This article provided data on how to develop an immunoturbidimetric assay (ITA) of enzyme/mutants as proteins in cell lysates in high-throughput (HTP) mode together with HTP assay of their activities to derive their specific activities in cell lysates for comparison, with Pseudomonas aeruginosa arylsulfatase (PAAS) and Bacillus fastidious uricase (BFU) plus their mutants as models.     Plant Expression of Cocaine Hydrolase-Fc Fusion Protein for Treatment of Cocaine Abuse

    BACKGROUND: A recently reported cocaine hydrolase (CocH3) fused with fragment crystallizable (Fc) region of human immunoglobulin G1, denoted as CocH3-Fc, is known as a promising therapeutic candidate for the treatment of cocaine overdose and addiction. A challenge for practical therapeutic use of this enzyme exists in the large-scale protein production and, therefore, it is interesting to identify a low-cost and feasible, sustainable source of CocH3-Fc production. RESULTS: CocH3-Fc was transiently expressed in plant Nicotiana benthamiana leaves. The plant-expressed protein, denoted as pCocH3-Fc, was as active as that expressed in mammalian cells both in vitro and in vivo. However, compared to the mammalian-cell expressed CocH3-Fc protein, pCocH3-Fc had a shorter biological half-life, probably due to the lack of protein sialylation in plant. Nevertheless, the in vivo half-life was significantly extended upon the PEGylation of pCocH3-Fc. The Fc fusion did not prolong the biological half-life of the plant-expressed enzyme pCocH3-Fc, but increased the yield of the enzyme expression in the plant under the same experimental conditions. CONCLUSIONS: It is feasible to express pCocH3-Fc in plants. Further studies on the pCocH3-Fc production in plants should focus on the development of vectors with additional genes/promoters for the complete protein sialylation and for a better yield

    Higgs boson decays to BcB_c meson in the fragmentation-function approach

    In the paper, we present a calculation of the decay widths for the Higgs boson decays to the BcB_c, BcB_c^*, Bc(21S0)B_c(2^1S_0) and Bc(23S1)B_c^*(2^3S_1) mesons using the fragmentation-function approach. In the calculation, the fragmentation functions up to order αs3\alpha_s^3 based on the nonrelativistic QCD factorization theory are used, and the decay widths for HQ+XH\to Q+X and Hg+XH \to g+X at the partonic level are calculated up to order αs\alpha_s. The large logarithms of mH2/mBc2m_H^2/m_{Bc}^2 are resummed up to next-to-leading logarithmic accuracy by solving the evolution equations for the running quark masses and the fragmentation functions. Compared to the leading-order decay widths based on the nonrelativistic QCD approach, the decay widths based on the fragmentation-function approach that include the higher-order QCD corrections are reduced significantly. Our numerical results show that there are about 1.2×1051.2\times 10^5 BcB_c events via the Higgs decays to be produced at the HL-LHC with 3ab13ab^{-1}, and about 1.6×1061.6\times 10^6 BcB_c events via the Higgs decays to be produced at the HE-LHC with 15ab115ab^{-1}.Comment: 12 pages, 9 figure

    Synergism and rules from combination of Baicalin, Jasminoidin and Desoxycholic acid in refined Qing Kai Ling for treat ischemic stroke mice model.

    Refined Qing-Kai-Ling (QKL), a modified Chinese medicine, consists of three main ingredients (Baicalin, Jasminoidin and Desoxycholic acid), plays a synergistic effect on the treatment of the acute stage of ischemic stroke. However, the rules of the combination and synergism are still unknown. Based on the ischemic stroke mice model, all different kinds of combination of Baicalin, Jasminoidin, and Desoxycholic acid were investigated by the methods of neurological examination, microarray, and genomics analysis. As a result, it confirmed that the combination of three drugs offered a better therapeutical effect on ischemic stroke than monotherapy of each drug. Additionally, we used Ingenuity pathway Analysis (IPA) and principal component analysis (PCA) to extract the dominant information of expression changes in 373 ischemia-related genes. The results suggested that 5 principal components (PC1-5) could account for more than 95% energy in the gene data. Moreover, 3 clusters (PC1, PC2+PC5, and PC3+PC4) were addressed with cluster analysis. Furthermore, we matched PCs on the drug-target networks, the findings demonstrated that Baicalin related with PC1 that played the leading role in the combination; Jasminoidin related with PC2+PC5 that played a compensatory role; while Desoxycholic acid had the least performance alone which could relate with PC3+PC4 that played a compatible role. These manifestations were accorded with the principle of herbal formulae of Traditional Chinese Medicine (TCM), emperor-minister-adjuvant-courier. In conclusion, we firstly provided scientific evidence to the classic theory of TCM formulae, an initiating holistic viewpoint of combination therapy of TCM. This study also illustrated that PCA might be an applicable method to analyze the complicated data of drug combination

    Constraining Cosmology with High Convergence Regions in Weak Lensing Surveys

    We propose to use a simple observable, the fractional area of "hot spots" in weak gravitational lensing mass maps which are detected with high significance, to determine background cosmological parameters. Because these high-convergence regions are directly related to the physical nonlinear structures of the universe, they derive cosmological information mainly from the nonlinear regime of density fluctuations. We show that in combination with future cosmic microwave background anisotropy measurements, this method can place constraints on cosmological parameters that are comparable to those from the redshift distribution of galaxy cluster abundances. The main advantage of the statistic proposed in this paper is that projection effects, normally the main source of uncertainty when determining the presence and the mass of a galaxy cluster, here serve as a source of information.Comment: 14 pages, 4 figures, accepted for publication in Astrophysical Journa
