    Accurate PCR detection of influenza A/B and respiratory syncytial viruses by use of Cepheid Xpert Flu+RSV Xpress Assay in point-of-care settings: Comparison to Prodesse ProFlu+

    ABSTRACT The Xpert Flu+RSV Xpress Assay is a fast, automated in vitro diagnostic test for qualitative detection and differentiation of influenza A and B viruses and respiratory syncytial virus (RSV) performed on the Cepheid GeneXpert Xpress System. The objective of this study was to establish performance characteristics of the Xpert Flu+RSV Xpress Assay compared to those of the Prodesse ProFlu+ real-time reverse transcription-PCR (RT-PCR) assay (ProFlu+) for the detection of influenza A and B viruses as well as RSV in a Clinical Laboratory Improvement Amendments (CLIA)-waived (CW) setting. Overall, the assay, using fresh and frozen nasopharyngeal (NP) swabs, demonstrated high concordance with results of the ProFlu+ assay in the combined CW and non-CW settings with positive percent agreements (PPA) (100%, 100%, and 97.1%) and negative percent agreements (NPA) (95.2%, 99.5%, and 99.6%) for influenza A and B viruses and RSV, respectively. In conclusion, this multicenter study using the Cepheid Xpert Flu+RSV Xpress Assay demonstrated high sensitivities and specificities for influenza A and B viruses and RSV in ∼60 min for use at the point-of-care in the CW setting. </jats:p

    Man-made maternity roost selection by Scotophilus kuhlii (Lesser Asiatic yellow bat) in two anthropogenically altered habitats

    Scotophilus kuhlii is a highly gregarious bat that thrives in anthropogenically altered habitats readily roosting in man-made structures. Although widespread, information on the ecology of S. kuhlii is very scarce in Southeast Asia. We conducted this study to determine if the females used the same day roost as a maternity roost in buildings in an agricultural and suburban habitat from July 2013 to April 2015. The females were found to use the same day roost as a maternity roost as the buildings provided a higher temperature and more stable thermal conditions as compared to the ambient temperature. We found that there was a significant positive relationship between roost microclimate and ambient microclimate. Based on the observation of the external body condition, females were found to be seasonally monoestrous. Our findings are useful for conservation and management of these synantrophic bats where natural roost sites are lacking and in managing human-bat conflicts

    Isolation and characterization of antioxidant constituents of the fruit of Telfairia occidentalis Hook F (Cucurbitaceae)

    Purpose: To evaluate the antioxidant property of the fruit of Telfairia occidentalis and isolate the components responsible for the antioxidant activity.Methods: The fruit pericarp was macerated with methanol and the extract obtained successively partitioned with n-hexane, dichloromethane and ethyl acetate. The in vitro antioxidant activity of the extract and fractions was evaluated using 2,2-diphenyl-1-picryl hydrazyl (DPPH) radical scavenging, reducing power, nitric oxide scavenging, total antioxidant and hydrogen peroxide scavenging assays. The n-hexane fraction, which had the highest DPPH scavenging and total antioxidant activities, was subjected to column and thin layer chromatography to isolate the components. The isolated compounds were identified by ultraviolet-visible (UV), nuclear magnetic resonance (NMR), Fourier transform infrared (FTIR) spectroscopy and mass spectrometry.Results: Among the fractions tested, n-hexane had the best total antioxidant activity of 99.44 % at 20 mg/ml (p &lt; 0.05) compared to ascorbic acid at 99.71 % of 20 mg/ml. This fraction also had the highest DPPH radical scavenging activity of all the fractions (p &lt; 0.05) at all test concentrations. For nitric oxide scavenging activity, the whole extract, and the chloroform and aqueous fractions exhibited activity ranging from 92.29 to 97.66 % compared to 98.93 % for ascorbic acid. The hydrogen peroxide scavenging activity of the extract and fractions ranged from 92.60 to 96.23 % compared with of the standard, ascorbic acid (101.68 %). The major components of the n-hexane fractions were α- amyrin and β-amyrin.Conclusion: The fruit pericarp of Telfairia occidentalis possesses good DPPH radical scavenging activity. This is the first time the antioxidant activity of the fruit (pericarp) and the presence of α- and β- amyrins in Telfairia occidentalis have been reported.Keywords: Telfairia occidentalis, Fluted pumpkin, Amyrin, Antioxidant

    Energy efficient electrocoagulation using baffle-plates electrodes for efficient Escherichia Coli removal from Wastewater

    A new electrocoagulation reactor (EC), which utilises the concepts of baffle-plates, has been applied to remove Escherichia coli (E. coli) from wastewater. This new aluminium-based EC reactor utilises perforated baffle-plates electrodes to mix water, which reduces the need for mechanical or magnetic mixers that require extra power to work. This new reactor has been used to treat E. coli containing wastewater samples, considering the effects of different parameters such as treatment time (TT), inter-electrode distance (IED), and current density (CD). A statistical analysis has also been commenced to evaluate the influence of each parameter on the removal of E. coli. Additionally, an economic study has been conducted to assess the operating cost of the new reactor. The outcomes of the experimental work confirmed that the new reactor removes as high as 96% of the E. coli within 20 minutes of electrolysis at IED of 0.5 cm, and CD of 1.5 〖mA/cm〗^2. Additionally, it has been found that the operating cost of the new reactor is 0.11 US $/m3 (for E. coli removal), which is less than operating cost of traditional reactors. Finally, it has been found that the effect of the studied parameters on E. coli removal followed the order: TT>CD>IED

    The Malaysian bat conservation research unit from a national model to an international network

    To have even a small chance of averting the crisis decimating Southeast Asian biodiversity, conservation researchers and practitioners need to provide policy makers with information that can guide their decisions. Particularly needed are realistic projections of the responses of diversity to different planning or policy decisions, and the attendant ecological and economic consequences of these responses. Unfortunately, there is a substantial short-fall between the information needed and our capacity to provide it, but we suggest that collaborative efforts that integrate research, capacity building and outreach can both accelerate knowledge development and transfer within the research community, and promote public understanding and appreciation of biodiversity, particularly of low-profile or unpopular taxa. The Malaysian Bat Conservation Research Unit (MBCRU) was established in 2001 to promote the conservation of Malaysia’s unique but threatened bat diversity through long-term conservation research, capacity building and outreach. Here we summarize the main activities of this integrated program, and trace the scale-up of the MBCRU model to the regional level with the launch in 2007 of the Southeast Asian Bat Conservation Research Unit (SEABCRU)

    Volatilitas merupakan refleksi besarnya risiko dari naik turunnya harga saham atau valuta asing. Penelitian ini bertujuan untuk menganalisis pengaruh volatilitas nilai tukar terhadap volatilitas indeks sektor pertambangan saham syariah Indonesia. Pengambilan sampel 10 emiten dilakukan dengan teknik purposive sampling selama periode penelitian 12 Mei 2011 hingga 31 Desember 2019. Metode yang digunakan adalah model EGARCH. Volatilitas nilai tukar ditemukan berpengaruh signifikan terhadap sektor pertambangan saham syariah. Hasil dari penelitian ini memberi bukti empiris dalam memprediksi pergerakan nilai tukar dan indeks sektor pertambangan, serta dapat menjadi sumber informasi bagi pemerintah untuk mempertimbangkan kebijakan insentif terhadap emiten di sektor pertambangan yang sensitif akibat fluktuasi nilai tukar, dan informasi bagi investor dalam mempertimbangan investasi di sektor pertambangan
