    Inhibitory effect of NRP-1mAb on the growth of gastric cancer cell BGC-823

    目的观察自主研发的抗NRP-1b1/b2IgG单克隆抗体(NRP-1mAb)对胃癌BGC-823细胞生长的影响,并初步探讨可能的作用机制。方法实验室制备NRP-1mAb,采用SDS-PAGE检测纯度。采用0、25、100、200、400μg/ml NRP-1mAb培养胃癌BGC-823细胞,光学显微镜下观察细胞形态学变化;采用MTT法、集落形成实验、流式细胞术观察胃癌细胞BGC-823的增殖、集落形成和凋亡的情况;Western blotting法检测NRP-1mAb作用后Akt、p38、ERK、JNK信号蛋白磷酸化情况。结果SDS-PAGE检测显示,NRP-1mAb的纯度在95%以上。光学显微镜观察显示,NRP-1mAb作用后,BGC-823细胞形态呈凋亡改变。MTT实验显示,NRP-1mAb抑制BGC-823细胞的增殖作用呈浓度和时间依赖性(P〈0.01)。集落形成实验显示,不同浓度NRP-1mAb均能显著抑制BGC-823细胞的集落形成,其抑制率呈浓度依赖性(P〈0.01)。流式细胞术检测显示,不同浓度NRP-1mAb均明显促进BGC-823细胞凋亡,且大部分集中在早期凋亡。Western blotting检测显示,BGC-823细胞的Akt磷酸化受到抑制,ERK、p38、JNK的磷酸化水平无明显变化。结论NRP-1mAb能抑制胃癌细胞BGC-823的生长、促进凋亡,可能与抑制Akt磷酸化有关。Objective To observe the effect of NRP-1b1/b2 IgG monoclonal antibody( NRP-1) on the growth of gastric cancer cell BGC-823,and to explore the possible mechanism of the antibody. Methods NRP-1mAb was prepared in laboratory,and the purity of antibody was detected by SDS-PAGE. Gastric cancer BGC-823 cells were cultured by 0,25,100,200,400 μg/ml NRP-1 mAb. The morphological changes of BGC-823 cells were observed by microscope. The proliferation,colony formation and apoptosis of gastric cancer BGC-823 cells were observed by MTT assay,colony forming assay and flow cytometry. The phosphorylation of related signal proteins was detected by Western blotting analysis. Results SDS-PAGE test showed that the purity of NRP-1mAb was above95%. Microscopy showed apoptotic changes of BGC-823 cells treated by NRP-1mAb. MTT assay showed that NRP-1mAb could inhibit the proliferation of BGC-823 cells in a time and dose dependent manner( P〈0. 01). Colony forming assay showed that different doses of NRP-1mAb could inhibit the colony formation of BGC-823 cells in a dose dependent manner( P〈0. 01). Flow cytometry showed that different doses of NRP-1mAb could promote the apoptosis of BGC-823 cells mainly at early apoptosis stage. It was found that the level of Akt phosphorylation was decreased after treated by NRP-1mAb,and there was no significant phosphorylation of ERK,p38 and JNK protein. Conclusion NRP-1mAb can inhibit the growth of gastric cancer cell BGC-823 and promote apoptosis,which may be related to the inhibition of Akt phosphorylation.厦门市科技计划创新资助项目(3502z20134026,3502z20144034

    Study on the Nutrient Content of Bamboo Leaves and Soil in Coastal Region of Fujian

    第一作者简介:陈松河,男, 1968年出生,福建长泰人,研究员,硕士生导师,硕士,主要从事园林植物(竹类)分类、栽培与抗逆性研究。通信地址: 361003厦门市思明区虎园路 25号, Tel:0592-2039619,E-mail:[email protected]。[中文文摘]为了研究福建滨海地区竹类叶片与土壤养分状况,对福建滨海3地厦门园博苑百竹园、莆田赤港华侨农场竹园和莆田湄洲岛竹类植物叶片和土壤养分进行实地调查取样分析。结果表明:3地17种竹类成熟叶片和衰老叶片氮(N)、磷(P)含量存在显著差异,且存在明显的地域性差异;竹类生长土壤氮(N)含量均值大小依次为厦门园博苑0.3697mg/g、莆田赤港0.2820mg/g、莆田湄洲岛0.1921mg/g;磷(P)含量均值大小依次为莆田湄洲岛0.3926mg/g、厦门园博苑0.3134mg/g、莆田赤港0.2152mg/g;莆田赤港和莆田湄洲岛10种竹类成熟叶片叶绿素a含量(Ca)和叶绿素b含量(Cb)均值比达到2.17,Ca和Cb间经11种单因子模型拟合,最优的拟合方程为:YA=0.9699+3.7374B-0.7166B2+0.0439B(3A代表Ca,B代表Cb),相关系数的平方达0.934,呈极显著相关关系;竹类植物盐害等级为0级的有5种,盐害等级为1级的有10种,盐害等级为2级的有2种,盐害等级为3级以上的未见。[英文文摘]In order to study nutrient status in bamboo leaves and soil,3 coastal region in Fujian, Xiamen Garden & Flower Expo (XMYB), Farm for Returned Overseas Chinese in Putian (PTCG) and Meizhou Island of Putian (PTMZ) were studied in this paper by field investigation and sampling analysis. The results were shown as follows: the content N and content P in mature and senescent of 17 species leaves in 3 coastal region were significant difference, also significant difference in region. The average value content N of bamboo soil inXMYB, PTCG, PTMZ were 0.3697, 0.2820, 0.1921 mg/g in turn. The average value content P of bamboo soil inPTMZ, XMYB, PTCG were 0.3926, 0.3134, 0.2152 mg/g in turn. The proportion of 10 mature bamboo speciesaverage value between content Ca and content Cb was 2.17, through 11 models of single factor analysis, the relation between the bamboo’s chlorophyll a content and chlorophyll b content accorded with model cubic: YA= 0.9699 + 3.7374B-0.7166B2 + 0.0439B3 (A stands for content Ca, B stands for content Cb), the square of itscorrelation coefficient was 0.934, it was very significant.     Application of Color Calibration Methods for the Tongue-color Images under Nature Indoor Light

    目的建立一种室内非直射自然光条件下的医学图像颜色校正分析方法并初步应用。方法根据色彩还原算法结合自制彩色、灰度色标作为校正的监督色标建立TrM颜色校正方法,并对410幅舌色图像进行了校正。结果舌色A*、b*值除淡白舌组外,校正前后各组均有显著性差异(P<0.05),校正后A*、b*值离散度和变异系数较校正前减小。49例典型舌象校正后fISHEr'S分类方法与人工分类比较总体一致率75.5%,一致性检验结果kAPPA值为0.675(P<0.05)。结论不同舌象颜色经拓扑剖分-影射还原法校正后能较好的体现其l*A*b*颜色特征。拓扑剖分-影射还原模型校正方法能明显降低自然光条件下舌色图像的色差,具有良好的颜色校正效果。OBJECTIVE To establish a calibration analysis approach of medical image color under the circumstance of non-direct natural indoor light,and use it.METHODS TRM(Topology Resolve-Map) calibration method was established in accordance with the color reduction algorithm,with self-made color and grey patches being supervision color control patches;it was applied to correct 410 cases of clinical tongue image.RESULTS The before and after calibrated a *,b * values of each group's tongue color were significantly different(P<0.05) except for that of whitish group.Compared with pre-calibration,the dispersion and variation coefficient of post-calibration a*,b* values decreased.The overall consensus rate was 75.5% after using Fisher's classification method and artificial classification method to analyze 49 corrected typical tongue cases,with the Kappa value of consistency test result of 0.675(P<0.05).CONCLUSION The characteristics of L*a*b* in varied tongue colors can be better demonstrated after the calibration with TRM.TRM model approach can significantly reduce the color errors of tongue images under natural indoor light,which demonstrates its good color calibration effect.国家高技术研究发展计划(2008AA02Z407);国家自然科学基金(30873463;30300443

    生态控制线是生态空间管控的重要举措之一,也是保护城市生态环境和提升生态环境质量的有效手段。但是,由于复杂的产权纠纷和历史遗留问题,它的划定在实际操作过程中面临着诸多挑战。妥善处理生态线内已建社区和合理进行社区改造已成为重要议题。本文以深圳市为例,系统梳理生态线内社区的特征,并引入交易成本的概念,分析生态线内社区改造过程中政府、社区、居民和开发商等利益主体的成本与收益,指出生态线内社区改造的难点在于交易成本过高以及收益与成本缺口巨大,并在此基础上提出一系列针对性的改造策略,以期更好地推进生态线内社区改造,逐步完成生态修复工作,更好地实现生态价值与社会经济价值,并为其他城市提供借鉴和参考
