    MONTHLY ISOTOPIC SIGNAL OF THE PRECIPITATED WATER IN THE PROVINCE OF TRENTO: LAGRANGIAN ANALYSIS AND DISCUSSION OF MEASUREMENTS

    The present work has been performed in the framework of the research project AQUAPAST. The main focus of AQUAPAST is the reconstruction of past climate changes in the atmospheric circulation and in particular in the water vapour transport mechanism over the Mediterranean area starting from the analysis of the isotopic content of cave speleothemes in the Trentino Province (North-East of Italy). A Lagrangian methodology for the reconstruction and the analysis of the airstreams which govern the transport of water vapour has been applied to recent events. The average monthly isotopic signal of the precipitation water sampled in stations located in key geographic settings in the Province of Trento is used as tracer to infer the origin of the precipitating water and to validate the Lagrangian analysis. A comparison between the isotopic signal of November 2002 and November 2003 is here presented, as most of the yearly precipitation falls in November over the Eastern Alps. Hypotheses are proposed about the meteorological factors (e.g. monthly averaged sea surface temperature, atmospheric circulation, etc.) determining different isotopic signals in the two years

    Antibacterial Activity of Iranian Green and Black Tea on Streptococcus Mutans: An In Vitro Study

    Objective: Dental caries is a common infectious disease.Streptococcus mutans is the prevalent decay microorganism. The anti Streptococcus mutans activity of non fermented and semi-fermented tea has been shown. The aim of this study was to determine the anti Streptococcus mutans activity of Iranian green and black tea (non fermented and fermented type).Materials and Methods: The study was experimental. The aerial parts of wild-growing Camellia sinensis were collected from Lahijan province. The methanolic extract of green and black tea were examined on Streptococcus mutans (ATCC3566). Five different concentrations (50mg/ml, 100mg/ml, 200mg/ml, 300mg/ml and 400 mg/ml) of tea extracts were tested using the well assay method. The agar dilution method recommended by the NCCLS standards (National Committee for Clinical Laboratory Standards) was used. Theminimum inhibitory concentration (MIC) was determined as the lowest concentration of extract inhibiting visible growth of the organism on the agar media plate. Minimum bactericidal concentration (MBC) was detected from MIC.Results: The Iranian green and black tea had an antibacterial effect on 100 to 400 mg/ml concentrations. The minimum inhibitory concentration of green and black tea was 150 and 50 mg/ml, respectively. The mean diameter of inhibition zone were 9.5 mm and 10.9 mm for methanolic extract of green and black tea, respectively.Conclusion: Both Iranian non fermented (green tea) and fermented (black tea) have anti Streptococcus mutans activity in vitro. The anti Streptococcus mutans activity of black tea appears on a lower concentration than green tea

    Human monoclonal antibodies to domain C of tenascin-C selectively target solid tumors in vivo

    We had previously reported that splice isoforms of tenascin-C containing the extra-domain C are virtually absent in normal adult tissues but are highly abundant in high-grade astrocytomas, with a prominent peri-vascular pattern of expression. We now report that the extra-domain C of tenascin-C is strongly expressed in the majority of lung cancers, with a vascular and stromal pattern of expression. Using antibody phage technology, we have generated a human monoclonal antibody (G11), with a dissociation constant KD = 4.2 nM for the human domain C. The G11 antibody, expressed in scFv and in mini-antibody (SIP) format, as well as a scFv-interleukin-2 fusion protein, was then characterized in quantitative biodistribution studies using mice grafted subcutaneously with U87 gliomas, revealing a selective tumor uptake, with tumor/blood ratios up to 11.8:1 at 24 h. A radioiodinated preparation of SIP(G11) was also investigated in a double tracer study using an orthotopic rat glioma model, confirming the antibody's ability to preferentially localize at the tumor site, with tumor/brain ratios superior to the ones observed with 18F-fluorodeoxyglucose. These tumor-targeting properties, together with the strong immunohistochemical staining of human tumor sections, indicate that the G11 antibody may be used as a portable targeting moiety for the selective delivery of imaging and therapeutic agents to gliomas and lung tumor

    Selective formation, reactivity, redox and magnetic properties of MnIII and FeIII dinuclear complexes with shortened salen-type schiff base ligands

    The reactivity of the shortened salen-type ligands H3salmp, H2salmen and H2sal(p-X)ben with variable para-substituent on the central aromatic ring (X = tBu, Me, H, F, Cl, CF3, NO2) towards the trivalent metal ions manganese(III) and iron(III) is presented. The selective formation of the dinuclear complexes [M2(m-salmp)2], M = Mn (1a), Fe (2a), [M2(m-salmen)2(m-OR)2)], R = Et, Me, H and M = Mn (3a\u2013c) or Fe (4a\u2013c), and (M2(m-sal[p-X]ben)2(m-OMe)2), X = tBu, Me, H, F, Cl, CF3, NO2 and M = Mn (5a\u2013g) or Fe (6a\u2013g), could be identified by reaction of the Schiff bases with metal salts and the base NEt3, and their characterization through elemental analysis, infrared spectroscopy, mass spectrometry and single-crystal X-ray diffraction of 2a.2AcOEt, 2a.2CH3CN and 3c.2DMF was performed. In the case of iron(III) and H3salmp, when using NaOH as a base instead of NEt3, the dinuclear complexes [Fe2(m-salmp)(m-OR)(salim)2], R = Me, H (2b\u2013c) could be isolated and spectroscopically characterized, including the crystal structure of 2b.1.5H2O, which showed that rupture of one salmp3\u2013 to two coordinated salim\u2013 ligands and release of one salH molecule occurred. The same hydrolytic tendency could be identified with sal(p-X)ben ligands in the case of iron(III) also by using NEt3 or upon standing in solution, while manganese(III) did not promote such a C\u2013N bond breakage. Cyclic voltammetry studies were performed for 3b, 4b, 5a and 6a, revealing that the iron(III) complexes can be irreversibly reduced to the mixed-valence FeIIFeIII and FeII2 dinuclear species, while the manganese(III) derivatives can be reversibly oxidized to either the mixed-valence MnIIIMnIV or to the MnIV2 dinuclear species. The super-exchange interaction between the metal centers, mediated by the bridging ligands, resulted in being antiferromagnetic (AFM) for the selected dinuclear compounds 3b, 4b, 5a, 5e, 5f, 6a and 6e. The coupling constants J (\u20132J \u15c1\ub7\u15c2 formalism) had values around \u201313 cm\u20131 for manganese(III) compounds, among the largest AFM coupling constants reported so far for dinuclear MnIII2 derivatives, while values between \u20133 and \u201310 cm\u20131 were obtained for iron(III) compounds

    The properties of charge-exchange excitations of 208{}^ {208}Pb with ΔL=0\Delta L = 0, i.e., the isobaric analog and Gamow-Teller resonances, are studied within a self-consistent model making use of an effective force of the Skyrme type. The well-known isobaric analog case is used to assess the reliability of the model. The calculated properties of the Gamow-Teller resonance are compared with recent experimental measurements with the aim of better understanding the microscopic structure of this mode.Comment: 26 pages including references, figure captions and tables.     The Molecular and Cellular Basis of Tumor Rejection After Vaccination With Mammary Adenocarcinoma Cells Transduced With the MHC Class II Transactivator CIITA

    CD8+ T cell responses are major players of tumor eradication in various vaccination protocols. However, an optimal stimulation of CD4+ T helper cells is required for both priming and maintenance of the effector CTL response against the tumor. In this study we show that the murine mammary adenocarcinoma cell line TS/A, a highly malignant MHC-II-negative tumor, is rejected in vivo if genetically engineered to express MHC-II molecules by transfer of the MHC-II transactivator CIITA. TS/ACIITA cells are fully rejected by 93% of the syngeneic recipients and have a significantly lower growth rate in the remaining 7% of animals. Rejection requires CD4+ and CD8+ cells. CD4+ T cells are fundamental in the priming phase, whereas CTLs are the major anti-tumor effectors. All tumor rejecting animals are protected against rechallenge with the parental TS/A tumor. Immunohistochemical data at day 5 post-inoculation showed an higher infiltrate of CD4+ T cells in mice bearing TS/A-CIITA, than in mice bearing the TS/A tumor. Subsequently, from day 7 trough day 10, TS/A-CIITA tumors showed higher number of both CD4+ and CD8+ cells, dendritic cells, together with massive necrosis. The frequency of IFN-αsecreting splenocytes early after inoculations was also assessed by an ex vivo ELISPOT assay. Only the rejecting TS/A-CIITA animals showed an high frequency of IFN-αsecreting cells (between 80 and 120/106 splenocytes). Importantly, CD4 and CD8 depletion experiments revealed that at the time of tumor resolution the major cell population recognizing the TS/A-CIITA cells was of CD4 origin. This is the first example of successful tumor vaccination by genetic transfer of CIITA.     We present a new method of obtaining time-dependent matrix elements of the electromagnetic pulse produced by a highly-relativistic projectile. These matrix elements are used in a coupled-channel calculation to predict the cross-sections for population of 1- and 2-phonon states of the giant dipole resonance.
