuvieran el término Bioética, tanto en inglés como en español. Se constató que el número de publicaciones con el término bioética fue de 1 en 1971 creciendo hasta 2997 en 2011. Se registraron 31 revistas que cumplían con el requisito de búsqueda, desde el año 1976, de las cuales 23 continúan publicándose. Respecto de las temáticas preponderantes, más allá de los temas habituales como aborto, eutanasia, transplantes, etc., se destacan los aportes en torno a la educación de los profesionales en el tema, la creación y sostenimiento de los comités de bioética, la vinculación de la bioética con la política y los medios de comunicación, hasta el surgimiento en los últimos años de subespecializaciones tales como la neurobioética

Bioética consolidada: abordaje histórico a más de 40 años de surgimiento

In this paper we attempt a historical approach on some aspects related to the emergence, evolution and consolidation of Bioethics as a discipline. We focus on analyzing the number of publications containing the word Bioethics from 1971 to 2011. We analyze briefly the dominant theme in every year in that period and also the registering indexed journals containing the term Bioethics in English or Spanish. We found that the number of publications with the term bioethics was 1 in 1971 rising to 2997 in 2011. There were 31 journals that met the search requirement, since 1976, of which 23 are still running. Regarding the prevailing theme, beyond the usual topics such as abortion, euthanasia, transplants, etc., highlights the contributions around the education of professionals in the field, the creation and maintenance of bioethic committees, the linking between Bioethics and politics and the media, and the emergence of subspecialties such as Neurobioethics in recent years.En este trabajo realizamos un abordaje histórico sobre algunos aspectos relacionados con el surgimiento, evolución y consolidación disciplinaria de la bioética. Surgery after Neoadjuvant Chemotherapy: A Clip-Based Technique to Improve Surgical Outcomes, a Single-Center Experience

SIMPLE SUMMARY: Neoadjuvant chemotherapy (NACT) has an important role in the treatment of locally advanced breast cancer. After NACT, some lesions may be no longer visible at preoperative imaging, making breast and axillary conservative surgery more difficult. Among others, radiopaque clips are the most commonly used method to mark lymph nodes and tumor sites to tailor surgery in the post neoadjuvant setting. ABSTRACT: Background: This study aims to describe the surgical management of breast cancer patients after neoadjuvant chemotherapy, with attention to the impact on surgical outcomes of a clip-based marking technique. Methods: Patients who underwent NACT at the Breast Unit of the A. O Ordine Mauriziano of Turin from January 2018 and had a surgical intervention by January 2022 were included. Data on the feasibility of clip insertion, after-treatment visibility, and successful removal during surgery were collected prospectively. Surgical outcomes in terms of breast-conserving surgery and axillary dissection reduction were described. Results: In 51 patients who had surgery after NACT, 55 clips were placed (34 breast and 21 axillary clips). Ultrasound visibility of the clips was optimal (91%) as well as preoperative localization and retrieval within the surgical specimen. Moreover, the use of the clip positively affected surgical outcomes. In our study, clip insertion allowed to avoid mastectomy and axillary dissection in patients with a complete radiological response. Conclusions: In our findings, the use of breast and/or lymph node clips has proved to be a simple and effective method to improve surgical conservative management of breast cancer patients after NACT

8-Chloro-cAMP-Related Changes on Mice Uteri

Histopathological effects of cAMP analog (8-Chloro-cAMP), tamoxifen, and medroxyprogesterone, alone or combined, upon BALB/c mice uteri are reported. 8-Chloro-cAMP diminished uterine weight, but did not modify its histopathology or estral cycle significantly. Tamoxifen diminished uterine weight showing cystic hyperplasia and an estral cycle arrested at diestrus. Medroxyprogesterone increased uterine weight, caused a swelling of the endometrium and a pseudopregnancy estrus. When combined with 8-Chloro-cAMP, tamoxifen or medroxyprogesterone always had a predominant effect. We concluded that the effects of 8-Chloro-cAMP on mice uteri did not cause significant changes on its histopathology, but diminished its weight

Study of an Impact Mill-Based Mechanical Method for NdFeB Magnet Recycling

Nowadays, the circular economy is gaining more and more attention in sectors where the raw material supply is critical for both cost and geo-political reasons. Moreover, the environmental impact issue calls for recycling. From this perspective, the recovery of rare earth elements represents a strategic point. On the other hand, the high cost and the dangerous standard recovery methods that apply to NdFeB magnets limits options for traditional recycling. A new mechanical method is proposed, not requiring hydrogen, high temperature, or chemical processes, but instead using an impact mill designed to operate in vacuum. A traditional impact mill operating in a glove box filled with Ar atmosphere has also been used for comparison. The obtained NdFeB powders were analyzed in terms of magnetic properties and chemical composition, particularly in terms of the oxygen content

Development of data-driven PHM solutions for robot hemming in automotive production lines

Robotic roller hemming is currently one of the most used solution for joining metal sheets in automotive industry, especially for those production lines which need to favor flexibility with respect to raw productivity and is mostly employed to assemble car doors. Hemming is a fairly delicate process since it does not only suffice a technical requirement – to join two panels together – but also an aesthetical one, since the joint panels are an integral part of the vehicle design and as such an important selling point. An unpredicted rupture or an advanced degradation condition of the system would lead to a significant loss in the quality of the final product or to a sudden stoppage of the production line. The development of a PHM system for hemming devices would hence provide a significant advantage, especially if designed to work for both new and legacy equipment. In this paper, we provide the results of a preliminary analysis of a new PHM framework for robotic roller hemming studied to work without having access to PLC data; the employed data-driven methodology is detailed and applied to the case of increasing wear in the head finger roll. Results from different prognostics routines are hence presented and compared
