    Analysis of Spatial Travel Association Rules for Rail Transit Based on AFC and POI Data

    In order to explore the spatial distribution rules and causes of urban rail transit passenger travel, this paper mines the spatial 1-frequent itemset and 2-frequent itemsets of weekdays and weekends metro passenger travel based on Apriori algorithm using the continuous week of Automatic Fare Collection System (AFC) swipe card. At the same time, the K-Means algorithm is used to cluster the subway stations and explore the causes of association rules by combining the Point of Interest (POI) data of the same period within the radiation range of the subway stations. The study shows that the spatial distribution pattern of inbound and outbound passenger flow of Shanghai rail transit is consistent between weekdays and weekends, and the outbound passenger flow is more concentrated than the inbound passenger flow, and the significance of weekends is higher; the spatial distribution of metro stations is "circled"; the analysis of the high-lift association rules show that a large passenger flow group centered on the type 3 station is formed in the spatial location, and the passenger flow within the group is mainly commuter flow with separation of employment and residence. The association rule mining of metro passenger travel data is beneficial to understanding the spatial distribution pattern and causes of metro ridership, which can provide reference for rail network planning and operation management

    Pre-Treatment with Melatonin Enhances Therapeutic Efficacy of Cardiac Progenitor Cells for Myocardial Infarction

    Background/Aims: Melatonin possesses many biological activities such as antioxidant and anti-aging. Cardiac progenitor cells (CPCs) have emerged as a promising therapeutic strategy for myocardial infarction (MI). However, the low survival of transplanted CPCs in infarcted myocardium limits the successful use in treating MI. In the present study, we aimed to investigate if melatonin protects against oxidative stress-induced CPCs damage and enhances its therapeutic efficacy for MI. Methods: TUNEL assay and EdU assay were used to detect the effects of melatonin and miR-98 on H2O2-induced apoptosis and proliferation. MI model was used to evaluate the potential cardioprotective effects of melatonin and miR-98. Results: Melatonin attenuated H2O2-induced the proliferation reduction and apoptosis of c-kit+ CPCs in vitro, and CPCs which pretreated with melatonin significantly improved the functions of post-infarct hearts compared with CPCs alone in vivo. Melatonin was capable to inhibit the increase of miR-98 level by H2O2 in CPCs. The proliferation reduction and apoptosis of CPCs induced by H2O2 was aggravated by miR-98. In vivo, transplantation of CPCs with miR-98 silencing caused the more significant improvement of cardiac functions in MI than CPCs. MiR-98 targets at the signal transducer and activator of the transcription 3 (STAT3), and thus aggravated H2O2-induced the reduction of Bcl-2 protein. Conclusions: Pre-treatment with melatonin protects c-kit+ CPCs against oxidative stress-induced damage via downregulation of miR-98 and thereby increasing STAT3, representing a potentially new strategy to improve CPC-based therapy for MI

    Relationship between freight accessibility and logistics employment in US counties

    This paper analyzes the relationship between freight accessibility and logistics employment in the US. It develops an accessibility measure relevant for logistics companies based on a gravity model. This allows for an analysis of the accessibility of US counties focusing on four different modes of transportation: road, rail, air, and maritime. Using a Partial Least Squares model, these four different freight accessibility measures are combined into two constructs, continental and intercontinental freight accessibility, and related to logistics employment. Results show that highly accessible counties attract more logistics employment than other counties. The analyses show that it is very important to control for the effect of the county population on both freight accessibility and logistics employment. While county population explains the most variation in the logistics employment per county, there is a significant relationship between freight accessibility and logistics employment, when controlling for this effect

    Recently, increasing amounts of multi-source geospatial data (raster data of satellites and textual data of meteorological stations) have been generated, which can play a cooperative and important role in many research works. Efficiently storing, organizing and managing these data is essential for their subsequent application. HBase, as a distributed storage database, is increasingly popular for the storage of unstructured data. The design of the row key of HBase is crucial to improving its efficiency, but large numbers of researchers in the geospatial area do not conduct much research on this topic. According the HBase Official Reference Guide, row keys should be kept as short as is reasonable while remaining useful for the required data access. In this paper, we propose a new row key encoding method instead of conventional stereotypes. We adopted an existing hierarchical spatio-temporal grid framework as the row key of the HBase to manage these geospatial data, with the difference that we utilized the obscure but short American Standard Code for Information Interchange (ASCII) to achieve the structure of the grid rather than the original grid code, which can be easily understood by humans but is very long. In order to demonstrate the advantage of the proposed method, we stored the daily meteorological data of 831 meteorological stations in China from 1985 to 2019 in HBase; the experimental result showed that the proposed method can not only maintain an equivalent query speed but can shorten the row key and save storage resources by 20.69% compared with the original grid codes. Meanwhile, we also utilized GF-1 imagery to test whether these improved row keys could support the storage and querying of raster data. We downloaded and stored a part of the GF-1 imagery in Henan province, China from 2017 to 2018; the total data volume reached about 500 GB. Then, we succeeded in calculating the daily normalized difference vegetation index (NDVI) value in Henan province from 2017 to 2018 within 54 min. Therefore, the experiment demonstrated that the improved row keys can also be applied to store raster data when using HBase

    Ramelteon alleviates myocardial ischemia/reperfusion injury (MIRI) through Sirt3-­dependent regulation of cardiomyocyte apoptosis

    Reperfusion stands as a pivotal intervention for ischemic heart disease. However, the restoration of blood flow to ischemic tissue always lead to further damage, which is known as myocardial ischemia/reperfusion injury (MIRI). Ramelteon is an orally administered drug used to improve sleep quality, which is famous for its high bioadaptability and absence of notable addictive characteristics. However, the specific mechanism by which it improves MIRI is still unclear. Sirtuin-3 (Sirt3), primarily located in mitochondria, is crucial in mitigating many cardiac diseases, including MIRI. Based on the structure of Sirt3, we simulated molecular docking and identified several potential amino acid binding sites between it and ramelteon. Therefore, we propose a hypothesis that ramelteon may exert cardioprotective effects by activating the Sirt3 signaling pathway. Our results showed that the activation levels and expression level of Sirt3 were significantly decreased in MIRI tissue and H2O2 stimulated H9C2 cells, while ramelteon treatment upregulated Sirt3 activity and expression. After treat with 3-TYP, a classic Sirt3 activity inhibitor, we constructed myocardial ischemia/reperfusion surgery in vivo and induced H9C2 cells with H2O2 in vitro. The results showed that the myocardial protection and anti-apoptotic effects of ramelteon were antagonized by 3-TYP, indicating that the activation of Sirt3 is a key mechanism for ramelteon to exert myocardial protection. In summary, our results confirm a novel mechanism by which ramelteon improves MIRI by activating Sirt3 signaling pathway, providing strong evidence for the treatment of MIRI with ramelteon
