    Investigation and Analysis of Present Situation and Educational Countermeasures and Cultural Needs of Farmers

    The samples, from 21 provinces, municipalities directly under the central government, 107 villages, 30 towns, are altogether 3,577. The survey found, the cultural demand of farmers is generally healthy and positive. What they need most are cultural knowledge of computer technology, disease prevention knowledge, legal knowledge to deal with disputes, migrant workers rights of women and children protection law, recreational activities are singing and dancing, favorite sports activities the first three are the small ball (table tennis badminton), major sports (basketball and volleyball football) and the short or long runs. And there are some differences in age, gender, and cultural degree of life state between these demands. Investigation and analysis reflect that, some activities already undertaken can’t meet the need, and some farmers on the surface needn’t culture, but actually reflect the lag of cultural spiritual pursuit. By contrast with sports, entertainment is more short of physical activity. Farmers’ favorite access to cultural knowledge is face-to-face teaching.     HOW DO ONLINE BOOKSTORES INFLUENCE THE SALES OF TRADITIONAL BOOKSTORES IN CHINA?

    Bookstores have been strongly affected by the rise of electronic commerce. Traditional (physical) bookstores now have to compete with online counterparts that possess several advantages over them, such as lower prices and 24/7 availability. This paper investigates the influence that online bookstores have on the sales of physical bookstores in China. The geographic restriction is based on the particularities of the Chinese market that differentiate China from other markets, both in the book market as well as in the adoption of E-commerce. Since there is a lack of overview in the English academic literature on this topic, we conducted a literature review to summarize the current state of research. Our research highlights multiple advantages and disadvantages of physical and online bookstores in the Chinese market, which in turn influence the sales of traditional bookstores. This paper offers a theoretical and a practical contribution. Participants of the Chinese book market can use the results to better assess and improve their market position by addressing the disadvantages or building on the strengths identified during the review. Furthermore, this paper can serve as a starting point for English researchers, as almost no research on this topic has been published in English

    Polarized growth of pollen tubes is a critical step for successful reproduction in angiosperms and is controlled by ROP GTPases. Spatiotemporal activation of ROP (Rho GTPases of plants) necessitates a complex and sophisticated regulatory system, in which guanine nucleotide exchange factors (RopGEFs) are key components. It was previously shown that a leucine-rich repeat receptor-like kinase, Arabidopsis pollen receptor kinase 2 (AtPRK2), interacted with RopGEF12 for its membrane recruitment. However, the mechanisms underlying AtPRK2-mediated ROP activation in vivo are yet to be defined. It is reported here that over-expression of AtPRK2 induced tube bulging that was accompanied by the ectopic localization of ROP-GTP and the ectopic distribution of actin microfilaments. Tube depolarization was also induced by a potentially kinase-dead mutant, AtPRK2K366R, suggesting that the over-expression effect of AtPRK2 did not require its kinase activity. By contrast, deletions of non-catalytic domains in AtPRK2, i.e. the juxtamembrane (JM) and carboxy-terminal (CT) domains, abolished its ability to affect tube polarization. Notably, AtPRK2K366R retained the ability to interact with RopGEF12, whereas AtPRK2 truncations of these non-catalytic domains did not. Lastly, it has been shown that the JM and CT domains of AtPRK2 were not only critical for its interaction with RopGEF12 but also critical for its distribution at the plasma membrane. These results thus provide further insight into pollen receptor kinase-mediated ROP activation during pollen tube growth

    Seed Oil of Brucea javanica Induces Apoptotic Death of Acute Myeloid Leukemia Cells via Both the Death Receptors and the Mitochondrial-Related Pathways

    Seed oil of Brucea javanica (BJO) is extracted from the seeds of herb medicine Brucea javanica (L.), and its emulsion formulation (BJOE) has been used clinically to treat carcinomas for many years in China. The antileukemia potential of BJO was investigated in human acute myeloid leukemia cell lines (AML) U937 and HL-60 in vitro and in a mouse U937 xenograft tumor model. BJO induced AML cell apoptosis through activation of caspase-8 and modulation of apoptosis-related proteins. Meanwhile, the inhibition of survivin and XIAP increased the cytotoxicity of BJO. Consistent with these findings, BJO also increased subG1 phase cells and cause PARP cleavage in AML patients' leukemia cells. In contrast, only weak cytotoxicity of BJO was found in peripheral blood lymphocytes (PBLs) of healthy volunteers. Moreover, oleic acid and linoleic acid were found to be the active components of BJO. Our study provided strong evidence for the first time that BJO induced apoptosis of both cultured and primary AML cells. Furthermore, intravenous injection of BJO significantly inhibited U937 tumor growth in the xenograft mouse model. These results suggest that BJO may have a therapeutic role in the treatment of human leukemia

    A novel coordination polymer, [Co3(OABDC)2(bipy)3 (H2O)6]n(CH3OH)n(H2O)4.5n (1), has been synthesised by hydrothermal method with the coordination of 4,4′-bipy ligand and 3,5-dicarboxyphenoxyacetic acid (H3OABDC) to cobalt(II) ions. The treatment and nursing application values of 1 on hypothermia in patients undergoing surgery has been examined, and the specific mechanism is explored. The application of the compound on the body temperature is measured with an in vitro infrared thermometer. Then, the relative expression of vascular α-receptors is determined by real-time RT-PCR. The toxicity and biocompatibility of the new compound is also determined

    Study of neutron density fluctuation and neutron-proton correlation in Au+Au collisions using PYTHIA8/Angantyr

    Utilizing the PYTHIA8 Angantyr model, which incorporates the multiple-parton interactions (MPI) based color reconnection (CR) mechanism, we study the relative neutron density fluctuation and neutron-proton correlation in Au+Au collisions at sNN\sqrt{s_\text{NN}} = 7.7, 11.5, 14.5, 19.6, 27, 39, 62.4, and 200 GeV. In this study, we have not only delved into the dependence of these two remarkable observations on rapidity, centrality, and energy, but also presented an analysis of their interplay with the MPI and CR. Our results have shown that the light nuclei yield ratio of proton, deuteron, and triton, expressed by the elegant expression NtNp/Nd2N_tN_p/N_d^2, remains unchanged even as the rapidity coverage and collision centrality increase. Interestingly, we have also revealed that the effect of CR is entirely dependent on the presence of MPI; CR has no impact on the yield ratio if MPI is off. Our findings further demonstrate that the light nuclei yield ratio experiences a slight increase with increasing collision energy as predicted by the PYTHIA8 Angantyr model, but it cannot describe the non-monotonic trend observed by the STAR experiment. Based on the Angantyr model simulation results, it is essential not to overlook the correlation between neutron and proton fluctuations. The Angantyr model is a good baseline for studying collisions in the absence of a Quark-Gluon Plasma (QGP) system, given its lack of flow and jet quenching.Comment: arXiv admin note: text overlap with arXiv:2211.03297 by other author

    Non-Sequential Graph Script Induction via Multimedia Grounding

    Online resources such as WikiHow compile a wide range of scripts for performing everyday tasks, which can assist models in learning to reason about procedures. However, the scripts are always presented in a linear manner, which does not reflect the flexibility displayed by people executing tasks in real life. For example, in the CrossTask Dataset, 64.5% of consecutive step pairs are also observed in the reverse order, suggesting their ordering is not fixed. In addition, each step has an average of 2.56 frequent next steps, demonstrating "branching". In this paper, we propose the new challenging task of non-sequential graph script induction, aiming to capture optional and interchangeable steps in procedural planning. To automate the induction of such graph scripts for given tasks, we propose to take advantage of loosely aligned videos of people performing the tasks. In particular, we design a multimodal framework to ground procedural videos to WikiHow textual steps and thus transform each video into an observed step path on the latent ground truth graph script. This key transformation enables us to train a script knowledge model capable of both generating explicit graph scripts for learnt tasks and predicting future steps given a partial step sequence. Our best model outperforms the strongest pure text/vision baselines by 17.52% absolute gains on F1@3 for next step prediction and 13.8% absolute gains on Acc@1 for partial sequence completion. Human evaluation shows our model outperforming the WikiHow linear baseline by 48.76% absolute gains in capturing sequential and non-sequential step relationships

    Factors associated with suicidal attempts in female patients with mood disorder

    AimThis study aims to establish a nomogram model to predict the relevance of SA in Chinese female patients with mood disorder (MD).MethodThe study included 396 female participants who were diagnosed with MD Diagnostic Group (F30–F39) according to the 10th Edition of Disease and Related Health Problems (ICD-10). Assessing the differences of demographic information and clinical characteristics between the two groups. LASSO Logistic Regression Analyses was used to identify the risk factors of SA. A nomogram was further used to construct a prediction model. Bootstrap re-sampling was used to internally validate the final model. The Receiver Operating Characteristic (ROC) curve and C-index was also used to evaluate the accuracy of the prediction model.ResultLASSO regression analysis showed that five factors led to the occurrence of suicidality, including BMI (β = −0.02, SE = 0.02), social dysfunction (β = 1.72, SE = 0.24), time interval between first onset and first dose (β = 0.03, SE = 0.01), polarity at onset (β = −1.13, SE = 0.25), and times of hospitalization (β = −0.11, SE = 0.06). We assessed the ability of the nomogram model to recognize suicidality, with good results (AUC = 0.76, 95% CI: 0.71–0.80). Indicating that the nomogram had a good consistency (C-index: 0.756, 95% CI: 0.750–0.758). The C-index of bootstrap resampling with 100 replicates for internal validation was 0.740, which further demonstrated the excellent calibration of predicted and observed risks.ConclusionFive factors, namely BMI, social dysfunction, time interval between first onset and first dose, polarity at onset, and times of hospitalization, were found to be significantly associated with the development of suicidality in patients with MD. By incorporating these factors into a nomogram model, we can accurately predict the risk of suicide in MD patients. It is crucial to closely monitor clinical factors from the beginning and throughout the course of MD in order to prevent suicide attempts

    Comparative genomic analyses of Mycoplasma hyopneumoniae pathogenic 168 strain and its high-passaged attenuated strain

    Background: Mycoplasma hyopneumoniae is the causative agent of porcine enzootic pneumonia (EP), a mild, chronic pneumonia of swine. Despite presenting with low direct mortality, EP is responsible for major economic losses in the pig industry. To identify the virulence-associated determinants of M. hyopneumoniae, we determined the whole genome sequence of M. hyopneumoniae strain 168 and its attenuated high-passage strain 168-L and carried out comparative genomic analyses. Results: We performed the first comprehensive analysis of M. hyopneumoniae strain 168 and its attenuated strain and made a preliminary survey of coding sequences (CDSs) that may be related to virulence. The 168-L genome has a highly similar gene content and order to that of 168, but is 4,483 bp smaller because there are 60 insertions and 43 deletions in 168-L. Besides these indels, 227 single nucleotide variations (SNVs) were identified. We further investigated the variants that affected CDSs, and compared them to reported virulence determinants. Notably, almost all of the reported virulence determinants are included in these variants affected CDSs. In addition to variations previously described in mycoplasma adhesins (P97, P102, P146, P159, P216, and LppT), cell envelope proteins (P95), cell surface antigens (P36), secreted proteins and chaperone protein (DnaK), mutations in genes related to metabolism and growth may also contribute to the attenuated virulence in 168-L. Furthermore, many mutations were located in the previously described repeat motif, which may be of primary importance for virulence. Conclusions: We studied the virulence attenuation mechanism of M. hyopneumoniae by comparative genomic analysis of virulent strain 168 and its attenuated high-passage strain 168-L. Our findings provide a preliminary survey of CDSs that may be related to virulence. While these include reported virulence-related genes, other novel virulence determinants were also detected. This new information will form the foundation of future investigations into the pathogenesis of M. hyopneumoniae and facilitate the design of new vaccines

    A Gene Encoding Sialic-Acid-Specific 9-O-Acetylesterase Found in Human Adult Testis

    Using differential display RT-PCR, we identified a gene of 2750 bp from human adult testis, named H-Lse, which encoded a putative protein of 523 amino acids and molecular weight of 58 kd with structural characteristics similar to that of mouse lysosome sialic-acid-specific 9-O-acetylesterase. Northern blot analysis showed a widespread distribution of H-Lse in various human tissues with high expression in the testis, prostate, and colon. In situ hybridization results showed that while H-Lse was not detected in embryonic testis, positive signals were found in spermatocytes but not spermatogonia in adult testis of human. The subcellular localization of H-Lse was visualized by green fluorescent protein (GFP) fused to the amino terminus of H-Lse, showing compartmentalization of H-Lse in large dense-core vesicles, presumably lysosomes, in the cytoplasm. The developmentally regulated and spermatogenic stage-specific expression of H-Lse suggests its possible involvement in the development of the testis and/or differentiation of germ cells
