    Phylogeography of a widespread terrestrial vertebrate in a barely-studied Palearctic region: green toads (Bufo viridis subgroup) indicate glacial refugia in Eastern Central Asia.

    The phylogeography of western Palearctic species is relatively well studied, but data on Eastern Central Asia are scarce. We present one of the first data sets from a widespread terrestrial vertebrate (Bufo pewzowi) inhabiting Eastern Central Asian mountains and deserts to gain knowledge on its phylogeography in this region. We applied combined phylogenetic and demographic analyses to understand the evolutionary history using mitochondrial DNA D-loop variation of toads from 37 locations. Genetic structure of Bufo pewzowi is strongly affected by landscape: we found three haplotype groups in eastern Kazakhstan, Dzungaria and Tarim Basin, divided by the Tian Shan and Dzungarian Alatau ranges. A vicariant hypothesis may explain divergence among groups. The divergence time of the three major clades was estimated about 0.9 million years ago (confidence interval 0.5-1.4), and is discussed with respect to Quaternary uplifting and glaciation in the Tian Shan. Demographic analyses provided evidence for both historical bottlenecks and population expansions and suggested Pleistocene signatures. Glacial refugia were inferred in the Tarim Basin (around the Turpan depression), in southern Dzungaria (Urumqui region), at the northern foot of the Tian Shan (Gongnaisi) and perhaps at the Altai range (Terekti). Regional Post-Last Glacial Maximum dispersal patterns are proposed. A taxonomic hypothesis is presented. This study provides a detailed history of how a widespread terrestrial vertebrate responded to geological change and Quaternary glacial events in Eastern Central Asia and may have significance for future phylogeographic research in this understudied region

    Novel species of Calonectria associated with Eucalyptus leaf blight in Southeast China

    Leaf blight caused by Calonectria spp. is an important disease occurring on Eucalyptus trees grown in plantations of Southeast Asia. Symptoms of leaf blight caused by Calonectria spp. have recently been observed in commercial Eucalyptus plantations in FuJian Province in Southeast China. The aim of this study was to identify these Calonectria spp. employing morphological characteristics, DNA sequence comparisons for the β-tubulin, histone H3 and translation elongation factor-1α gene regions and sexual compatibility. Four Calonectria spp. were identified, including Ca. pauciramosa and three novel taxa described here as Ca. crousiana, Ca. fujianensis and Ca. pseudocolhounii. Inoculation tests showed that all four Calonectria spp. found in this study were pathogenic on two different E. urophylla × E. grandis hybrid clones, commercially utilised in eucalypt plantations in China

    Evolution of Anisotropy in Granular Materials: Effect of Particle Rolling and Particle Crushing

    The effect of particle rolling and crushing on the evolutions of the two types of anisotropy, i.e., anisotropy of particle packing (microstructure) and anisotropy of force chains, is investigated numerically using the discrete element method. To this end, the classical fabric tensor is adopted to describe the anisotropy of microstructure, while two similar orientation tensors defined by the directions of contact forces are used to characterize the anisotropy of force chains. Numerical results show that the evolutions of anisotropy follows the same tendency as the stress–strain curve, and the anisotropy of force chains is more intense than that of the microstructure. In addition, particle rolling exerts different effect on anisotropy before and after the peak stress state, and particle crushing decreases the anisotropy of granular materials.Представлено чисельне дослідження за допомогою методу дискретних елементів впливу скочування і дроблення частинок на еволюцію анізотропій скочування частинок (мікроструктура) і силового ланцюжка. Для опису анізотропії мікроструктури використовується структурний класичний тензор, а два аналогічних тензора орієнтації, що характеризуються напрямком контактних зусиль, – для визначення анізотропії силового ланцюжка. Результати чисельного дослідження показали, що еволюція анізотропій має той же характер, що і залежність деформації від напруження, однак анізотропія силового ланцюжка є більш інтенсивною порівняно з анізотропією мікроструктури. Більш того, скочування частинок по-різному впливає на анізотропію до і після досягнення максимального значення напруження, в той час як дроблення частинок зменшує анізотропію гранульованих матеріалів.Представлено численное исследование с помощью метода дискретных элементов влияния скатывания и дробления частиц на эволюцию анизотропий скатывания частиц (микроструктура) и силовой цепочки. Для описания анизотропии микроструктуры используется структурный классический тензор, а два аналогичных тензора ориентации, характеризующихся направлением контактных усилий, – для определения анизотропии силовой цепочки. Результаты численного исследования показали, что эволюция анизотропий имеет тот же характер, что и зависимость деформации от напряжения, однако анизотропия силовой цепочки является более интенсивной по сравнению с анизотропией микроструктуры. Более того, скатывание частиц по-разному влияет на анизотропию до и после достижения максимального значения напряжения, тогда как дробление частиц уменьшает анизотропию гранулированных материалов

    High-pressure behaviors of carbon nanotubes

    In this paper, we have reviewed the experimental and theoretical studies on pressure-induced polygonization, ovalization, racetrack–shape deformation, and polymerization of carbon nanotubes (CNTs). The corresponding electronic, optical, and mechanical changes accompanying these behaviors have been discussed. The transformations of armchair (n, n) CNT bundles (n = 2, 3, 4, 6, and 8) under hydrostatic or nonhydrostatic pressure into new carbons, including recently proposed superhard bct-C₄, Cco-C₈, and B-B1AL2R2 carbon phases have also been demonstrated. Given the diversity of CNTs from various chiralities, diameters, and arrangements, pressure-induced CNT polymerization provides a promising approach to produce numerous novel metastable carbons exhibiting unique electronic, optical, and mechanical characteristics.Розглянуто експериментальні та теоретичні дослідження з індукованою тиском полігонізації, овалізації, деформації у формі бігової доріжки і полімеризації вуглецевих нанотрубок (ВНТ). Обговорено відповідні електронні, оптичні і механічні зміни, що супроводжують ці процеси. Також продемонстровано перетворення в ВНТ у формі крісла (n, n), зібраних в пучок (n = 2, 3, 4, 6 і 8) під гідростатичним або негідростатичним тиском в нові вуглецеві алотропи, в тому числі недавно запропоновані надтверді bct-C₄, Cco-C₈ і B-B1AL2R2-вуглецеві фази. Різноманітність ВНТ з різними хіральністю, діаметрами та упаковками, а також полімеризація ВНТ, викликана тиском, забезпечує перспективний підхід для отримання численних нових метастабільних вуглецевих фаз, що демонструють унікальні електронні, оптичні і механічні характеристики.Рассмотрены экспериментальные и теоретические исследования по индуцированной давлением полигонизации, овализации, деформации в форме беговой дорожки и полимеризации углеродных нанотрубок (УНТ). Обсуждены соответствующие электронные, оптические и механические изменения, сопровождающие эти процессы. Также продемонстрированы преобразования в УНТ в форме кресла (n, n), собранных в пучок (n = 2, 3, 4, 6 и 8) под гидростатическим или негидростатическим давлением в новые углеродные аллотропы, в том числе недавно предложенные сверхтвердые bct-C₄, Cco-C₈ и B-B1AL2R2-углеродные фазы. Разнообразие УНТ с различными хиральностью, диаметрами и упаковками, а также полимеризация УНТ, вызванная давлением, обеспечивает перспективный подход для получения многочисленных новых метастабильных углеродных фаз, демонстрирующих уникальные электронные, оптические и механические характеристики

    Comparison of Structural Development and Biochemical Accumulation of Waxy and Non-waxy Wheat Caryopses

    This study was conducted to compare structural development and biochemical accumulation of waxy and non-waxy wheat (NW) caryopses. The caryopses’ microstructure of the waxy wheat (WW) and NW cultivars at different developmental stages were observed under light, fluorescence, and scanning electron microscope. The results were as follows: Compared with NW,WWhad a shorter maturation duration, which was reflected in several following characteristics. Programmed cell death of the pericarp began earlier, and the chlorophyll-containing layer in the pericarp was smaller. Vacuoles in chalazal cells accumulated more tannins at different developmental stages. Starch granules and protein bodies in the endosperm showed a higher accumulation level in developing caryopses, and aleurone cells were larger in size with larger numbers of aleurone grains. An analysis of the element content indicated that the mineral elements Mg, P, K, and Ca exhibited a higher content, while the heavy elements Cr, Cd, and Pb exhibited a lower content in the aleurone layer

    Nonsymbiotic hemoglobins (ns-Hbs) previously have been found in monocots and dicots; however, very little is known about the tissue and cell type localization as well as the physiological function(s) of these oxygen-binding proteins. We report the immunodetection and immunolocalization of ns-Hbs in rice (Oryza sativa L.) by Western blotting and in situ confocal laser scanning techniques. Ns-Hbs were detected in soluble extracts of different tissues from the developing rice seedling by immunoblotting. Levels of ns-Hbs increased in the germinating seed for the first six days following imbibition and remained relatively constant thereafter. In contrast, ns-Hb levels decreased during leaf maturation. Roots and mesocotyls contained detectable, but low levels of ns-Hbs. Split-seed experiments revealed that ns-Hbs are synthesized de novo during seed germination and are expressed in the absence of any signal originating from the embryo. Immunolocalization of ns-Hbs by con- focal microscopy indicated the presence of ns-Hbs primarily in differentiated and differentiating cell types of the developing seedling, such as the aleurone, scutellum, root cap cells, sclerenchyma, and tracheary elements. To our knowledge, this is the first report of the specific cellular localization of these proteins during seedling development

    Boty-II, a novel LTR retrotransposon in Botrytis cinerea B05.10 revealed by genomic sequence

    Botrytis cinerea is a necrotrophic pathogen causing pre- and post-harvest diseases in at least 235 plant species. It manifests extraordinary genotype and phenotype variation. One of the causes of this variation is transposable elements. Two transposable elements have been discovered in this fungus, the retrotransposon (Boty), and the transposon (Flipper). In this work, two complete (Boty-II-76 and Boty-II-103) and two partial (Boty-II-95 and Boty-II-141) long terminal repeat (LTR) retrotransposons were identified by an in silico genomic sequence analysis. Boty-II-76 and Boty-II-103 contain 6439 bp nucleotides with a pair of LTRs at both ends, and an internal deduced pol gene encoding a polyprotein with reverse transcriptase and DDE integrase domains. They are flanked by 5 bp direct repeats (ACCAT, CTTTC). In Boty-II-141, two LTRs at both ends, and a partial internal pol gene encoding a protein with a DDE integrase domain were identified. In Boty-II-95, a right LTR and a partial internal pol gene encoding a protein with no conserved domains were identified. Boty-II uses a self-priming mechanism to initiate synthesis of reverse transcripts. The sequence of the presumed primer binding site for first-strand reverse transcription is 5'-TTGTACCAT-3'. The polypurine-rich sequence for plus-strand DNA synthesis is 5'-GCCTTGAGCGGGGGGTAC-3'. Fourteen Boty-II LTRs that contain 125-158 bp nucleotides and share 69.1 ~ 100% identities with the short inverted terminal repeats of 5 bp (TGTCA\u2026TGACA) were discovered. Analysis of structural features and phylogeny revealed that Boty-II is a novel LTR retrotransposon. It could potentially be used as a novel molecular marker for the investigation of genetic variation in B. cinerea

    Knockout of p75 neurotrophin receptor attenuates the hyperphosphorylation of Tau in pR5 mouse model

    p75 neurotrophin receptor (p75NTR) has been implicated in Alzheimer's disease (AD). However, whether p75NTR is involved in Tau hyperphosphorylation, one of the pathologies observed in AD, remains unclear. In our previous study, the extracellular domain of p75NTR blocked amyloid beta (Aβ) toxicity and attenuated Aβ-induced Tau hyperphosphorylation. Here we show that, in the absence of Aβ, p75NTR regulates Tau phosphorylation in the transgenic mice with the P301L human Tau mutation (pR5). The knockout of p75NTR in pR5 mice attenuated the phosphorylation of human Tau. In addition, the elevated activity of kinases responsible for Tau phosphorylation including glycogen synthase kinase 3 beta; cyclin-dependent-kinase 5; and Rho-associated protein kinase was also inhibited when p75NTR is knocked out in pR5 mice at 9 months of age. The increased caspase-3 activity observed in pR5 mice was also abolished in the absence of p75NTR. Our study also showed that p75NTR is required for Aβ- and pro-brain derived neurotrophin factor (proBDNF)-induced Tau phosphorylation, in vitro. Overall, our data indicate that p75NTR is required for Tau phosphorylation, a key event in the formation of neurofibrillary tangles, another hallmark of AD.     Measurement of Trace I-129 Concentrations in CsI Powder and Organic Liquid Scintillator with Accelerator Mass Spectrometry

    Levels of trace radiopurity in active detector materials is a subject of major concern in low-background experiments. Procedures were devised to measure trace concentrations of I-129 in the inorganic salt CsI as well as in organic liquid scintillator with Accelerator Mass Spectrometry (AMS) which leads to improvement in sensitivities by several orders of magnitude over other methods. No evidence of their existence in these materials were observed. Limits of < 6 X 10^{-13} g/g and < 2.6 X 10^{-17} g/g on the contaminations of I-129 in CsI and liquid scintillator, respectively, were derived.These are the first results in a research program whose goals are to develop techniques to measure trace radioactivity in detector materials by AMS.Comment: Proceedings of 10th International Conference on Accelerator Mass Spectrometr
