“Des-andar lo cotidiano” en el Museo de Antropología (FFYH-UNC) en la Noche de los Museos 2020

In this work we want to narrate, systematize and interpret the anthropological intervention carried out in the educational proposal "Un-walk the everyday. Tell us about your experiences in the pandemic.” We built it for the "Night of the Museums" in the City of Córdoba (Argentina), which took place on November 20, 2020, and had the motto "Museums for diversity. Dialogues from virtuality” It was planned from the Education Area of the Museum of Anthropology of the Faculty of Philosophy and Humanities (FFyH-UNC). As anthropologists we found ourselves being fellow citizens of the world that we wanted to document and learn about, and we were equally affected by the pandemic caused by the COVID19 virus as the visitors. So how to create an activity for the Night of Museums in the midst of preventive social isolation that involved the active participation of visitors mediated by "screens"? How to generate a meeting space to tell what we were experiencing? How to achieve the different, the special trait that this event has in virtuality?En este trabajo queremos narrar, sistematizar e interpretar la intervención antropológica realizada en la propuesta educativa “Des-andar lo cotidiano. Construyendo espacios comunes en el Valle de Ambato, Catamarca, Argentina: Proyecto de valorización y protección del patrimonio arqueológico local

En este artículo se dan a conocer resultados obtenidos en los trabajos realizados junto a miembros de la comunidad educativa de la ciudad de Catamarca y Los Castillos, esta última, situada en el Valle de Ambato. Se trabajó con docentes y alumnos de nivel inicial, medio y superior, con el objetivo de conformar una red de relaciones y acciones, donde los diversos actores intervinientes aporten sus experiencias y saberes tratando de descentralizar la figura del arqueólogo en el proceso investigativo. Los resultados, si bien aún preliminares, posibilitaron constituir un entramado de relaciones donde los participantes desarrollan actividades conjuntas y desjerarquizadas persiguiendo como principal objetivo la valorización y el resguardo del patrimonio arqueológico, así como la construcción del conocimiento del pasado prehispánico local.This article presents the first results obtained in the works carried out together with members of the educational community of the city of Catamarca and Los Castillos, this last one, located in the Ambato Valley. We worked with teachers and students of initial, intermediate and superior levels, with the objective of forming a net of relationships and actions, where the various actors involved contribute their experiences and knowledge in equal proportions, decentralizing the figure of the archaeologist in the research process. The results, although still preliminary, made it possible to constitute a network of relationships where the participants develop a series of activities together and in a non-hierarchical way, pursuing as their main goal the valorization and guard of the archaeological heritage, as well as the construction of the knowledge related to the local prehispanic past.Fil: Figueroa, Germán Gabriel. Glucocorticoid-Induced Leucine Zipper Inhibits the Raf-Extracellular Signal-Regulated Kinase Pathway by Binding to Raf-1

Glucocorticoid-induced leucine zipper (GILZ) is a leucine zipper protein, whose expression is augmented by dexamethasone (DEX) treatment and downregulated by T-cell receptor (TCR) triggering. Stable expression of GILZ in T cells mimics some of the effects of glucocorticoid hormones (GCH) in GCH-mediated immunosuppressive and anti-inflammatory activity. In fact, GILZ overexpression inhibits TCR-activated NF-κB nuclear translocation, interleukin-2 production, FasL upregulation, and the consequent activation-induced apoptosis. We have investigated the molecular mechanism underlying GILZ-mediated regulation of T-cell activation by analyzing the effects of GILZ on the activity of mitogen-activated protein kinase (MAPK) family members, including Raf, MAPK/extracellular signal-regulated kinase (ERK) 1/2 (MEK-1/2), ERK-1/2, and c-Jun NH(2)-terminal protein kinase (JNK). Our results indicate that GILZ inhibited Raf-1 phosphorylation, which resulted in the suppression of both MEK/ERK-1/2 phosphorylation and AP-1-dependent transcription. We demonstrate that GILZ interacts in vitro and in vivo with endogenous Raf-1 and that Raf-1 coimmunoprecipitated with GILZ in murine thymocytes treated with DEX. Mapping of the binding domains and experiments with GILZ mutants showed that GILZ binds the region of Raf interacting with Ras through the NH(2)-terminal region. These data suggest that GILZ contributes, through protein-to-protein interaction with Raf-1 and the consequent inhibition of Raf-MEK-ERK activation, to regulating the MAPK pathway and to providing a further mechanism underlying GCH immunosuppression

Suppression of Myc oncogenic activity by ribosomal protein haploinsufficiency.

The Myc oncogene regulates the expression of several components of the protein synthetic machinery, including ribosomal proteins, initiation factors of translation, RNA polymerase III and ribosomal DNA. Whether and how increasing the cellular protein synthesis capacity affects the multistep process leading to cancer remains to be addressed. Here we use ribosomal protein heterozygote mice as a genetic tool to restore increased protein synthesis in Emu-Myc/+ transgenic mice to normal levels, and show that the oncogenic potential of Myc in this context is suppressed. Our findings demonstrate that the ability of Myc to increase protein synthesis directly augments cell size and is sufficient to accelerate cell cycle progression independently of known cell cycle targets transcriptionally regulated by Myc. In addition, when protein synthesis is restored to normal levels, Myc-overexpressing precancerous cells are more efficiently eliminated by programmed cell death. Our findings reveal a new mechanism that links increases in general protein synthesis rates downstream of an oncogenic signal to a specific molecular impairment in the modality of translation initiation used to regulate the expression of selective messenger RNAs. We show that an aberrant increase in cap-dependent translation downstream of Myc hyperactivation specifically impairs the translational switch to internal ribosomal entry site (IRES)-dependent translation that is required for accurate mitotic progression. Failure of this translational switch results in reduced mitotic-specific expression of the endogenous IRES-dependent form of Cdk11 (also known as Cdc2l and PITSLRE), which leads to cytokinesis defects and is associated with increased centrosome numbers and genome instability in Emu-Myc/+ mice. When accurate translational control is re-established in Emu-Myc/+ mice, genome instability is suppressed. Our findings demonstrate how perturbations in translational control provide a highly specific outcome for gene expression, genome stability and cancer initiation that have important implications for understanding the molecular mechanism of cancer formation at the post-genomic level

Arqueología y comunidad: Valorización y conservación del patrimonio arqueológico del Valle de Ambato, Catamarca, Argentina

La arqueología del Valle de Ambato, Catamarca, Argentina, cuenta con una larga trayectoria, solo detenida durante la última dictadura militar. Los aportes logrados desde ese entonces fueron y son aún profusos, generando, no sólo consensos, sino también profundos debates acerca del modo de vida de las poblaciones Aguada que habitaron la zona entre los siglos VI al XI d.C. Sin embargo, si realizamos un breve repaso sobre la historiografía de las investigaciones, podemos advertir que dentro de las ausencias más destacadas se halla la escasa participación por parte de la población local. Este hecho, originó en muchos casos una profunda desconfianza hacia los arqueólogos y a la disciplina en general, generándose fantasías en torno a los hallazgos realizados y distanciando cada vez más la relación entre ambos actores. Precisamente, en el contexto nacional actual, iniciado hace unas décadas atrás, se fueron dando las condiciones necesarias para propiciar la práctica de una arqueología inclusiva y comprometida socialmente. De este modo, es que iniciamos a fines del año 2017 un proyecto que busca poner en valor y conservar el patrimonio arqueológico de Ambato, el que actualmente se encuentra en serio peligro de destrucción y/o desaparición. Para ello, el trabajo en conjunto con alumnos y docentes de la escuela de Los Castillos, constituye el puntapié inicial de una labor que pretende extenderse en un futuro a otros puntos del valle y a otras franjas etarias de la población. Si bien, el proyecto aún se encuentra en sus primeras etapas, ya está generando resultados, los cuales constituirán el eje este escrito.Fil: Figueroa, Germán Gabriel. EGFR mutations cause a lethal syndrome of epithelial dysfunction with progeroid features

The epidermal growth factor receptor (EGFR) is part of a large family of receptors required for communicating extracellular signals through internal tyrosine kinases. Epidermal growth factor (EGF) signaling is required for tissue development, whereas constitutive activation of this signaling pathway is associated with oncogenic transformation. We identified homozygous c.1283G>A (p.Gly428Asp) mutations in the extracellular domain of EGFR in two siblings. The children were born prematurely, had abnormalities in skin and hair, suffered multisystem organ failure, and died in the neonatal period from intestinal perforation. EGF failed to induce mutated receptor phosphorylation in patient-derived fibroblasts and activation of downstream targets was suppressed. The heterologously expressed extracellular domain was impaired in stability and the binding of EGF. Cells from the affected patient undergo early senescence with accelerated expression of β-galactosidase and shortened telomeres at all passages when compared to controls. A comparison of homozygous inherited regions from a separate report of a patient from the same ethnic background and EGFR genotype confirms the pathogenicity of EGFR mutations in congenital disease
