    在福建东南沿海局部地域献血员中检出HTLV-I

    [目的 ]进一步阐明福建沿海 HTL V小流行区 HTL V- 的地理分布特点及毒株的基因类型。 [方法 ]对福建东南沿海莆田地区献血员以国产双抗原夹心法 EL ISA试剂筛选 ,对 EL ISA阳性血清用 Western blot(WB)进行确证。对抗体确证阳性者 ,采集外周血分离淋巴细胞用巢式 PCR扩增 HTL V- env区 gp46片段进行序列分析鉴定病毒亚型。 [结果 ]从 1998年 10月至 2 0 0 0年 4月止 ,共检测 45 6 4份标本 ,结果确证出 16例 HTL V- 抗体阳性者 ,感染率为 0 .35 %。HTL V- 抗体阳性者主要分布在某村献血人群中 ,并具有区域集中的特点。笏石秀屿一带某村献血员中感染率高达 4.7%。7例代表性毒株经病毒序列分析为 HTL V- C亚型 (COSMOPOL ITAN)。[结论 ]福建东南沿海某村献血员中发现 HTL V- 流行。提出在对某一地区人群进行 HTL V筛选时 ,既要注意整个地区“面”的筛查 ,又要注重某个村镇“点”的调查

    [中文文摘]针对固体中杂质发光光谱的特殊问题 ,以FORTRAN 77语言编写了多套曲线拟合程序 ,可分别适用于光滑型 ,散粒噪音叠加型及混合型光谱曲线的分解。并介绍了正确使用程序的技巧。对如何获得正确、可靠的拟合结果进行了讨论。[英文文摘]At first we stress that a successful fit result is determined by three factors, namely the high resolution spectrum data, effective computer program and skill for correctly undertaking curve fitting. The latter two factors are considered in this article. The algorithm of curve fitting are based on the damped least squares optimization methods. It is programmed in FORTRAN 77.The general principles of a satisfactory optimization routine are given in the context of degree of convergence and the computation time needed to achieve an acceptable fit. The computer grograms developed are capable of separating 24 overlapping bands at the most in a conr posite profile having 500 experimental data points.In the case of a composite profile strongly perturbed by the shot noises,a weighting function must be introduced into the computer program for its effectively running.Corresponding to the shape of a profile (a smooth curve or a curve revealing a high-level of noises)and the constraint conditions,one may use a suitable program to undertake courve fitting.The functions of six programs are listed in Table 1. Furthermore,we stress that the mere ability to undertake the required calculations,in terms of rapid data handing capability of the computer,is not the guarantee that meaningful fits will be obtained.It is shown that a preliminary data processing is needed.In order to eliminate“insensible parameters”during the fitting calculations we should make the conversion of the values of original data.The value of abscissa for every point should be converted into the value expressed in the unit of meV. The value of intensity for every experimental point must be converted into a value as the maximum intensity is set at 100 a.u.Curve fitting may be facilitated if the initial estimates of parameters were chosen appropriately.The peak energy position must be accurately fixed.The discrepancy of a fit may be employed to locate any regions of poor fit in con2 junction with visual plot. The regions of poor fit are att ributed to incorrect choice of the initial estimates of parameters or the incorrect fit model adopted. A successful fit result is determined by the fit model which include the number of peaks,peak energy position and degree of band asymmet ry. These factors are discussed in detail. At last we have exemplified the curve fitting of the 25K luminescence spect rum involving NN1-bound exciton in GaP:N sample.     【中文摘要】 用低压金属有机气相外延 (LP MOCVD)在InP衬底上生长出较高质量的GaAs材料 ,并对材料进行Raman、光致发光 (PL)谱的测试分析 ,结果表明在GaAs InP外延层中存在伸张应力致使Raman的LO模式的频率红移 ;PL谱峰较强 ,16K下测到GaAs的特征激子峰和杂质相关的激子峰 ,表明了GaAs外延层的晶体质量较好 .以此生长方法制备了金属 半导体场效应晶体管 (MESFET) ,其单位跨导可达 2 0 0ms mm .该器件已经达到GaAs同质结构的器件水平 【英文摘要】 High quality GaAs epilayers have been grown on InP substract by low pressure metal organic vapour phase epitaxy (LP-MOVPE). The measurement of Raman and PL spectra of GaAs epilayer have been carried out, which show a red shift of the LO mode resulting from the stretching stress in the GaAs epilayer; Strong PL spectrum peak, GaAs charateristic excition peak and corresponding impurity excition peak have also been detected at 16 K, which indicates the crystal is in good quality.     钛合金表面渗氮层与TiAlN耦合涂层的结构及性能

    研究了在钛合金表面渗氮-离子镀TiAlN膜复合处理与单一离子镀TiAlN膜处理工艺对钛合金力学性能、海水环境下的摩擦学的影响。采用SEM S4800、XRD、XPS、纳米压痕仪,观察了薄膜的表面及截面形貌,分析了渗氮后的钛合金表面、单涂层及复合涂层的相组成变化,化学成分和元素价态,测试了两种样品硬度随深度的变化。结果表明,复合涂层中择优取向(200)晶面明显,复合涂层渗氮后基底硬度增加,薄膜硬度下降速度慢,有利于提高涂层的耐磨性,且在海水润滑作用下,短时间摩擦过程中复合涂层的磨损量最小

    Characterization of Coenzyme Q and Preliminary Investigation on Its Effect on Life-span in Drosophila melanogaster

    辅酶Q是存在于线粒体内膜上的脂溶性电子传递体,它是呼吸链的重要组成部分.辅酶Q在不同生物体内其侧链异戊二烯单位的数目不同,目前对黑腹果蝇(drOSOPHIlA MElAnOgASTEr)中辅酶Q的存在形式和含量还不清楚.利用高效液相色谱法分析鉴定了模式生物黑腹果蝇中辅酶Q的种类及含量.实验结果表明:黑腹果蝇中辅酶Q9与Q10并存,辅酶Q9的含量约为Q10的2.5倍.辅酶Q含量具性别差异,其中辅酶Q9在雌性黑腹果蝇中含量较雄性约高30%,而辅酶Q10在雌性果蝇中则比雄性果蝇高约33%;辅酶Q9与Q10含量在辅酶Q合成酶COQ2的突变体中明显降低并且COQ2突变体的寿命较野生型略短.这些实验结果将为利用黑腹果蝇模型研究辅酶Q的功能提供重要基础.Coenzyme Q is an essential component of the mitochondrial respiratory chain as a lipid-soluble electron transporter located in the inner membrane of mitochondria.The number of isoprenoid subunits in the coenzyme Q side chain varies in different species.The content of coenzyme Q in Drosophila has not been reported yet.In the present report,we analyzed the content of coenzyme Q in adult Drosophila by HPLC.We provided evidence that Drosophila contains both CoQ9 and CoQ10.The amount of CoQ9 is about 2.5 folds than that of CoQ10.We further showed the content of CoQ9 and CoQ10 in females are respectively about 30%,33% higher than that in males.In addition,both concentrations of CoQ9 and CoQ10 are significantly reduced in the mutant of CoQ synthase gene coq2 which appeared with shortened life-span.These results provided a good basis to further investigate in vivo biological functions of CoQ in Drosophila.国际科技合作项目(2009DFA32760

    Clone and preparation for transgenic flies of tubeworm Ridgeia piscesae TAK1

    Ridgeia piscesae是唯一一种存在于东北太平洋Juan de Fuca Ridge热液区的管状蠕虫,我们成功通过RACE PCR获得Ridgeia piscesae TAK1(Rp TAK1)基因全长c DNA序列.为深入研究TAK1基因在管状蠕虫中的作用,本文通过显微注射获得Rp TAK1转基因果蝇,并在果蝇体内成功过表达Rp TAK1基因.我们发现由Rp TAK1编码的氨基酸序列具有一定保守性,可能暗示其功能上的保守性,同时也暗示TAK1蛋白在物种进化中的保守性.而过表达突变体果蝇的眼睛出现变异,包括眼睛变得粗糙、单眼排列不整齐及整体形状变小,可以看出Rp TAK1基因影响果蝇的机体发育,我们猜测在Rp TAK1基因在管状蠕虫的生长发育过程中也可能起到重要作用.Ridgeia piscesae,the only tubeworm living at the hydrothermal vents on the Juan de Fuca Ridge,northern Pacific. We successfully got the full-length c DNA sequence of Ridgeia piscesae TAK1. To study the roles TAK1 gene in R. piscesae,we used microinjection to get R. piscesae TAK1 transgenic flies and overexpress R. piscesae TAK1 gene in vivo. We found that the considerably conservative amino acid sequence of R. piscesae TAK1 seemed to imply an evolutionary conserved role for Rp TAK1 and the eye of R. piscesae TAK1 overexpression mutants varied,including shape,appearance and arrangement of ocellus.
