Shedding light on miRNA targeting through structure
In miRNAs, it is known that seed pairing is an important parameter for targeting, but there are still missing links on why it targets a certain mRNA and how the decision between degradation or translation repression is made.
In our work, we use liquid state NMR spectroscopy, EMSA and UV melting experiments to biophysically characterise the complex of miRNA-34a with the target mRNA of CD44 and PNUTS. Here we present the NMR sample preparation as well as the first steps for the biophysical description of the mRNA.miRNA complexes.
Our EMSA data suggests both miRNA.mRNA complexes are transient and miRNA.gCD44 possibility has a higher Kd then miRNA.PNUTS. Through NMR miRNA.PNUTS complex was deemed more structurally dynamic, yet interestingly with UV melting it had a higher Tm then miRNA.gCD44 complex. Finally, we also performed several simulations on several different mRNA targets with MCFold unveiling a kink on the mRNA as a structural trend which could be a necessity for docking of the duplex into Ago.
Still further studies must be conducted in order to solve the structures of both duplexes in study and a bigger population of simulated structures must be collected
Business intelligence to improve the quality of local government services : case-study in a local government Town Hall
The use of business intelligence (BI) systems by organizations is increasingly considered as an asset, which goal is to provide access to information in a timely manner in order to support the decision-making process. However, in specific cases such as local government organizations, there are very specific challenges. Some of them like privacy rights and applicable law compliance must be carefully observed, making the necessary adaptations of these BI solutions. The developed solution brings some important contributions and represents some advances in the eGovernment context applied to local governments where the information is normally used/stored are not normalized and pre-defined. Being this a big barrier to development of this type of solutions, the developed architecture is prepared to improve the data quality and avoid this type of mistakes. Security in information systems for smart cities
Com o crescimento das populações nos grandes centros urbanos, surgem
cada vez mais problemas ao nível da gestão de recursos das cidades. Graças
à evolução das tecnologias ligadas à internet é possível utilizar essas
tecnologias para auxiliar na solução de problemas, criando assim as cidades
inteligentes. Este trabalho analisa uma solução de cidade inteligente existente
ao nível da segurança dos dados e aplicação e apresenta uma solução
para colmatar as suas falhas e mostrar como desenvolver aplicações com
uma camada de segurança adequada. É apresentada uma solução que tem
em conta a criticalidade dos dados, tal como exigido no novo Regulamento
de Proteção de Dados da União Europeia, e depois é executada a implementação.
A implementação da solução é por fim validada por vários testes de
desempenho e também funcionais o que permite avaliar o nível de sucesso
do trabalho apresentado nesta dissertação.With a significant population growth in large urban centers, the problems in the
management of cities’ resources are rapidly increasing. But due to the evolution
of the technologies connected to the internet it is possible to use those to
aid in the solution of problems, thus creating the so called Smart Cities. This
dissertation examines an existing Smart City solution, and presents a solution
to address its flaws and show how to develop applications with a proper security
layer for the data and application layer. A solution is presented which
takes into account the criticality of the data as required by the new EU Data
Protection Regulation, and then implements using the appropriate technologies.
The implementation of the solution is validated by several performance
and functional tests, which allows to evaluate the level of success of the work

Introdução: O carcinoma da próstata é a neoplasia mais comum e a terceira causa de morte por cancro no homem. Atualmente, existe uma variedade de opções disponíveis para o tratamento do carcinoma da próstata metastizado e resistente à castração (mCRPC) como é o exemplo da abiraterona e da enzalutamida, focados nesta análise.
Objetivo do estudo: Analisar as caraterísticas, o tratamento e outcomes de pacientes com mCRPC tratados com abiraterona ou enzalutamida e correlacionar com a sobrevida livre de progressão e sobrevida média global. Averiguar a influência das variáveis idade, score de Gleason, duração do tratamento antineoplásico anterior e PSA no início do tratamento na resposta à terapêutica.
Metodologia: Foi realizado um estudo retrospetivo baseado em dados de um período pregresso (desde a introdução destes medicamentos no hospital, 19 de Dezembro de 2013 até dia 31 de Julho de 2017) no Centro Hospitalar Cova da Beira. Foram recolhidas diversas variáveis como idade, ECOG, score de Gleason, existência ou não de metastização, locais de metastização, terapêuticas antineoplásicas anteriores, terapêutica associada, comorbilidades, doentes quimio-naive ou pós-quimioterapia, PSA no início do tratamento e a duração do tratamento.
Resultados: A amostra é constituída por 21 doentes com mCRPC sem quimioterapia prévia. A idade média dos doentes foi de 72,1 anos (52–90 anos). Não se verificou uma diferença estatisticamente significativa (p>0,05), entre a abiraterona e a enzalutamida no que concerne à percentagem de doentes com redução de PSA a 6 meses =50%, nem na sobrevida média global (22,0 meses para a abiraterona vs não alcançado para a enzalutamida). De igual modo, não foi observada uma influência significativa (p>0,05) das seguintes variáveis analisadas: a) idade dos doentes na redução da PSA a 6 meses =50%; b) score de Gleason no valor médio e distribuição de valores da redução PSA (%) a 6 meses; c) tratamento antineoplásico anterior =12 meses no valor médio e distribuição de valores da redução PSA (%) a 6 meses; d) valor do PSA no início do tratamento na percentagem de doentes com redução de PSA a 6 meses =50%.
Conclusões: O tratamento com abiraterona ou com enzalutamida não conduziu a diferenças significativas nos outcomes de doentes com mCRPC sem quimioterapia prévia. Do mesmo modo, nenhuma das variáveis analisadas influenciou significativamente os resultados do tratamento.Background: Prostate cancer is the most common malignancy and the third leading cause of cancer death in man. Currently, there are a variety of options available for the treatment of metastatic castration-resistant prostate cancer (mCRPC), as is the case with abiraterone and enzalutamide, focused on this analysis.
Purpose: Analyze the characteristics, treatment and outcomes of patients receiving mCRPC treated with abiraterone or enzalutamide and correlate with progression-free survival and overall mean survival. Check the influence of the variables age, Gleason score, duration of previous antineoplastic treatment and PSA at the beginning of treatment in response to treatment.
Methodology: A retrospective study based on data from a previous period (from the introduction of these drugs in the hospital until July 31, 2017) was performed at our hospital. Several variables such as age, ECOG, Gleason score, metastatic existence or not, metastatic sites, previous antineoplastic therapies, associated therapeutics, comorbidities, chemo-naive or post-chemotherapy patients, PSA at the beginning of treatment and the duration of treatment.
Results: The sample consisted of 21 mCRPC patients without prior chemotherapy. The mean age of the patients was 72.1 years (52-90 years). There was no statistically significant difference (p> 0.05) between abiraterone and enzalutamide in the percentage of patients with a 6 month PSA reduction =50%, or overall mean survival (22.0 months for abiraterone vs not obtained for enzalutamide). Likewise, a significant influence (p> 0.05) of the following analyzed variables was not observed: a) patients' age at 6 month PSA reduction =50%; b) Gleason score in the mean value and distribution of PSA reduction (%) at 6 months; c) previous antineoplastic treatment =12 months in the mean value and distribution of PSA reduction values (%) at 6 months; d) PSA value at baseline in the proportion of patients with 6 month PSA reduction =50%.
Conclusions: Treatment with abiraterone or with enzalutamide did not lead to significant differences in the outcomes of patients with mCRPC without prior chemotherapy. Likewise, none of the analyzed variables significantly influenced treatment outcomes
Modelos Supervisionados Aplicados à Deteção de Fraude em Seguros de Saúde
Estima-se que a fraude na área da saúde represente um problema na ordem dos 3%
a 10% dos orçamentos de alguns países para este setor e que, no caso particular dos
seguros de saúde, os prejuízos ascendam a vários milhões de euros. Este problema tem
levado estados e seguradoras a implementar Sistemas de Deteção de Fraude sofisticados,
compostos por ferramentas automáticas que procuram identificar os padrões de fraude
conhecidos seguido de um processo manual de inspeção por especialistas.
Contudo, estes sistemas apresentam várias limitações, e algumas técnicas de deteção
de fraude baseadas em modelos data driven têm vindo a ser incorporadas nestes sistemas.
Apesar de se terem revelado eficientes, a natureza dos datasets deste setor - o conjunto
de pedidos de reembolso feitos às seguradoras - traz também muitos desafios à sua implementação,
nomeadamente a distribuição enviesada de classes (uma proporção muito
elevada de pedidos de reembolso legítimos face aos pedidos de reembolso suspeitos) ou
o concept drift (a natureza dos padrões de fraude muda com o tempo). Estas características
dificultam a aplicação de técnicas de aprendizagem automática e são necessárias
abordagens específicas para a sua resolução.
Nesta tese apresenta-se uma solução de aprendizagem automática supervisionada de
deteção de fraude, solicitada por um grupo internacional privado que faz a gestão dos
seguros de saúde de algumas seguradoras. Usou-se, para esse efeito, o datatset constituído
pelos pedidos de reembolso respeitantes aos anos de 2017 e 2018 que haviam sido classificados
como legítimos ou suspeitos pelos auditores. Foram consideradas 4 famílias de
classificadores - Regressão Logística, Random Forest e Support Vector Machine e XGBoost
e os seus desempenhos foram medidos e comparados.
Os resultados obtidos evidenciaram a utilidade destes classificadores, tendo o Random
Forest e o XGBoost apresentado melhores resultados
Desenvolvimento de ferramenta de apoio ao dimensionamento de sistemas de autoconsumo fotovoltaico em ambiente doméstico
Nos últimos anos, tem-se assistido a uma maior preocupação com o meio ambiente, a atual
conjuntura mundial está cada vez mais direcionada para a eficiência energética e para a
utilização de fontes de energias renováveis. Os principais governos mundiais, incluindo o
português, já perceberam a necessidade de enveredar por esse caminho e nesse sentido
aplicam medidas que direcionam e consciencializam a população para a eficiência energética
e para as energias renováveis.
Em Portugal, o setor das energias renováveis assume atualmente uma posição de extrema
importância, resultante da expressão que governo português tem vindo a implementar no
panorama energético nacional, da qual resulta uma importante contribuição para o
desenvolvimento económico, na criação de riqueza e geração de emprego. Neste contexto, e
no caso particular da energia fotovoltaica têm sido implementadas medidas que incentivam
a aposta nesta tecnologia, prova disso é o Decreto-Lei n.º 153/2014 aprovado em conselho
de ministros em Setembro de 2014, que promove essencialmente o autoconsumo.
O autoconsumo consiste na utilização de painéis fotovoltaicos para produção de energia
elétrica para consumo próprio com ou sem recurso a equipamentos de acumulação. Em
termos práticos, este sistema permite que os consumidores produzam a sua própria energia
através de uma fonte renovável ao invés de adquirir essa energia na rede elétrica de serviço
público.
As políticas de incentivo ao autoconsumo proporcionam uma oportunidade para os
consumidores interessados em investir na produção da própria energia elétrica, neste sentido
e de forma a ajudar no dimensionamento de unidades de produção de autoconsumo foi
desenvolvida, no âmbito desta tese, uma ferramenta de apoio ao dimensionamento de
sistemas de autoconsumo fotovoltaico sem acumulação em ambiente doméstico, com o
objetivo de estimar as necessidades de potência fotovoltaica a instalar em habitações de
baixa tensão normal.
Na base da construção desta ferramenta estiveram essencialmente os perfis de consumo,
aprovados pela Entidade Reguladora dos Serviços Energéticos, de todos os clientes finais que não dispõem de equipamento de medição com registo de consumos e também a
estimativa de produção fotovoltaica desenvolvida pelo Centro Comum de Investigação da
Comissão Europeia.
A aplicação desenvolvida tem como principal funcionalidade proporcionar ao utilizador o
dimensionamento de unidades de produção de autoconsumo fotovoltaico, mediante a
introdução de alguns dados tais como o distrito, a potência contratada, a tarifa e o consumo
energético anual. Esta aplicação apresenta resultados relativos ao dimensionamento do
sistema, como é o caso da potência a instalar e da estimativa de produção fotovoltaica anual,
e resultados relativos à análise económica do sistema como é o caso do valor atual líquido,
da taxa interna de rentabilidade e do payback do investimento.Over the last few years we have seen greater concern with the environment, the current world
situation is increasingly directed to energy efficiency and the use of renewable energy
sources. The world's leading governments, including the Portuguese, have realized the need
to go in this direction applying measures that drive and rise the population’s consciousness
for energy efficiency and renewable energy.
In Portugal, the renewable energy sector currently takes a position of utmost importance,
result of the expression that the Portuguese government has been implementing in the
national energy panorama, which shows an important contribution to economic
development, to increase wealth and employment creation. In this context, and in the
particular case of photovoltaic have been implemented measures to encourage the betting on
this technology, proof of this is Decree-Law No. 153/2014 approved by the Council of
Ministers in September 2014, which essentially promotes photovoltaic self-consumption.
The photovoltaic self-consumption consists of using photovoltaic panels to produce
electricity for own consumption with or without an accumulation equipment. In practical
terms, this system allows consumers to produce their own energy through a renewable source
rather than to purchase electricity on the power distribution grid.
Self-consumption encouraging policies provide an opportunity for consumers interested in
investing in the production of own electricity, in this sense and in order to help in the design
of self-consumption production units, it has been developed as part of this thesis, a support
tool for the design of PV self-consumption systems without accumulation in domestic
environment to estimate the photovoltaic power needs that should be installed in a standard
low voltage housing.
In the basis of the construction of this tool were essentially consumption profiles, approved
by the Regulatory Authority for Energy Services, of all final users who do not have
measuring equipment with consumption registration and also photovoltaic production
estimate developed by the Joint Research Centre of the European Commission. This application's main functionality provides the user with the sizing of PV selfconsumption
production units by introducing some data such as the district, the contracted
power, the rate and the annual energy consumption. This application presents the results for
sizing of the system, such as the power to be installed, and the estimated annual photovoltaic
production, and results on economic analysis system such as net present value, the internal
rate of return and payback on investment
Growing from incipient to potentially large cycle networks: screening the road network of the consolidated urban area of Lisbon
Many arguments sustaining active modes of transport are related to ameliorating urban life. Biking should be one option for our daily trips including utilitarian trips and not only trips for leisure. Accordingly, designing cycle networks that meet citizens’ requirements for daily trips is important to increase modal share. Particularly, it should take into account coherence, directness and attractiveness/comfort, not overlooking safety issues towards other road vehicles. Furthermore, it must consider also the current limitations of existing roads (lane width or gradient), in order not to hinder excessively existing road traffic while rearranging the carriageway for suitable cycle network. These objectives become more challenging when aiming to enlarge an incipient cycle network and make it grow to a large and ubiquitous one, in hilly and consolidated urban areas. The present research proposes a practice-ready method to be implemented for screening the existing road network to potentially rearrange the existing carriageway and accommodate a cycling network in urban areas with such characteristics. The methodology suggests also key indicators to analyze the potential performance of the cycle network, at an early planning stage. Although applied to Lisbon, the cycle network screening method is potentially relevant for countries where major urban agglomerations stand in hilly land and still want to see their cyclists grow in the next years. We conclude that it is possible to potentially fit a large cycle network of almost 20% of Lisbon’s total road network. The possible network configurations obtained are assessed in terms of coherence, directness and attractiveness/comfort for bikers.
