    Platelet hyperaggregability in high-fat fed rats: A role for intraplatelet reactive-oxygen species production

    <p>Abstract</p> <p>Background</p> <p>Adiposity greatly increases the risk of atherothrombotic events, a pathological condition where a chronic state of oxidative stress is reported to play a major role. This study aimed to investigate the involvement of (NO)-soluble guanylyl cyclase (sGC) signaling pathway in the platelet dysfunction from high fat-fed (HFF) rats.</p> <p>Methods</p> <p>Male Wistar rats were fed for 10 weeks with standard chow (SCD) or high-fat diet (HFD). ADP (10 μM)- and thrombin (100 mU/ml)-induced washed platelet aggregation were evaluated. Measurement of intracellular levels of ROS levels was carried out using flow cytometry. Cyclic GMP levels were evaluated using ELISA kits.</p> <p>Results</p> <p>High-fat fed rats exhibited significant increases in body weight, epididymal fat, fasting glucose levels and glucose intolerance compared with SCD group. Platelet aggregation induced by ADP (<it>n </it>= 8) and thrombin from HFD rats (<it>n </it>= 8) were significantly greater (<it>P </it>< 0.05) compared with SCD group. Platelet activation with ADP increased by 54% the intraplatelet ROS production in HFD group, as measured by flow cytometry (<it>n </it>= 6). N-acetylcysteine (NAC; 1 mM) and PEG-catalase (1000 U/ml) fully prevented the increased ROS production and platelet hyperaggregability in HFD group. The NO donors sodium nitroprusside (SNP; 10 μM) and SNAP (10 μM), as well as the NO-independent soluble guanylyl cyclase stimulator BAY 41-2272 (10 μM) inhibited the platelet aggregation in HFD group with lower efficacy (<it>P </it>< 0.05) compared with SCD group. The cGMP levels in response to these agents were also markedly lower in HFD group (<it>P </it>< 0.05). The prostacyclin analogue iloprost (1 μM) reduced platelet aggregation in HFD and SCD rats in a similar fashion (<it>n </it>= 4).</p> <p>Conclusions</p> <p>Metabolic abnormalities as consequence of HFD cause platelet hyperaggregability involving enhanced intraplatelet ROS production and decreased NO bioavailability that appear to be accompanied by potential defects in the prosthetic haem group of soluble guanylyl cyclase.</p

    Effect of 6-months of physical exercise on the nitrate/nitrite levels in hypertensive postmenopausal women

    <p>Abstract</p> <p>Background</p> <p>Evidences have showed that the incidence of arterial hypertension is greater in postmenopausal women as compared to premenopausal. Physical inactivity has been implicated as a major contributor to weight gain and abdominal obesity in postmenopausal women and the incidence of cardiovascular disease increases dramatically after menopause. Additionally, more women than men die each year of coronary heart disease and are twice as likely as men to die within the first year after a heart attack. A healthy lifestyle has been strongly associated with the regular physical activity and evidences have shown that physically active subjects have more longevity with reduction of morbidity and mortality. Nitric oxide (NO) produced by endothelial cells has been implicated in this beneficial effect with improvement of vascular relaxing and reduction in blood pressure in both laboratory animals and human. Although the effect of exercise training in the human cardiovascular system has been largely studied, the majority of these studies were predominantly conducted in men or young volunteers. Therefore, the aim of this work was to investigate the effects of 6 months of dynamic exercise training (ET) on blood pressure and plasma nitrate/nitrite concentration (NOx<sup>-</sup>) in hypertensive postmenopausal women.</p> <p>Methods</p> <p>Eleven volunteers were submitted to the ET consisting in 3 days a week, each session of 60 minutes during 6 months at moderate intensity (50% of heart rate reserve). Anthropometric parameters, blood pressure, NOx<sup>- </sup>concentration were measured at initial time and after ET.</p> <p>Results</p> <p>A significant reduction in both systolic and diastolic blood pressure values was seen after ET which was accompanied by markedly increase of NOx<sup>- </sup>levels (basal: 10 ± 0.9; ET: 16 ± 2 μM). Total cholesterol was significantly reduced (basal: 220 ± 38 and ET: 178 ± 22 mg/dl), whereas triglycerides levels were not modified after ET (basal: 141 ± 89 and ET: 147 ± 8 mg/dl).</p> <p>Conclusion</p> <p>Our study shows that changing in lifestyle promotes reduction of arterial pressure which was accompanied by increase in nitrite/nitrate concentration. Therefore, 6-months of exercise training are an important approach in management arterial hypertension and play a protective effect in postmenopausal women.</p

    Centralized cytogenetic analysis of pediatric acute leukemia: results of an italian collaborative experience

    Background and Objective. Cytogenetic analysis of acute leukemia yields important information which has been demonstrated to be correlated to patient survival. A reference laboratory was created in order to perform karyotype analysis on all cases of acute leukemia enrolled in the AIEOP (Associazione Italiana Emato-Oncologia Pediatrica) protocols. Methods. From January 1990 to December 1995, 1115 samples of children with ALL or AML were sent in for cytogenetic analysis. The results of cell cultures were screened in the Reference Laboratory and then the fixed metaphases were sent to one of the six cytogenetic laboratories for analysis. Results. The leukemic karyotypes of 556 patients were successfully analyzed. An abnormal clone was detected in 49% of cases of ALL and in 66% of AML. In ALL the most frequent abnormality was 9p rearrangement. Other recurrent abnormalities were t(9;22), t(4;11) and t(1;19). In AML t(8;21), t(15;17) and 11q23 rearrangement were the most frequent structural abnormalities. These findings are similar to the results obtained in other multicenter studies using a similar approach. Interpretation and Conclusions. Our data confirm the feasibility of performing cytogenetic analysis in a centralized laboratory on mailed samples of bone marrow and/or peripheral blood; this is very important considering that cytogenetic analysis of neoplastic tissue requires a special laboratory and expert staff

    Apesar do filme para passivação padrão da indústria de células solares atual ser SiNx, as últimas tendências do mercado fazem com que seja necessário reavaliar o SiO2 para passivação das superfícies de células solares. Para crescer estes óxidos, no entanto, são necessárias etapas de processamento em altas temperaturas, que deixam as lâminas de silício suscetíveis a contaminações metálicas. Para evitar essa contaminação, o composto orgânico Trans-LC é utilizado para limpeza do tubo de oxidação. O objetivo deste trabalho é avaliar a eficácia deste composto para limpeza dos tubos e determinar a qualidade da passivação que os óxidos produzidos desta forma proporcionam para as lâminas de Si. Observou-se que as limpezas realizadas não foram capazes de prevenir a contaminação de lâminas de Si de alto tempo de vida inicial. Porém, o Trans-LC foi suficientemente eficaz para oxidações realizadas em lâminas de baixo tempo de vida dos portadores minoritários.Even though the standard film for passivation currently utilized in the silicon solar cell industry is SiNx, the late tendencies of the market make it necessary to reevaluate the SiO2 for surface passivation of solar cells. In order to grow these oxides, however, high-temperature processing steps are necessary, which leave the wafers susceptible to metallic contamination. To avoid this contamination, the organic compound Trans-LC is used to clean the oxidation tube. The goal of this work is to evaluate the efficacy of this compound for the tube cleaning and determine the passivation quality that the oxides produced this way provide to the Si wafers. It was observed that the cleanings were not capable to prevent the contamination of high initial lifetime Si wafers.
