    基于大数据平台的城市灾害社会恢复力指标初探——以“莫兰蒂”台风为例

    气候变化导致许多灾害的发生频率加快,破坏强度增加,提高城市的韧性是当前应对气候变化的重要途径。作为韧性的核心内容,社会恢复力对于灾后恢复评估及灾害的社会影响评估具有重要意义。本文以大数据为背景,关注公众心理恢复,探究反映社会恢复能力的指标及其与受灾情况之间的关系,对厦门市展开了实证分析。研究结果表明:厦门市的公众心理恢复时间最长,三明市最短;公众对莫兰蒂台风的关注度与其登陆的时间和地点有关,随台风在时空上的逼近而增高;表征关注度的公众搜索指数和公众心理恢复时间这两个反映社会恢复能力的指标都表现出与受灾情况良好的正相关性,可以作为社会恢复力的评价指标,但各自的影响机制不同,有待深入研究

    阿胶-菊粉饮料制备工艺研究

    以阿胶为主料,菊粉、果粉、甜味剂等为辅料,依次经过原料预处理、熬煮、过滤、杀菌后得到阿胶-菊粉饮 料。本文概述了阿胶-菊粉饮料的制备工艺流程和操作要点,研究了阿胶-菊粉饮料促进小鼠肠道蠕动的功能。研究结果表明:阿胶、菊粉、木糖醇、果粉、甜菊糖的添加量分别为1% ~2%、3% ~10%、3% ~5%、5% ~10%、0.005%,经过预处理、熬煮、过滤、杀菌后可制备风味俱佳,稳定性好的产品;通过小鼠小肠墨汁推进率实验证明该产品具有较好的促进肠道蠕动的效果

    Development and application of CK-MB specific monoclonal antibodies

    本研究拟建立肌酸激酶同工酶MB(CK-MB)特异性单克隆抗体(mAb)的研制方法,对抗CK-MB单抗进行评价分类及性质鉴定,并初步建立CK-MB; 定量检测试剂。以CK-MB抗原免疫BALB/c小鼠,利用常规单抗制备技术,使用间接和捕获ELISA差异筛选法筛选单抗。利用肌酸激酶同工酶(CK-; MM/BB/MB)抗原对所制备单抗的抗原识别表位进行鉴定,另通过免疫印迹法及合成CK-MM、CK-BB差异性的线性表位肽鉴定对所制备的单抗进行评; 价分类。使用双抗体夹心ELISA方法筛选检测CK-MB抗原的配对mAb,并初步建立CK-MB定量检测试剂。使用74例临床标本初步评价该试剂与罗氏; 试剂的检测一致性。最终,我们成功筛选到22株稳定分泌抗CK-MB抗体的杂交瘤细胞株,这些单抗可以分为线性、偏构象的CK-MB和CK-MM或者CK; -BB交叉的单抗以及与CK-MB特异反应的偏构象型单抗,并使用偏构象型单抗研制出CK-MB定量检测试剂,该试剂与罗氏试剂相关系数r达到0.930; 9。综上所述,本研究建立了研制CK-MB偏构象型特异性单抗的筛选方法,通过对所筛选的单抗进行分析鉴定并建立了CK-MB定量检测试剂,与罗氏试剂检; 测结果符合率高。The aim of this study is to develop creatine kinase isoenzyme MB (CK-MB); specific monoclonal antibodies (mAb), and characterize the monoclonal; antibody and further development of quantitative detection assay for; CK-MB. The BALB/c mice were immunized with purchased CK-MB antigen, then; monoclonal antibodies were prepared according to conventional hybridoma; technique and screened by indirect and capture ELISA method. To identify; the epitopes and evaluate the classification, purchased creatine kinase; isoenzyme MB (CK-MM/BB/MB) antigen was used to identify the epitopes,; with immunoblotting and synthetic CK-MM and CK-BB in different linear; epitope. A double antibody sandwich ELISA was applied to screen the mAb; pairs for CK-MB detection, and the quantitative detection assay for; CK-MB was developed. We used 74 cases of clinical specimens for; comparison of our assay with Roche's CK-MB assay. We successfully; developed 22 strains of hybridoms against CK-MB, these mAbs can be; divided into linear, partial conformational CK-MB, CK-MM or CK-BB cross; monoclonal antibody and CK-MB specific reaction with partial; conformational monoclonal antibody, and CK-MB quantitative detection; assay was developed by using partial conformational monoclonal antibody.; The correlation coefficient factor r of our reagent and Roche's was; 0.930 9. This study established a screening method for CK-MB partial; conformational specific monoclonal antibody, and these monoclonal; antibodies were analyzed and an established quantitative detection assay; was developed.     70年来中国化学海洋学研究的主要进展

    Development of detection kit for T cell infected with Mycobacterium tuberculosis based on fullautomatic chemiluminescence immune analyzer

    目的建立基于化学发光平台的人结核感染T细胞检测方法。方法将一株γ-干扰素单抗标记在磁微粒上,另一株γ-干扰素单抗标记在吖啶酯上,然后将检测体系与已有的细胞刺激培养体系相结合。结果本研究成功建立基于化学发光平台的人结核感染T细胞检测方法。以ElISA平台检测试剂的检测结果作为参考,该方法的灵敏度为98.3%,特异性为99.2%,总体符合率达到98.8%。结论该方法具有更高的分析灵敏度(可达0.27 Pg/M l)、更宽的线性范围(1 Pg/M l~5 000 Pg/M l)、更好的重复性(批内与批间变异系数均<6.0%)及更易实现高通量检测,为临床诊断结核感染提供了有力的工具。Objective To develop the detection kit for T cell infected with Mycobacterium tuberculosis based on full-automatic chemiluminescence immune analyzer.Mehtods An anti IFN-γ Mab was coated on the surface of microparticle.Another anti IFN-γ Mab was labelled to acridinium ester.After that, the detection system was combined with the in vitro cell culture system.Results This research developed the detection kit for T cell infected with Mycobacterium tuberculosis based on full-automatic chemiluminescence immune analyzer.Compared with the testing results of the ELISA kit, the sensitivity, specificity and total matching ratio of the CLIA kit was 98.3%, 99.2% and 98.8%, respectively.Conclusion The CLIA kit has better sensitivity(0.27 pg/m L), wilder linear range(1 pg/m L ~ 5 000 pg/m L), and better repeatability(intra and inter coefficient of variation < 6.0%).It makes a high throughput detection available.It will contribute to the clinical diagnosis of Mycobacterium tuberculosis.国家高技术研究发展计划(863计划)(2011AA02A101

    Application of Mycobacterium Tuberculosis Specific Protein InterferonGamma Release Assay in Tuberculosis Diagnosis and Therapeutic Monitoring

    目的:目的:探讨结核分枝杆菌相关γ-干扰素释放试验(Tb-IgrA)在结核病诊断及临床抗结核治疗效果评价中的应用价值。方法:对196例临床确诊结核患者进行相关病史收集,同时进行Tb-IgrA、结核菌素皮肤试验、结核杆菌培养、涂片镜检、结核抗体检测,结核患者的治疗过程中按用药疗程(初治患者第2、5、6个月,复治患者第2、5、8个月)对其进行随访,在治疗结束后6个月和1年后各随访一次,比较Tb-IgrA与结核菌素皮肤试验在治疗过程中的变化。结果:确诊结核病例196例,其中肺结核158例,肺外结核38例,Tb-IgrA在结核诊断中灵敏度为82.1%,特异性为91.7%,阳性预测值为93.1%,阴性预测值为79.3%,与传统实验室检查方法相比有显著性差异(P<0.01)。在结核患者的抗结核治疗过程中,Tb-IgrA阳性率及浓度水平治疗前与治疗后2、6月以及复治后2、5、8月比较显著下降(P<0.01),Tb-IgrA水平与结核分枝杆菌数量存在较好相关性,与传统的TST方法评估疗效相比有显著性差异(P<0.01)。结论:Tb-IgrA在结核诊断中敏感性和阳性预测值较高,可作为较好的结核诊断的依据,同时Tb-IgrA对肺外结核的灵敏度较高,为肺外结核患者的诊断提供了较为可靠的依据,通过治疗过程Tb-IgrA检测分析,外周血Tb-IgrA的变化随着抗结核治疗的进程下降的趋势明显,与结核涂片镜检结果符合率较高,是临床疗效评估较为理想的指标。Objective To investigate the applicative value of mycobacterium tuberculosis specific protein interferon gamma release test in early diagnosis of tuberculosis and the evaluation of clinical efficacy.Methods 196 clinical diagnosis of tuberculosis patients were related to the history collection,and detected by TB-IGRA simueltaneously,tuberculin skin test( purified protein derivative of tuberculin 5 units),tubercle bacillus culture,directly smear,tuberculosis antibody detection.Treatment of tuberculosis patients were followed up( 2,5,6 months,retreatment patients at 2,5,8 months),in 6 months and 1 years after the end of treatment followup once.Compared with difference between the IGRA and tuberculin skin test and other traditional laboratory results.Results Confirmed tuberculosis cases in 196 cases,including 158 cases of pulmonary tuberculosis,38 cases of extra-pulmonary tuberculosis.The sensitivity of TB-IGRA in the pulmonary tuberculosis and extra-pulmonary tuberculosis showed 81% and 86.8% respectively,at the same time,TB-IGRA in the diagnosis of tuberculosis,the negative predictive value express 86.3%,specificity was 91.7%,and the positive predictive value was 88.2%.In anti-tuberculosis treatment the positive rate of TB-IGRA and concentration levels before treatment and after treatment 2,5,6months,retreatment 2,5,8months decreased significantly( P < 0.01).There is better correlation TB-IGRA concentration level with number of mycobacterium tuberculosis.It is significant difference compared to TB-IGRA with the traditional TST method to the evaluation of clinical treatment( P < 0.01).Conclusion The sensitivity of TB-IGRA in the diagnosis of tuberculosis and the postive predictive value is higher; it can be used as a good basis for the diagnosis of tuberculosis.TB-IGRA provides a more reliable diagnosis method in patients with extra-pulmonary tuberculosis.Peripheral blood TB-IGRA decreased significantly in anti- tuberculosis treatment process.Coincidence rate between TB-IGRA concentration level and number of mycobacterium tuberculosis is higher.It is an ideal index to evaluate the clinical efficacy.浙江省医药卫生科技计划项目(2009A167

    苯硫酚对固氮酶催化活性的影响

    研究了苯硫酚对棕色固氮菌固氮酶催化底物还原活性的影响。结果表明:当反应体系中固氮酶钼铁蛋白与苯硫酚的摩尔比为1:4时,固氮酶的乙炔还原活性比对照组下降了62.3%,下降的幅度随着苯硫酚浓度的升高而增大;但在乙炔存在的情况下,固氮酶的放H_2活性随着苯硫酚的浓度的升高而升高。在氩气氛下,苯硫酚浓度的升高对固氮酶放H_2活性的影响不明显。这一现象有可能是由于苯硫酚取代与铁钼辅基的Fe_1原子连接的半胱氨酸的巯基而引起的

    一种A型流感病毒NP抗原快速检测试剂的建立

    目的建立一种适合现场检测需要的A型流感快速诊断试剂。方法以自制的抗A型流感(FluA)病毒核蛋白(NP)单抗为原料,建立快速检测A型流感病毒的双抗体夹心酶免疫渗滤试验,并对其灵敏度和特异性进行了初步评价。结果该试剂对不同地区流行的各种亚型的A型流感病毒株均有较高的反应性,而对非FluA病毒株无交叉反应。比较该试剂与BD公司的两种流感快速诊断试剂,发现该试剂对随机选取的3株FluA病毒的检测分析灵敏度高出Directigen EZ Flu A试剂5~125倍,对2株FluA病毒的分析灵敏度高出Directigen Flu A试剂约20倍。另外,用该试剂对57份含漱液标本和170份动物拭子标本进行检测,结果显示:本试剂的灵敏度(>85%)和特异性(>95%)均优于当前主流的商品化A型流感快速诊断试剂。结论利用抗FluANP单抗为原料建立了A型流感快速诊断试剂,该试剂的应用无需任何专用仪器,操作简便快速,可满足现场检测需要
