    The presentation and regulation of the IL-8 network in the epithelial cancer stem-like cell niche in patients with colorectal cancer

    Background: Accumulative evidence suggests that the biological behavior of cancer stem-like cells (CSCs) is regulated by their surrounding niche, in which cytokines function as one of the main mediators for the interaction between CSCs and their microenvironment in the colorectal cancer (CRC). Methods: We characterized the presentation of CSCs and the interleukin (IL)− 8 network in the adenoma/CRC epithelium using quantitative real-time PCR (q-PCR), immunohistochemistry (IHC) and double immunofluorescence. In addition, the capacity of IL-1β to stimulate epithelial IL-8 production in colon cancer Caco-2 cells was examined in vitro and the IL-8 product was measured by enzyme-linked immunosorbent assay (ELISA). Results: IHC observation showed increased expression of both CSCs and IL-8 in the adenoma and CRC epithelium, and q-PCR results revealed that increased expression of IL-1β transcript was strongly correlated with increased IL-8 transcript levels in both adenoma and CRC tissues. Double immunofluorescence images demonstrated the coexpression of the IL-8 receptors IL-8RA and IL-8RB with LGR5 labeled CSCs in CRC tissue sections. Consistently, in vitro experiments showed that coculture of Caco-2 cells with IL-1β at concentrations of 1, 5, 10 and 20 ng/ml resulted in a dose-dependent release of IL-8, which could be specifically inhibited by cotreatment with the IL-1β receptor antagonist. Conclusions: These results demonstrate activation of the IL-8 network in the niche of CSCs from the precancerous adenoma stage to the CRC stage, which is potentially stimulated by IL-1β in CRC cells

    CBSeq: A Channel-level Behavior Sequence For Encrypted Malware Traffic Detection

    Machine learning and neural networks have become increasingly popular solutions for encrypted malware traffic detection. They mine and learn complex traffic patterns, enabling detection by fitting boundaries between malware traffic and benign traffic. Compared with signature-based methods, they have higher scalability and flexibility. However, affected by the frequent variants and updates of malware, current methods suffer from a high false positive rate and do not work well for unknown malware traffic detection. It remains a critical task to achieve effective malware traffic detection. In this paper, we introduce CBSeq to address the above problems. CBSeq is a method that constructs a stable traffic representation, behavior sequence, to characterize attacking intent and achieve malware traffic detection. We novelly propose the channels with similar behavior as the detection object and extract side-channel content to construct behavior sequence. Unlike benign activities, the behavior sequences of malware and its variant's traffic exhibit solid internal correlations. Moreover, we design the MSFormer, a powerful Transformer-based multi-sequence fusion classifier. It captures the internal similarity of behavior sequence, thereby distinguishing malware traffic from benign traffic. Our evaluations demonstrate that CBSeq performs effectively in various known malware traffic detection and exhibits superior performance in unknown malware traffic detection, outperforming state-of-the-art methods.Comment: Submitted to IEEE TIF

    Publishing in English or Chinese: a qualitative analysis of Chinese researchers’ academic language choice

    Non-native language scholars often struggle to choose between English and their native language in scholarly publishing. This study aims to identify the mechanism by which journal attributes influence language choice by investigating the perspectives of 18 Chinese scholars through semi-structured interviews. Drawing on grounded theory, this study develops a model for how journal attributes influence researchers’ language preferences. We find that journal attributes influence researchers’ perceived value which, in turn, affects their particular language choice, with contextual factors playing a moderating role. By examining the motivations underlying Chinese scholars’ language choice, this study provides a critical understanding of the factors shaping their decision-making processes. These findings have significant implications for Chinese scholars, policymakers, and journal operators, shedding light on the issue of discrimination in academic publishing. Addressing these concerns is crucial for fostering a fair and inclusive academic environment

    MLVA distribution characteristics of Yersinia pestis in China and the correlation analysis

    <p>Abstract</p> <p>Background</p> <p><it>Yersinia pestis</it>, the aetiological agent of plague, has been well defined genotypically on local and worldwide scales. In November 2005, five cases of severe pneumonia of unknown causes, resulting in two deaths, were reported in Yulong, Yunnan province. In this study, we compared <it>Y. pestis </it>isolated from the Yulong focus to strains from other areas.</p> <p>Results</p> <p>Two hundred and thirteen <it>Y. pestis </it>strains collected from different plague foci in China and a live attenuated vaccine strain of <it>Y. pestis </it>(EV76) were genotyped using multiple-locus variable-number tandem repeat analysis (MLVA) on 14 loci. A total of 214 <it>Y. pestis </it>strains were divided into 85 MLVA types, and Nei's genetic diversity indices of the various loci ranged between 0.02 - 0.76. Minimum spanning tree analysis showed that <it>Y. pestis </it>in China could be divided into six complexes. It was observed that Microtus strains were different from the other three biovar strains. Each plague focus had its own unique MLVA types.</p> <p>Conclusion</p> <p>The strains isolated from Yulong, Yunnan province had a unique MLVA type, indicating a new clone group. Our results suggest that Yulong strains may have a close relationship with strains from the Qinghai-Tibet Plateau plague focus.</p

    Mechanistic examination of causes for narrow distribution in an endangered shrub: a comparison of its responses to drought stress with a widespread congeneric species

    Although deep rooting is usually considered a drought-tolerant trait, we found that Syringapinnatifolia, a deep rooting and hydrotropic shrub, has a limited distribution in arid areas. To elucidate the mechanisms for its narrow distribution, we conducted two experiments to examine the physiological and morphological responses to water availability and heterogeneity in S. pinnatifolia and a widespread congeneric species, S. oblata. We measured gas exchange, water use efficiency, and plasticity index in plants of these two species grown at different levels of soil water regimes and in containers with patched water distribution. Our results showed that high photosynthetic capacity in the narrowly distributed S. pinnatifolia was an important factor enabling its survival in the harsh sub-alpine environment. High photosynthetic capacity in S. pinnatifolia, however, was obtained at the expense of high transpiratory water loss, resulting in lower integrative water use efficiency. Biomass allocation to roots in S. pinnatifolia increased by 73 % when soil water increased from 75 to 95 % field capacity, suggesting that S. pinnatifolia could be less competitive for above-ground resources under favorable water regimes. The horizontal root hydrotropism and vertical root hydrotropism of S. pinnatifolia in soil with patched water patterns were likely related to compensation for leaf water loss at low soil water level, indicating a limited capacity for homeostasis within the plant for water conservation and lower level of inherent drought-tolerance. In summary, greater degree of morphological plasticity but lower degree of physiological adjustment may be the main causes for the hydrotropism and narrow distribution of S. pinnatifolia in the sub-alpine habitats
