    Achieving short-cut nitrification and denitrification in modified intermittently aerated constructed wetland

    This study aim to enhance nitrogen removal performance via shifting nitrogen removal pathway from nitrate to nitrite pathway. It was demonstrated that nitrite pathway was successfully and stably achieved in CWs by using modified intermittent aeration control with aeration 20 min/non-aeration 100 min and reducing DO concentration during aeration, nitrite in the effluent could accumulate to over 70% of the total oxidized nitrogen. Q-PCR analysis showed that nitrifying microbial communities were optimized under the alternating anoxic and aerobic conditions, ammonia oxidizing bacteria increased from 7.15 x 10(6) to 8.99 x 10(6) copies/g, while nitrite oxidizing bacteria decreased approximately threefold after 234 days operation. Most importantly, high nitrogen removal efficiency with ammonium removal efficiency of 94.6%, and total nitrogen removal efficiency of 82.6% could be achieved via nitrite pathway even under carbon limiting conditions. In comparison to the nitrate pathway, the nitrite pathway could improve the TN removal by about 55%. (C) 2017 Elsevier Ltd. All rights reserved.</p

    The AI-2/luxS Quorum Sensing System Affects the Growth Characteristics, Biofilm Formation, and Virulence of Haemophilus parasuis

    Haemophilus parasuis (H. parasuis) is a kind of opportunistic pathogen of the upper respiratory tract of piglets. Under certain circumstances, virulent strains can breach the mucosal barrier and enter the bloodstream, causing severe GlĂ€sser's disease. Many virulence factors are found to be related to the pathogenicity of H. parasuis strain, but the pathogenic mechanism remains unclear. LuxS/AI-2, as a kind of very important quorum sensing system, affects the growth characteristics, biofilm formation, antibiotic production, virulence, and metabolism of different strains. In order to investigate the effect of luxS/AI-2 quorum sensing system on the virulence of H. parasuis, a deletion mutant strain (ΔluxS) and complemented strain (C-luxS) were constructed and characterized. The results showed that the luxS gene participated in regulating and controlling stress resistance, biofilm formation and virulence. Compared with wild-type strain, ΔluxS strain decreased the production of AI-2 molecules and the tolerance toward oxidative stress and heat shock, and it reduced the abilities of autoagglutination, hemagglutination, and adherence, whereas it increased the abilities to form biofilm in vitro. In vivo experiments showed that ΔluxS strain attenuated its virulence about 10-folds and significantly decreased its tissue burden of bacteria in mice, compared with the wild-type strain. Taken together, the luxS/AI-2 quorum sensing system in H. parasuis not only plays an important role in growth and biofilm formation, but also affects the pathogenicity of H. parasuis

    Genome-wide association mapping for yield-related traits in soybean (Glycine max) under well-watered and drought-stressed conditions

    Soybean (Glycine max) productivity is significantly reduced by drought stress. Breeders are aiming to improve soybean grain yields both under well-watered (WW) and drought-stressed (DS) conditions, however, little is known about the genetic architecture of yield-related traits. Here, a panel of 188 soybean germplasm was used in a genome wide association study (GWAS) to identify single nucleotide polymorphism (SNP) markers linked to yield-related traits including pod number per plant (PN), biomass per plant (BM) and seed weight per plant (SW). The SLAF-seq genotyping was conducted on the population and three phenotype traits were examined in WW and DS conditions in four environments. Based on best linear unbiased prediction (BLUP) data and individual environmental analyses, 39 SNPs were significantly associated with three soybean traits under two conditions, which were tagged to 26 genomic regions by linkage disequilibrium (LD) analysis. Of these, six QTLs qPN-WW19.1, qPN-DS8.8, qBM-WW1, qBM-DS17.4, qSW-WW4 and qSW-DS8 were identified controlling PN, BM and SW of soybean. There were larger proportions of favorable haplotypes for locus qPN-WW19.1 and qSW-WW4 rather than qBM-WW1, qBM-DS17.4, qPN-DS8.8 and qSW-DS8 in both landraces and improved cultivars. In addition, several putative candidate genes such as Glyma.19G211300, Glyma.17G057100 and Glyma.04G124800, encoding E3 ubiquitin-protein ligase BAH1, WRKY transcription factor 11 and protein zinc induced facilitator-like 1, respectively, were predicted. We propose that the further exploration of these locus will facilitate accelerating breeding for high-yield soybean cultivars

    Clinical application of superselective transarterial embolization of renal tumors in zero ischaemia robotic-assisted laparoscopic partial nephrectomy

    ObjectiveTo assess the feasibility and safety of zero ischaemia robotic-assisted laparoscopic partial nephrectomy (RALPN) after preoperative superselective transarterial embolization (STE) of T1 renal cancer.MethodsWe retrospectively analyzed the data of 32 patients who underwent zero ischaemia RALPN after STE and 140 patients who received standard robot-assisted laparoscopic partial nephrectomy (S-RALPN). In addition, we selected 35 patients treated with off-clamp RALPN (O-RALPN) from September 2017 to March 2022 for comparison. STE was performed by the same interventional practitioner, and zero ischaemia laparoscopic partial nephrectomy (LPN) was carried out by experienced surgeon 1-12 hours after STE. The intraoperative data and postoperative complications were recorded. The postoperative renal function, routine urine test, urinary Computed Tomography (CT), and preoperative and postoperative glomerular filtration rate (GFR) data were analyzed.ResultsAll operations were completed successfully. There were no cases of conversion to opening and no deaths. The renal arterial trunk was not blocked. No blood transfusions were needed. The mean operation time was 91.5 ± 34.28 minutes. The mean blood loss was 58.59 ± 54.11 ml. No recurrence or metastasis occurred.ConclusionFor patients with renal tumors, STE of renal tumors in zero ischaemia RALPN can preserve more renal function, and it provides a safe and feasible surgical method

    Improvement of Muscle Quality of Grass Carp (Ctenopharyngodon idellus) With a Bio-Floating Bed in Culture Ponds

    Muscle quality and the physiological condition of fish are affected greatly by the culture environment. In aquaculture ponds, a bio-floating bed planted with vegetation is often used to improve water quality. This study investigated the growth and muscle quality of grass carp (Ctenopharyngodon idellus) cultured in ponds equipped with bio-floating beds. Fish were cultured in two replicated pond groups from May to November. Fish in the first group were cultured in experimental ponds equipped with a bio-floating bed planted with Ipomoea aquatica, whereas fish in the other group were reared in control ponds without a bio-floating bed. Compared with control ponds, the experimental ponds had better water quality with significantly lower concentrations of nitrite and ammonia. Grass carp in the experimental group had greater muscle weight gain, a significantly higher content of crude protein, and a significantly lower crude fat level than fish in the control group. The levels of pH, water-holding capacity, and antioxidant capacity of muscle decreased significantly in the control group compared to the experimental group. Texture profile analysis revealed higher values of hardness, springiness, gumminess, and chewiness and lower values of cohesiveness and resilience of white muscle in the experimental group compared to the control group. The filets of fish in the experimental group also received higher grades in the sensory evaluation of springiness, overall acceptability, aroma, and palatability. These results indicate that growth performance and muscle quality of grass carp were improved by the presence of bio-floating beds in the culture ponds
