    A multinode quantum network over a metropolitan area

    Towards realizing the future quantum internet, a pivotal milestone entails the transition from two-node proof-of-principle experiments conducted in laboratories to comprehensive, multi-node setups on large scales. Here, we report on the debut implementation of a multi-node entanglement-based quantum network over a metropolitan area. We equipped three quantum nodes with atomic quantum memories and their telecom interfaces, and combined them into a scalable phase-stabilized architecture through a server node. We demonstrated heralded entanglement generation between two quantum nodes situated 12.5 km apart, and the storage of entanglement exceeding the round-trip communication time. We also showed the concurrent entanglement generation on three links. Our work provides a metropolitan-scale testbed for the evaluation and exploration of multi-node quantum network protocols and starts a new stage of quantum internet research.Comment: 21 pages in total, 4 figures and 1 table in the main text, 5 figures and 8 tables in the supplementary materia

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    The R2R3-MYB transcription factor FveMYB10 is a major regulator of anthocyanin pigmentation in the red fruits of strawberry. fvemyb10 loss-of-function mutants form yellow fruits but still accumulate purple-colored anthocyanins in the petioles, suggesting that anthocyanin biosynthesis is under distinct regulation in fruits and petioles. From chemical mutagenesis in the diploid wild strawberry Fragaria vesca, we identified a green petioles (gp)-1 mutant that lacks anthocyanins in petioles. Using mapping-by-sequencing and transient functional assays, we confirmed that the causative mutation resides in a FveMYB10-Like (FveMYB10L) gene and that FveMYB10 and FveMYB10L function independently in the fruit and petiole, respectively. In addition to their tissue-specific regulation, FveMYB10 and FveMYB10L respond differently to changes in light quality, produce distinct anthocyanin compositions, and preferentially activate different downstream anthocyanin biosynthesis genes in their respective tissues. This work identifies a new regulator of anthocyanin synthesis and demonstrates that two paralogous MYB genes with specialized functions enable tissue-specific regulation of anthocyanin biosynthesis in fruit and petiole tissues.Strawberry plants accumulate anthocyanin in petioles through the action of the transcription factor MYB10-Like.     Silver Nanocluster-Embedded Zein Films as Antimicrobial Coating Materials for Food Packaging

    Highly efficient antimicrobial agents with low toxicity and resistance have been enthusiastically pursued to address public concerns on microbial contamination in food. Silver nanoclusters (AgNCs) are known for their ultrasmall sizes and unique optical and chemical properties. Despite extensive studies of AgNCs for biomedical applications, previous research on their application as antimicrobials for food applications is very limited. Here, for the first time, by incorporating AgNCs (~2 nm in diameter) into zein films that are widely used as food packaging materials, we developed a novel coating material with potent antimicrobial activity, low toxicity to human cells, and low potential to harm the environment. In addition, we systematically evaluated the antimicrobial activities and cytotoxicity of AgNCs-embedded zein films and compared them to zein films embedded with AgNO3 or Ag nanoparticles with diameters of 10 and 60 nm (AgNP10 and AgNP60, respectively). At equivalent silver concentrations, AgNCs and AgNO3 solutions exhibited considerably higher antimicrobial activities than those of AgNP10 and AgNP60 solutions. Moreover, AgNCs exhibited less cytotoxicity to human cells than AgNO3, with a half maximal inhibitory concentration (IC50) of 34.68 ÎĽg/mL for AgNCs, compared to 9.14 ÎĽg/mL for AgNO3. Overall, the novel AgNCs coating developed in this research has great potential for antimicrobial applications in food packaging materials due to its high antimicrobial efficacy, ultrasmall size, and low cytotoxicity

    Improved metabolomic approach for evaluation of phytochemicals in mustard, kale, and broccoli microgreens under different controlled environment agriculture conditions

    The fast-growing field of controlled environment agriculture (CEA) offers unprecedented opportunities for targeted improvement in concentrations of bioactive compounds in fresh produce achieved through precise modulation of production conditions. To gain full sight of the phytochemical profiles of vegetables grown under different conditions, a rapid analytical strategy is needed for the evaluation of different CEA growing conditions. In this study, Brassica microgreens including ruby streaks mustard (B. juncea), red kale (B. oleracea), and broccoli (B. oleracea) were used as model plants for the evaluation of CEA conditions. Analysis of two first leaves (cotyledons) in microgreens with minimum sample extraction and ultra-high performance liquid chromatography-high-resolution mass spectrometry (UHPLC-HRMS) based metabolomic approach was applied for phytochemical analysis for evaluation of the brassica microgreens grown under four different light sources, namely white (W), dark (D), white and far-red (WF) and far-red (F) and fertilizer conditions (CaCl2 and K2SO4). An image-based normalization method using leaf area coupled with chemometrics-based strategies including principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA) was performed for the post-acquisition data analysis. The method successfully distinguished between Brassica microgreens grown under different CEA settings in a shortened cycle with less organic solvent and labor, which is more environmentally friendly and sustainable. Marker compounds that are responsible for differentiating the Brassica microgreens under various CEA conditions were tentatively identified. Among the tentatively identified marker compounds, gingerglycolipid A was first reported in red kale and broccoli. The results from the present study may serve as a scientific foundation for the rapid and simple assessment to optimize CEA conditions

    Organic materials in fresh-cut produce wash water deplete free chlorine that is required to prevent pathogen survival and cross-contamination. This research evaluated water quality parameters frequently used to describe organic load for their fitness to predict chlorine demand (CLD) and chemical oxygen demand (COD), which are major needs identified by the industry-led produce food safety taskforce. Batches of romaine lettuce, iceberg lettuce, or carrot of different cut sizes and shapes were washed in 40 liters of water. Physicochemical properties of wash water including CLD, COD, total organic carbon (TOC), total suspended solids (TSS), total dissolved solids (TDS), turbidity, total sugar content, and pH, were monitored. Results indicate that pH is primarily commodity dependent, while organic load is additionally impacted by cutting and washing conditions. Significant linear increases in COD, TOC, CLD, TDS, and turbidity resulted from increasing product-to-water ratio, and decreasing cut size. Physicochemical parameters, excluding pH, showed significant positive correlation across different cut sizes within a commodity. High correlations were obtained between CLD and COD and between COD and TOC for pooled products. The convenient measurement of TDS, along with its strong correlation with COD and CLD, suggests the potential of TDS for predicting organic load and chlorine reactivity. Finally, the potential application and limitation of the proposed models in practical produce processing procedures are discussed extensively

    Cationic β‑Lactoglobulin Nanoparticles as a Bioavailability Enhancer: Protein Characterization and Particle Formation

    Cationic β-lactoglobulin (CBLG) was developed as a bioavailability enhancer for poorly absorbed bioactives. At most 11 anionic amino acid residues of β-lactoglobulin (BLG) were substituted by ethylenediamine (EDA), resulting in a highly positive surface charge (zeta potential up to 39 mV at pH 7.0) and significantly increased surface hydrophobicity. These changes conferred CBLG with desirable water solubility and improved mucoadhesion by at most 252%, according to quartz crystal microbalance (QCM) study. Furthermore, CBLG inherited the unique resistance to gastric digestion from BLG, while the digestion under simulated intestinal condition was significantly improved. The latter was possibly due to the formation of aspartic acid-EDA conjugates, together with the randomization of protein conformation related with decreased percentage of β-sheet. Compared to BLG, CBLG formed smaller (75–94 nm), more uniform nanoparticles by the acetone-desolvation method. These merits made CBLG a useful material that provides desirable solubility, controlled release, and enhanced absorption to nutraceuticals or drugs

    Inhibition of α5 GABAA receptors has preventive but not therapeutic effects on isoflurane-induced memory impairment in aged rats

    The α5 subunit-containing gamma-amino butyric acid type A receptors (α5 GABAARs) are a distinct subpopulation that are specifically distributed in the mammalian hippocampus and also mediate tonic inhibitory currents in hippocampal neurons. These tonic currents can be enhanced by low-dose isoflurane, which is associated with learning and memory impairment. Inverse agonists of α5 GABAARs, such as L-655,708, are able to reverse the short-term memory deficit caused by low-dose isoflurane in young animals. However, whether these negative allosteric modulators have the same effects on aged rats remains unclear. In the present study, we mainly investigated the effects of L-655,708 on low-dose (1.3%) isoflurane-induced learning and memory impairment in elderly rats. Young (3-month-old) and aged (24-month-old) Wistar rats were randomly assigned to receive L-655,708 0.5 hour before or 23.5 hours after 1.3% isoflurane anesthesia. The Morris Water Maze tests demonstrated that L-655,708 injected before or after anesthesia could reverse the memory deficit in young rats. But in aged rats, application of L-655,708 only before anesthesia showed similar effects. Reverse transcription-polymerase chain reaction showed that low-dose isoflurane decreased the mRNA expression of α5 GABAARs in aging hippocampal neurons but increased that in young animals. These findings indicate that L-655,708 prevented but could not reverse 1.3% isoflurane-induced spatial learning and memory impairment in aged Wistar rats.     Impacts and interactions of organic compounds with chlorine sanitizer in recirculated and reused produce processing water.

    Water conservation and economics dictate that fresh produce processors reuse/recirculate the process water. However, the ensuing accumulation of organic matter in water depletes the chlorine sanitizer required for food safety. In this study, we comprehensively investigated chemical compounds that are responsible for water quality in relation to chemical oxygen demand (COD) and chlorine demand (CLD), the two most critical factors associated with water treatment and chlorine replenishment. Simulating commercial fresh-cut wash operations, multiple batches of diced cabbage (0.3 x 0.3 cm2) were washed in the same tank of water. The major components were isolated from the wash water and analyzed by HPLC. Sugars were the predominant compounds (82.7% dry weight) and the major contributor to COD (81.6%), followed by proteins/peptides (7.3% dry weight, 5.3% COD), organic acids (6.2% dry weight, 3.6% COD), and phenolics (0.5% dry weight, 0.5% COD). By repeated time course measures, the effect of these chemicals on CLD are dependent on the chemical structure, concentration in the wash water, and their rate of reaction. Proteins/peptides accounted for about 50% of the total CLD over a 120-min period and phenolics was 21% at 5 min, but diminished with time. The contribution by organic acids and sugars increased continuously, reaching 22% and 16% of total CLD at 120 min of chlorination, respectively. Collectively, these compounds represented 86% of the CLD in cabbage wash water at 5 min and greater than 94% CLD afterwards. This is the first systematic report on the source of COD and CLD during fresh produce washing. It provides essential information for the produce processors to develop safe, effective, and economical wash water treatment/reuse and chlorine replenishment strategies
