    In Vivo and In Vitro Characterization of Primary Human Liver Macrophages and Their Inflammatory State

    Liver macrophages (LMs) play a central role in acute and chronic liver pathologies. Investigation of these processes in humans as well as the development of diagnostic tools and new therapeutic strategies require in vitro models that closely resemble the in vivo situation. In our study, we sought to gain further insight into the role of LMs in different liver pathologies and into their characteristics after isolation from liver tissue. For this purpose, LMs were characterized in human liver tissue sections using immunohistochemistry and bioinformatic image analysis. Isolated cells were characterized in suspension using FACS analyses and in culture using immunofluorescence staining and laser scanning microscopy as well as functional assays. The majority of our investigated liver tissues were characterized by anti-inflammatory LMs which showed a homogeneous distribution and increased cell numbers in correlation with chronic liver injuries. In contrast, pro-inflammatory LMs appeared as temporary and locally restricted reactions. Detailed characterization of isolated macrophages revealed a complex disease dependent pattern of LMs consisting of pro- and anti-inflammatory macrophages of different origins, regulatory macrophages and monocytes. Our study showed that in most cases the macrophage pattern can be transferred in adherent cultures. The observed exceptions were restricted to LMs with pro-inflammatory characteristics

    Approaches to the application of magnetic fluids in electromechanical drive systems

    This paper shows the approach of applications of magnetic liquids in electromechanical drive systems. Magnetic fluids consist of colloidal ferromagnetic nanoparticles, a particle surfactant and carrier liquid. These fluids are divided into two groups called ferrofluids and magneto-rheological fluids (MRF). Both liquids are examined in two different kinds of electric motor prototypes. Following the ideas of Nethe [4], a ferrofluid is located in the air gap of an electrical drive. The influence on torque and especially heat transfer is shown by experiments. The system is also studied analytically as a classical Taylor-Couette-System. A second motor prototype is a novel and innovative magnetorheological assisted electrical machine. The construction and the functional principle are presented in this paper. In addition, some of first measurements are shown

In-Memory-basierte betriebliche Informationssysteme bieten viele Vorteile und können bei der Definition neuer SCM-Systeme zugrunde gelegt werden. Allerdings sind noch Defizite in der Datenorganisation und den User Interfaces zu überwinden sowie Geschäftsprozesse anzupassen. Basierend auf den Erfahrungen aus mehreren Forschungsprojekten und den dort praktisch realisierten Demonstratoren werden die Eigenschaften von Real-Time-SCP-Lösungen definiert und anhand des Sales and Operations Planning (SOP)-Prozesses Perspektiven zur Überwindung bestehender Defizite aufgezeigt

    3D Spectral Nonrigid Registration of Facial Expression Scans

    In this paper, we introduce a new template-based spectral nonrigid registration method in which the target is represented using multilevel partition of unity (MPU) implicit surfaces and the template is embedded in a discrete Laplace-Beltrami based spectral representation using the manifold harmonics transform (MHT). The implicit surface parametrization of the target allows us to avoid computing correspondences during the registration as in classical nonrigid iterative closest point (ICP) techniques. It also allows us to denoise the 3D scans and fill the holes by interpolating the noisy 3D data and to incorporate different types of 3D surfaces into our model, independently of their original parametrization. We take advantage of spectral geometry processing methods to compute a spectral embedding of the template and use it as a parametric surface deformation model. We optimize the nonrigid deformation directly in the spectral domain, thus effectively reducing the size of the parameter space as compared with the classical per vertex affine transformation deformation model. In addition, we introduce a new 3D facial expressions database, EPFL3DFace, on which we apply the proposed method to nonrigidly register 3D face scans that contain different expressions. This database consists of 3D scans of 120 subjects posing 35 different facial expressions. These include various standard prototypical facial expressions as well as individual action units, visemes, and the facial movement of biting one’s own upper lip, which are suitable for a large variety of applications

Целью работы было: изучить рынок автострахования России на примере компании "Коместра страхование".Основными задачами было:изучить сущность и структуру страхового рынка,дать характеристику участников страхового рынка,проанализировать состояние российского страхового рынка в части его динамики, масштабов, уровня оказываемых страховых услуг. Данная тема актуальна, так как множество различных компаний открываются и стараются конкурировать с уже устоявшимися компаниями. В данной работе предложен ряд решений которые могут увеличить уровень конкурентоспособности компании в целом.The object of research in this work is the car insurance market of Russia. The aim of the work was: to study the Russian auto insurance market using the example of the company "Comestra Insurance". The main tasks were: to study the nature and structure of the insurance market, to characterize the participants of the insurance market, to analyze the state of the Russian insurance market in terms of its dynamics, scale, level of insurance services provided. This topic is relevant, as many different companies open up and try to compete with already established companies. In this work, a number of solutions are proposed that can increase the level of competitiveness of the company as a whole

    Photonic and Optomechanical Thermometry

    Temperature is one of the most relevant physical quantities that affects almost all processes in nature. However, the realization of accurate temperature standards using current temperature references, like the triple point of water, is difficult due to the requirements on material purity and stability of the environment. In addition, in harsh environments, current temperature sensors with electrical readout, like platinum resistors, are difficult to implement, urging the development of optical temperature sensors. In 2018, the European consortium Photoquant, consisting of metrological institutes and academic partners, started investigating new temperature standards for self-calibrated, embedded optomechanical sensor applications, as well as optimised high resolution and high re- liability photonic sensors, to measure temperature at the nano and meso-scales and as a possible replacement for the standard platinum resistant thermometers. This article presents an overview of the results obtained with sensor prototypes that exploit photonic and optomechanical techniques for sensing temperatures over a large temperature range (5 K to 300 K). Different concepts are demon- strated, including ring resonators, ladder-like resonators and suspended membrane optomechanical thermometers, highlighting initial performance and challenges, like self-heating that need to be overcome to realize photonic and optomechanical thermometry applications.This work was carried out under the 17FUN05 PhotOQuanT project, which has received funding from the EMPIR program, co-financed by the Participating States and the European Union’s Horizon 2020 research and innovation progra

    The proteoglycan osteoglycin/mimecan is correlated with arteriogenesis

    Arteriogenesis or collateral growth is able to compensate for the stenosis of major arteries. Using differential display RT-PCR on growing and quiescent collateral arteries in a rabbit femoral artery ligation model, we cloned the rabbit full-length cDNA of osteoglycin/mimecan. Osteoglycin was present in the adventitia of collateral arteries as a glycosylated protein without keratan sulfate side chains, mainly produced by smooth muscle cells (SMCs) and perivascular fibroblasts. Northern blot, Western blot, and immunohistochemistry confirmed a collateral artery-specific downregulation of osteoglycin from 6 h to 3 weeks after the onset of arteriogenesis. Treatment of primary SMCs with the arteriogenic protein fibroblast growth factor-2 (FGF-2) resulted in a similar reduction of osteoglycin expression as observed in vivo. Application of the FGF-2 inhibitor polyanethole sulfonic acid (PAS) blocked the downregulation of osteoglycin and interfered with arteriogenesis. From our study we conclude that downregulation of osteoglycin is a fundamental requirement for proper arteriogenesis
