Hypospadias anatomy: Elastosonographic evaluation of the normal and hypospadic penis
Background Hypospadias is one of the most common congenital anomalies in childhood. The aim of this study is to apply elastosonography on normal and hypospadic penis to verify the structural differences in tissues composition and stiffness. Materials and methods We analyzed medical chart of patients treated at our Institution for hypospadias during the period December 2005 and December 2014 (group 1). Other two groups were enrolled for this study: group 2- patients with hypospadias waiting for surgery and group 3-patients without hypospadias. Inclusion and exclusion criteria were created; all patients underwent penile ultrasound and elastosonography. Elastographic index of elasticity was defined as soft, medium-hard or hard. We assigned the value 1 to soft tissue, 2 and 3 to medium-hard and hard respectively. Results During the study period 294 patients were treated for hypospadias. After reviewing medical chart 115 patients were considered for analysis (group 1). 22 patients were enrolled in group 2 and 38 patients were enrolled in group 3. Group 1: 7 proximal hypospadias, 29 penile hypospadias, 79 distal hypospadias. Patients with hypospadias had malformation also at corpus spongiosum and cavernosum respect to controls. Elastography showed a corpus spongiosum stiffness defined as medium-hard or hard in all cases of the pathologic group and soft in all the subjects of the control group (p < 0.05). Conclusions Elastosonography showed how the hypospadia anatomy is deeply altered, even in an anatomical area far from meatal abnormality: corpus spongiosum in hypospadic penis seems to be globally stiffer and less elastic and cavernous corpora are less developed.[Figure presented
Shrec'16 Track: Retrieval of Human Subjects from Depth Sensor Data
International audienceIn this paper we report the results of the SHREC 2016 contest on "Retrieval of human subjects from depth sensor data". The proposed task was created in order to verify the possibility of retrieving models of query human subjects from single shots of depth sensors, using shape information only. Depth acquisition of different subjects were realized under different illumination conditions, using different clothes and in three different poses. The resulting point clouds of the partial body shape acquisitions were segmented and coupled with the skeleton provided by the OpenNI software and provided to the participants together with derived triangulated meshes. No color information was provided. Retrieval scores of the different methods proposed were estimated on the submitted dissimilarity matrices and the influence of the different acquisition conditions on the algorithms were also analyzed. Results obtained by the participants and by the baseline methods demonstrated that the proposed task is, as expected, quite difficult, especially due the partiality of the shape information and the poor accuracy of the estimated skeleton, but give useful insights on potential strategies that can be applied in similar retrieval procedures and derived practical applications. A metabolite-derived protein modification integrates glycolysis with KEAP1-NRF2 signalling.
Mechanisms that integrate the metabolic state of a cell with regulatory pathways are necessary to maintain cellular homeostasis. Endogenous, intrinsically reactive metabolites can form functional, covalent modifications on proteins without the aid of enzymes1,2, and regulate cellular functions such as metabolism3-5 and transcription6. An important 'sensor' protein that captures specific metabolic information and transforms it into an appropriate response is KEAP1, which contains reactive cysteine residues that collectively act as an electrophile sensor tuned to respond to reactive species resulting from endogenous and xenobiotic molecules. Covalent modification of KEAP1 results in reduced ubiquitination and the accumulation of NRF27,8, which then initiates the transcription of cytoprotective genes at antioxidant-response element loci. Here we identify a small-molecule inhibitor of the glycolytic enzyme PGK1, and reveal a direct link between glycolysis and NRF2 signalling. Inhibition of PGK1 results in accumulation of the reactive metabolite methylglyoxal, which selectively modifies KEAP1 to form a methylimidazole crosslink between proximal cysteine and arginine residues (MICA). This posttranslational modification results in the dimerization of KEAP1, the accumulation of NRF2 and activation of the NRF2 transcriptional program. These results demonstrate the existence of direct inter-pathway communication between glycolysis and the KEAP1-NRF2 transcriptional axis, provide insight into the metabolic regulation of the cellular stress response, and suggest a therapeutic strategy for controlling the cytoprotective antioxidant response in several human diseases
From privileged natural product scaffolds to PNA-encoded chemical libraries
En croisant notre connaissance en synthèse de collections encodées par APN, notre expérience obtenue lors de la synthèse des quelques membres de la famille des lactones d'acide résorcylique, et notre constant intérêt dans l'inhibition de kinases et chaperonnes moléculaires, nous avons construit plusieurs collections moléculaires encodées par APN avec des pharmacophores variés incluant des hétérocycles, des acides aminés, des dérivés synthétiques de lactones d'acide résorcylique, et des médicaments autorisés par la FDA ou des fragments de ceux-ci, comme autant de points de diversité, dirigés contre différentes protéines cibles. La chaperonne moléculaire HSP70, les protéines contenant un bromodomaine et d'autres kinases ont toutes été exposées à ces collections encodées par APN et résolues spatialement sur micropuce ADN, démontrant ainsi la robustesse et la puissance de cette méthodologie de criblage dans une tentative de dépasser l'effet de goulot en découverte médicamenteuse

Peptide nucleic acid (PNA)-encoded chemical libraries along with DNA-encoded libraries have provided a powerful new paradigm for library synthesis and ligand discovery.
