    Get PDF
Rooting performance in microcuttings of five indigenous and one exotic landscape tree species was evaluated in a 60 days period. Rate of rooting varied greatly among species as Ilex cymosa, Tabebuia heterophylla and Agalaia korthalsii reached 100% rooting at 24, 42 and 48 days after insertion into the rooting medium followed by Lepisanthes rubiginosa and Hopea odorata species (95 & 80% rooting at 60 days, respectively). The poorest rooting ability (30% rooting at 60 days) was shown by Aporusa globifera and was first noticed at 30 days. Results of Ilex cymosa species for adventitious root induction per microcutting was markedly different from other species and it produced the highest number of adventitious roots (14.07 roots/microcutting). A. korthalsii generated the fewer, but the heaviest roots compared to I. cymosa. Rooting ability of microcuttings was highly correlated with its number of leaves produced during rooting (r = 0.869-0.981) as well as, with regenerative shoot length of the mother plants (r = 0.690-0.954). Regenerative leaf growth of mother plants and the rooting of their microcuttings was species-specific suggesting that developing association between the two parameters is difficult

Soils consist of complex mixtures of mineral, water, air, organic matter and as an ecosystem of a large number of countless microorganisms such as bacteria, fungi and nematodes. Due to the limitation on understanding of fungi biodiversity in soil, many people do not know there consists of very useful microbe that can help in suppression of plant pathogens. Trichoderma (Hypocreales, Ascomycota) is one of the microorganisms that living in all types of soil. Trichoderma species has been used as a biological control agent of phytopathogens due to its abundance and having an antagonistic property. This study will provide checklists of Trichoderma species that have been isolated from cultivated crop soils in Malaysia. The objectives of this study are to isolate and identify Trichoderma species from soil based on morphological characterization and to calculate diversity of Trichoderma species based on Shannon-Wiener Index. Soil samples from each cultivated agricultural crops were collected in three spots in 17 locations around Malaysia. Isolation of Trichoderma was completed using soil dilution technique to obtain a pure single colony-forming unit (CFU). A total of 253 of Trichoderma isolates were successfully isolated from rhizosphere of different crops. All isolates were tentatively identified into six species such as T. harzianum (114 isolates), T. asperellum (65 isolates), T. hamatum (22 isolates), T. viride (26 isolates),T. koningii (20 isolates) and T. virens (6 isolates). The differences between species were characterized based on their colony features, pigmentation on potato dextrose agar (PDA) and the branching of conidiophores. The diversity index of Trichoderma species found in the Malaysia soil was 1.43 and T. harzianum was dominated. Due to diverse species of Trichoderma found in Malaysia, it will increase the opportunity in selection for biological control agent of plant diseases

    Get PDF
West Nile virus (WNV) is a zoonotic RNA virus maintained in enzootic cycles involving mosquito mainly Culex and Aedes spp. as the vector and birds as the reservoir host. WNV is endemic in Africa, Europe and Western Asia. Human infection results in asymptomatic illness such as fever, headache, tiredness, body aches, nausea, vomiting, skin rash, swollen lymph glands and neuroinvasive disease are seen in less than 1% of infected persons. The spectrum of symptoms in animals includes fever, weakness and paralysis of hind limbs, impaired vision, ataxia, head pressing, aimless wandering, seizures, inability to swallow, walking in circles and coma. Based on the previous study in Malaysia, the antibody and nucleic acid against WNV were detected in Orang Asli, captive birds and horses. In this paper, potential risk factors contributing to WNV occurrence are discussed. How the disease or infection is diagnosed and controlled was also discussed

Evaluation of Trichoderma asperellum B1902 in controlling Fusarium wilt of cavendish banana cultivar

    Get PDF
Trichoderma species is one of the microorganisms with antagonistic properties as biological control agents. In the banana industry, Fusarium wilt disease caused by Fusarium oxysporum f. sp. cubense (Foc) has been practically managed using chemical pesticides that led to environmental disruptions, ineffective conditions and disease resistance. In preliminary study, T. asperellum gave better result compared to other species in inhibiting the growth of Foc in in vitro condition. Therefore, the aim of this study was to examine the effects of T. asperellum as a biological control of Fusarium wilt disease of banana. A total of 326 fungal isolates were isolated from soil samples obtained around Malaysia and identified as Trichoderma species based on phenotype characteristics. The species identity for the best candidates from dual culture test was confirmed based on internal transcribed spacers (ITS) and translation elongation factor 1 alpha (TEF-1α) sequence identity. In dual culture test, findings showed that three isolates with a high percentage inhibition of radial growth (PIRG) were observed in plates of T. asperellum isolates B1902 (84.85%), T2007 (77.78%) and C1667 (75.76%), which successfully inhibited the growth of F. oxysporum f. sp. cubense isolate 9888. Based on in vivo test, the best candidate was T. asperellum B1902 with lower disease severity index (DSI) value of 0.2 compared to the inoculated control with DSI of 3.6. As a conclusion, T. asperellum B1902 can be used as an alternative treatment in managing Fusarium wilt disease. Hence, future study should be focused on applying T. asperellum as a biocontrol agent in the field and controlling other plant diseases in the agricultural plantation

Beta-Lactam Infusion in Severe Sepsis (BLISS): a prospective, two-centre, open-labelled randomised controlled trial of continuous versus intermittent beta-lactam infusion in critically ill patients with severe sepsis

    Get PDF
This study aims to determine if continuous infusion (CI) is associated with better clinical and pharmacokinetic/pharmacodynamic (PK/PD) outcomes compared to intermittent bolus (IB) dosing in critically ill patients with severe sepsis. This was a two-centre randomised controlled trial of CI versus IB dosing of beta-lactam antibiotics, which enrolled critically ill participants with severe sepsis who were not on renal replacement therapy (RRT). The primary outcome was clinical cure at 14 days after antibiotic cessation. Secondary outcomes were PK/PD target attainment, ICU-free days and ventilator-free days at day 28 post-randomisation, 14- and 30-day survival, and time to white cell count normalisation. A total of 140 participants were enrolled with 70 participants each allocated to CI and IB dosing. CI participants had higher clinical cure rates (56 versus 34 %, p = 0.011) and higher median ventilator-free days (22 versus 14 days, p MIC than the IB arm on day 1 (97 versus 70 %, p < 0.001) and day 3 (97 versus 68 %, p < 0.001) post-randomisation. There was no difference in 14-day or 30-day survival between the treatment arms. In critically ill patients with severe sepsis not receiving RRT, CI demonstrated higher clinical cure rates and had better PK/PD target attainment compared to IB dosing of beta-lactam antibiotics. Continuous beta-lactam infusion may be mostly advantageous for critically ill patients with high levels of illness severity and not receiving RRT
