    NEW DATA ON THE RARE CHENGJIANG (LOWER CAMBRIAN, SOUTH CHINA) LINGULOID BRACHIOPOD XIANSHANELLA HAIKOUENSIS

    ABSTRACT—The remarkable brachiopod Xianshanella haikouensis, from the Early Cambrian Chengjiang Lagerstätte, was briefly re-ported based on only two extraordinarily preserved specimens. Details of valve interiors remain unknown due to a lack of available material. We herein reillustrate the holotype of X. haikouensis, and, on the basis of many additional specimens from the type locality, provide further information on this brachiopod taxon, including shell ornamentation, a U-shaped digestive tract, mantle canal systems, lophophores, muscle scars, and well-defined visceral areas in the valves. This exceptional brachiopod is characterized by a biconvex, rounded, knob-shaped shell lacking visible pseudointerarea in the valves, an elongate and muscular pedicle, and, especially, its possession of exceptional setal fringes. A restudy of a number of new specimens justifies assigning Xianshanella to the family Obolidae. The elongate and horny setae of X. haikouensis, coupled with the massive pedicle in many cases with its end attached to shells of other animals, indicate that this species was an epibenthic, sessile suspension feeder, attached by a pedicle

    MiR-486 protects against acute myocardial infarction via regulation of PTEN

    Purpose: To investigate the effect of miR-486 on rats with acute myocardial infarction (AMI), and its mechanism of action.Methods: A rat model of AMI was established. They were randomly divided into 4 groups, namely, sham, model, agomiR-486 and antagomiR-486 groups, respectively. Rats in these different groups were treated with agomiR-21 (5 μL, 40 nmol/mL), antagomiR-21 (5 μL, 40 nmol/mL) or agomiR-NC (5 μL, 40 nmol/mL), respectively. Then, key miRNAs were sorted out using gene-chip assay and verified by quantitative reverse transcription polymerase chain reaction (qRT-PCR) assay. Luciferase reporter gene assay was conducted to determine the interaction between miR-486 and gene of PTEN. After intraperitoneal injection of agomiR-486 and antagomiR-486, hemodynamics was measured to determine the effect of miR-486 on myocardial function of the rats. The effect of miR-486 expression level on the expression of myocardial enzymes in serum, the morphology of myocardial tissues, and the apoptosis of myocardial tissues in rats, were investigated. Additionally, the effect of miR-486 expression level on PTEN/AKT signaling pathway in the rats was determined by Western blotting.Results: The results of gene-chip and qRT-PCR assays revealed that there were 8 differentially expressed genes in rat myocardial tissues in the model group when compared with the sham group. MiR-486 improved the cardiac function of rats and the morphology of myocardial tissues, but reduced AMI-induced apoptosis of myocardial cells and the expression of myocardial enzymes (markers of myocardial injury) in a dose-dependent manner (p < 0.05). The results of luciferase reporter gene assay showed that PTEN was a direct target of miR-486. In rat models of AMI, a raised expression of miR-486 remarkably suppressed the protein expression level of PTEN and up-regulated that of p-AKT/AKT (p < 0.05).Conclusion: MiR-486 protects against AMI in rats probably by targeting PTEN and activating the AKT signaling pathway. The results of the current study may provide new insights for the treatment of AMI

    Evidence for gill slits and a pharynx in Cambrian vetulicolians: implications for the early evolution of deuterostomes.

    BACKGROUND: Vetulicolians are a group of Cambrian metazoans whose distinctive bodyplan continues to present a major phylogenetic challenge. Thus, we see vetulicolians assigned to groups as disparate as deuterostomes and ecdysozoans. This divergence of opinions revolves around a strikingly arthropod-like body, but one that also bears complex lateral structures on its anterior section interpreted as pharyngeal openings. Establishing the homology of these structures is central to resolving where vetulicolians sit in metazoan phylogeny. RESULTS: New material from the Chengjiang Lagerstätte helps to resolve this issue. Here, we demonstrate that these controversial structures comprise grooves with a series of openings. The latter are oval in shape and associated with a complex anatomy consistent with control of their opening and closure. Remains of what we interpret to be a musculature, combined with the capacity for the grooves to contract, indicate vetulicolians possessed a pumping mechanism that could process considerable volumes of seawater. Our observations suggest that food captured in the anterior cavity was transported to dorsal and ventral gutters, which then channeled material to the intestine. This arrangement appears to find no counterpart in any known fossil or extant arthropod (or any other ecdysozoan). Anterior lateral perforations, however, are diagnostic of deuterostomes. CONCLUSIONS: If the evidence is against vetulicolians belonging to one or other group of ecdysozoan, then two phylogenetic options seem to remain. The first is that such features as vetulicolians possess are indicative of either a position among the bilaterians or deuterostomes but apart from the observation that they themselves form a distinctive and recognizable clade current evidence can permit no greater precision as to their phylogenetic placement. We argue that this is too pessimistic a view, and conclude that evidence points towards vetulicolians being members of the stem-group deuterostomes; a group best known as the chordates (amphioxus, tunicates, vertebrates), but also including the ambulacrarians (echinoderms, hemichordates), and xenoturbellids. If the latter, first they demonstrate that these members of the stem group show few similarities to the descendant crown group representatives.     Studies on Chemical Characterization of Ginkgo Amillaria Oral Solution and Its Drug–Drug Interaction With Piceatannol 3′-O-β-D-Glucopyranoside for Injection

    Ginkgo Amillaria oral solution (GAO) is commonly used for the treatment of cardiovascular and cerebrovascular diseases in China. Piceatannol-3′-O-β-D-glucopyranoside for injection (PGI) is mainly used for the prevention and treatment of ischemic cerebrovascular diseases. With the spread of cerebrovascular disease, the possibility of combining the two drugs has increased; however, there is no research on the drug–drug interaction (DDI) between these two medicines. In this paper, an ultrahigh-performance liquid chromatography/quadrupole–orbitrap mass spectrometry (UHPLC/Q-Orbitrap MS) method was established to characterize the chemical constituents of GAO first; 62 compounds were identified or tentatively identified based on their retention time (RT), MS, and MS/MS data. Nine main compounds were determined by ultrahigh-performance liquid chromatography/triple quadrupole mass spectrometry (UPLC-QQQ-MS). Furthermore, incubation with liver microsomes in vitro was fulfilled; the results showed that GAO had a significant inhibitory effect on UGT1A9 and UGT2B7 (p < 0.05), and PGI was mainly metabolized by UGT1A9. The identification results of in vivo metabolites of PGI showed that PGI mainly undergoes a phase II binding reaction mediated by UDP-glucuronosyltransferase (UGT) and sulfotransferase (SULT) in vivo. Therefore, pharmacokinetic studies were performed to investigate the DDI between GAO and PGI. The results showed that the AUC (p < 0.05) and T1/2 (p < 0.05) of PGI in vivo were significantly increased when administered together with GAO, whereas the CL was significantly decreased (p < 0.05).     Pediatric obstructive sleep apnea diagnosis: leveraging machine learning with linear discriminant analysis

    ObjectiveThe objective of this study was to investigate the effectiveness of a machine learning algorithm in diagnosing OSA in children based on clinical features that can be obtained in nonnocturnal and nonmedical environments.Patients and methodsThis study was conducted at Beijing Children's Hospital from April 2018 to October 2019. The participants in this study were 2464 children aged 3–18 suspected of having OSA who underwent clinical data collection and polysomnography(PSG). Participants’ data were randomly divided into a training set and a testing set at a ratio of 8:2. The elastic net algorithm was used for feature selection to simplify the model. Stratified 10-fold cross-validation was repeated five times to ensure the robustness of the results.ResultsFeature selection using Elastic Net resulted in 47 features for AHI ≥5 and 31 features for AHI ≥10 being retained. The machine learning model using these selected features achieved an average AUC of 0.73 for AHI ≥5 and 0.78 for AHI ≥10 when tested externally, outperforming models based on PSG questionnaire features. Linear Discriminant Analysis using the selected features identified OSA with a sensitivity of 44% and specificity of 90%, providing a feasible clinical alternative to PSG for stratifying OSA severity.ConclusionsThis study shows that a machine learning model based on children's clinical features effectively identifies OSA in children. Establishing a machine learning screening model based on the clinical features of the target population may be a feasible clinical alternative to nocturnal OSA sleep diagnosis

    The Mechanical Effect of MnO2 Layers on Electrochemical Actuation Performance of Nanoporous Gold

    This study investigated the electrochemical actuation behavior of nanoporous material during the capacitive process. The length change of nanoporous gold (npg) was in situ investigated in a liquid environment using the dilatometry technique. The mechanical effect of MnO2 layers was introduced in this work to improve the actuation characteristics of the npg samples. Our work found that the actuation behavior of npg sample could be significantly modulated with a covering of MnO2 layers. The electrochemical actuation amplitude was efficiently improved and strongly dependent on the thickness of MnO2 layers covered. Aside from the amplitude, the phase relation between the length change and the electrode potential was inverted when covering the MnO2 layer on the npg samples. This means the expansion of the npg samples and the contraction of samples covered with the MnO2 layer when electrochemical potential sweeps positively. A simple finite element model was built up to understand the effect of the MnO2 layer. The agreement between the simulation result and the experimental data indicates that the sign-inverted actuation-potential response of nanoporous gold contributes to the mechanical effect of MnO2. It is believed that our work could offer a deep understanding on the effect of the MnO2 layer on the electrochemical actuation and then provide a useful strategy to modulate the actuation performance of nanoporous metal materials

    Emotion Recognition and Management in the Tourism Industry During Emergency Events Using Improved Convolutional Neural Network

    The purpose is to optimize emotion recognition capabilities in the tourism industry during emergency events and enhance management efficiency. Hence, based on the principles of emergency events and emotion recognition, this work has outlined the fundamental emotion recognition process. Additionally, an emotion recognition model is constructed based on a Convolutional Neural Network (CNN), and the process of extracting emotions using a three-dimensional (3D) CNN is proposed. Finally, an attention mechanism is employed to optimize the 3D CNN model, and a comparative analysis of accuracy and precision is conducted using the Bimodal Face and Body Gesture Database (FABO). The research findings reveal that the optimized 3D CNN model exhibits lower error rates in recognizing anxiety emotions, with only 3 samples going unrecognized. Its overall recognition performance is superior to other models. There are variations in the recognition accuracy among different models, but in general, the optimized 3D CNN model performs relatively well across various datasets, achieving recognition accuracies of 83.92%, 82.33%, and 88.81%. Compared to other models, the optimized 3D CNN model demonstrates higher precision in recognizing different emotions, particularly excelling in identifying anger, disgust, and happiness, with precision rates of 97%, 91%, and 94%, respectively. This work has improved the accuracy and efficiency of emotion recognition, providing more intelligent and effective support for emergency event management in the tourism industry

    The Effect of an External Magnetic Field on the Electrochemical Capacitance of Nanoporous Nickel for Energy Storage

    This work investigates the effect of a magnetic field on the electrochemical performance of nanoporous nickel (np-Ni). We first compare the electrochemical capacitance of np-Ni electrodes, which were prepared using the chemical dealloying strategy under different magnetic flux densities (B = 0, 500 mT). Our experimental data show that np-Ni500 prepared under an external magnetic field of 500 mT exhibits a much better electrochemical performance, in comparison with that (np-Ni0) prepared without applying a magnetic field. Furthermore, the specific capacitance of the np-Ni0 electrode could be further enhanced when we increase the magnetic flux densities from 0 T to 500 mT, whereas the np-Ni500 electrode exhibits a stable electrochemical performance under different magnetic flux densities (B = 0 mT, 300 mT, 500 mT). This could be attributed to the change in the electrochemical impedance of the np-Ni0 electrode induced by an external magnetic field. Our work thus offers an alternative method to enhance the electrochemical energy storage of materials

    Mask to reconstruct: Cooperative Semantics Completion for Video-text Retrieval

    Recently, masked video modeling has been widely explored and significantly improved the model's understanding ability of visual regions at a local level. However, existing methods usually adopt random masking and follow the same reconstruction paradigm to complete the masked regions, which do not leverage the correlations between cross-modal content. In this paper, we present Mask for Semantics Completion (MASCOT) based on semantic-based masked modeling. Specifically, after applying attention-based video masking to generate high-informed and low-informed masks, we propose Informed Semantics Completion to recover masked semantics information. The recovery mechanism is achieved by aligning the masked content with the unmasked visual regions and corresponding textual context, which makes the model capture more text-related details at a patch level. Additionally, we shift the emphasis of reconstruction from irrelevant backgrounds to discriminative parts to ignore regions with low-informed masks. Furthermore, we design dual-mask co-learning to incorporate video cues under different masks and learn more aligned video representation. Our MASCOT performs state-of-the-art performance on four major text-video retrieval benchmarks, including MSR-VTT, LSMDC, ActivityNet, and DiDeMo. Extensive ablation studies demonstrate the effectiveness of the proposed schemes

    Skipping breakfast and physical fitness among school-aged adolescents

    OBJECTIVES: This study explored the relationship between skipping breakfast and physical fitness in a group of school-aged adolescents in China. METHODS: This cross-sectional study from the Chinese National Surveillance on Students’ Constitution and Health (CNSSCH) survey in Ningbo, China, used a standardized questionnaire to assess the frequency of breakfast consumption. Physical fitness was measured through standing long jump, 50-m sprint, 1,000 (or 800)- m run, and vital capacity tests. Multiple linear regression analysis was used to investigate the relationship between the frequency of breakfast consumption and physical fitness. RESULTS: Our study included a total of 1,849 school-aged adolescents (aged 15.53±1.80 years). Among boys, non-breakfast-skippers had good scores for 50-m sprints, 1,000-m run, and vital capacity tests when compared with breakfast skippers (all po0.05). Among girls, non-breakfast-skippers had a good scores for the standing long jump test compared with breakfast skippers (p=0.003). The multiple linear regression model showed that not skipping breakfast was positively associated with vital capacity (b=-173.78, p=0.004) and inversely associated with 50-m sprint (b=-0.12, p=0.018) and 1,000-m run times (b=-8.08, p=0.001) in boys. CONCLUSION: The results of this cross-sectional study revealed that skipping breakfast might be associated with lower physical fitness in Chinese adolescents aged 13–18 years, especially boys. Breakfast consumption should be promoted among Chinese school-aged boys
