The single tube quantitative rapid detection system for coliform
建立了单管定量检测食品中大肠菌群的快速检测系统。通过光子接收器采集样品在410; nm处吸光度变化数据,由固化在计算机内的程序来实现样品中大肠菌群的单管定量快速检测。结果表明,检测系统可检测出7; cfu/mL(g)样品,菌浓度在7~10~5 cfu/mL(g)范围内具有良好的线性关系。当菌浓度为7~10~5; cfu/mL(g)时所需检测时间为558 ~275; min,检测结果与国标法检测结果之间无显著差异。可满足常见食品样品大肠菌群的单管定量快速检测需求。To address the drawbacks of traditional coliform detection methods,such; as long operation time, tedious steps, huge workload and other issues,; the authorspropose a rapid, sensitive and accurate single tube; quantitative detection system for the determination of coliform in food.; The sample absorbance at 410 nm was collected by photon receiver, and; acomputer program was developed to achieve single tube quantitative; rapid detection of the coliform sample. The results show that the; detection system has high sensitivity, the limit of detection is 7; cfu/mL (g),and this system hasa good linear relationship with the; bacterial concentration in the range of 7 ~10~5 cfu/mL (g). The; detection time is only 558 ~ 275 min for the bacteria concentration of 7; ~10~5 cfu/mL (g),and no significant difference is found between the; results of this detection system and the national standard method. Research on the Fluorescence Characteristics of Synechococcus sp.and Salt Algae
[目的]研究聚球藻和盐藻的荧光特性,确定其在特征激发、发射波长下藻浓度和荧光强度的关系,寻找一种确定藻类浓度的简便方法。[方法]以岛津rf5301型荧光分光光度计分析检测聚球藻和盐藻的激发和发射光谱,并获得它们的荧光强度与以血球计数板计数的藻浓度之间的关系。[结果]聚球藻和盐藻的激发、发射光谱有着明显的不同。在它们特征的激发、发射波长下,其浓度与荧光强度呈良好的线性关系。[结论]通过测定藻类的荧光强度就可推算出特定藻类的浓度,且该方法比血球计数板计算藻类的浓度更为便捷。[Objective] A simple determining method of the concentration of the known algae:Synechococcus sp.and salt algae,was formed through the confirmation of the relationship between its concentration and fluorescence intensity under its characteristics of excitation and emission wavelength based on research on its fluorescence characteristics.[Method] The excitation and emission spectra of Synechococcus sp.and salt algae was measured with the fluorescence spectrophotometer(RF5301) and the relationship between the fluorescence intensity and the concentration of alga based on the number at blood counting plate was gotten.[Results] There was a marked difference in the excitation and emission spectra between Synechococcus sp.and salt algae and there was good linear relationship between its concentration and the fluorescence intensity under its special excitation and emission wavelength.[Conclusion] The concentration of specific algae could be calculated through the determination of its fluorescence intensity,and the method was more convenient in the concentration-testing of algae than that of the blood cell counting sheet

Establishment of Single Tube Quantitative Detection Technology for Coliforms Assay Based on Time of Growth Spectrophotometer Method
通过研究大肠菌群初始接种浓度与达到0.2吸光度所需时间的关系,建立基于生长时间光谱法的大肠菌群单管定量检测技术。通过与多管发酵法比较发现单管定量检测技术的相对标准偏差小于传统的多管发酵法,而且反应所需的时间短,当初始菌浓为101.3Cfu/Ml时反应时间仅需9H,可以作为大肠菌群快速检测的方法,具有良好的应用前景。By studying the initial inoculation concentration of coliforms and the time required to reach 0.2 absorbance,and establishment of single tube quantitative detection technology for coliforms assay based on time of growthspectrophotometer method.By comparison with the multi-tube fermentation method of quantitative detection of single-tube technology that the relative standard deviation less than the conventional multi-tube fermentation method,and the reaction time is short,when the initial bacterial concentration was 101.3cfu/mL,the reaction time was only 9h,can serve as a method of rapid detection of coliforms,has a good prospect.厦门市科技计划创新项目(3502Z20103046
