    Challenges and Techniques for Personal Environment Management

    People today use the computer for many simultaneous work projects and activities. The traditional file system was developed for storing and retrieving files and it and the desktop have not evolved with users' practices. The first part of the dissertation presents a user study that generates a better understanding of the issues and practices regarding the organization of documents in support of activities. The second part provides the design of an environment to organize information based on an activity paradigm as opposed to an archiving paradigm and delivers the instantiation and the evaluation of a system based on such a design. The system, called Docksy, provides an environment structured in workspaces. Each workspace is segmented in areas or panels. Users can use documents as elements to structure their workflow or to manage their activities by separating files in the different panels, and by adding comments, tags, and flags. The Docksy design aims to create a flexible, lightweight environment that is easy to use and can be incorporated into users' daily practice, old or new. Such a system could be used to learn about users' practices and their evolution. Docksy was therefore developed for a double purpose; the short term purpose of testing new features (panels, comments, flags, and tags) and the long term purpose of facilitating learning about user practices. A study of Docksy use was conducted in which twenty participants used Docksy for at least two weeks and they were then interviewed. The study showed that participants valued the panels and the comment features. The results of the study showed the potential for changing users' practice and the potential for the system to be adopted by users

    Algorithms Aside: Recommendation as the Lens of Life

    In this position paper, we take the experimental approach of putting algorithms aside, and reflect on what recommenders would be for people if they were not tied to technology. By looking at some of the shortcomings that current recommenders have fallen into and discussing their limitations from a human point of view, we ask the question: if freed from all limitations, what should, and what could, RecSys be? We then turn to the idea that life itself is the best recommender system, and that people themselves are the query. By looking at how life brings people in contact with options that suit their needs or match their preferences, we hope to shed further light on what current RecSys could be doing better. Finally, we look at the forms that RecSys could take in the future. By formulating our vision beyond the reach of usual considerations and current limitations, including business models, algorithms, data sets, and evaluation methodologies, we attempt to arrive at fresh conclusions that may inspire the next steps taken by the community of researchers working on RecSys

    Protocol for the PreventIT feasibility randomised controlled trial of a lifestyle-integrated exercise intervention in young older adults

    Introduction The European population is rapidly ageing. In order to handle substantial future challenges in the healthcare system, we need to shift focus from treatment towards health promotion. The PreventIT project has adapted the Lifestyle-integrated Exercise (LiFE) programme and developed an intervention for healthy young older adults at risk of accelerated functional decline. The intervention targets balance, muscle strength and physical activity, and is delivered either via a smartphone application (enhanced LiFE, eLiFE) or by use of paper manuals (adapted LiFE, aLiFE). Methods and analysis The PreventIT study is a multicentre, three-armed feasibility randomised controlled trial, comparing eLiFE and aLiFE against a control group that receives international guidelines of physical activity. It is performed in three European cities in Norway, Germany, and The Netherlands. The primary objective is to assess the feasibility and usability of the interventions, and to assess changes in daily life function as measured by the Late-Life Function and Disability Instrument scale and a physical behaviour complexity metric. Participants are assessed at baseline, after the 6 months intervention period and at 1 year after randomisation. Men and women between 61 and 70 years of age are randomly drawn from regional registries and respondents screened for risk of functional decline to recruit and randomise 180 participants (60 participants per study arm). Ethics and dissemination Ethical approval was received at all three trial sites. Baseline results are intended to be published by late 2018, with final study findings expected in early 2019.     Identification of Novel Pax8 Targets in FRTL-5 Thyroid Cells by Gene Silencing and Expression Microarray Analysis

    The differentiation program of thyroid follicular cells (TFCs), by far the most abundant cell population of the thyroid gland, relies on the interplay between sequence-specific transcription factors and transcriptional coregulators with the basal transcriptional machinery of the cell. However, the molecular mechanisms leading to the fully differentiated thyrocyte are still the object of intense study. The transcription factor Pax8, a member of the Paired-box gene family, has been demonstrated to be a critical regulator required for proper development and differentiation of thyroid follicular cells. Despite being Pax8 well-characterized with respect to its role in regulating genes involved in thyroid differentiation, genomics approaches aiming at the identification of additional Pax8 targets are lacking and the biological pathways controlled by this transcription factor are largely unknown.To identify unique downstream targets of Pax8, we investigated the genome-wide effect of Pax8 silencing comparing the transcriptome of silenced versus normal differentiated FRTL-5 thyroid cells. In total, 2815 genes were found modulated 72 h after Pax8 RNAi, induced or repressed. Genes previously reported to be regulated by Pax8 in FRTL-5 cells were confirmed. In addition, novel targets genes involved in functional processes such as DNA replication, anion transport, kinase activity, apoptosis and cellular processes were newly identified. Transcriptome analysis highlighted that Pax8 is a key molecule for thyroid morphogenesis and differentiation.This is the first large-scale study aimed at the identification of new genes regulated by Pax8, a master regulator of thyroid development and differentiation. The biological pathways and target genes controlled by Pax8 will have considerable importance to understand thyroid disease progression as well as to set up novel therapeutic strategies

    Low Dosage of Histone H4 Leads to Growth Defects and Morphological Changes in Candida albicans

    Chromatin function depends on adequate histone stoichiometry. Alterations in histone dosage affect transcription and chromosome segregation, leading to growth defects and aneuploidies. In the fungal pathogen Candida albicans, aneuploidy formation is associated with antifungal resistance and pathogenesis. Histone modifying enzymes and chromatin remodeling proteins are also required for pathogenesis. However, little is known about the mechanisms that generate aneuploidies or about the epigenetic mechanisms that shape the response of C. albicans to the host environment. Here, we determined the impact of histone H4 deficit in the growth and colony morphology of C. albicans. We found that C. albicans requires at least two of the four alleles that code for histone H4 (HHF1 and HHF22) to grow normally. Strains with only one histone H4 allele show a severe growth defect and unstable colony morphology, and produce faster-growing, morphologically stable suppressors. Segmental or whole chromosomal trisomies that increased wild-type histone H4 copy number were the preferred mechanism of suppression. This is the first study of a core nucleosomal histone in C. albicans, and constitutes the prelude to future, more detailed research on the function of histone H4 in this important fungal pathogen

In this thesis we describe a study performed on fifth grade students. The study describes how elementary school students used physical manipulations in conjunction with the digital interface of educational software for geometry. The blending of physical manipulations and digital interface helped the students to overcome the limits of the representation and interaction modalities of the digital interface. The addition of physical manipulations also helped them to consolidate their physical knowledge of the world and smooth the transition from the physical world to the abstract

    Patterns of Reading and Organizing Information in Document Triage

    People engaged in knowledge work must often rapidly identify valuable material from within large sets of potentially relevant documents. Document triage is a type of sensemaking task that involves skimming documents to get a sense of their content, evaluating documents to assess their worth in the context of the current activity, and organizing documents to prepare for their subsequent use and more in-depth reading. We have performed a study of document triage by collecting multiple forms of qualitative and quantitative data to characterize how 24 subjects read about a new topic and assessed and organized a set of 40 relevant Web documents. Our results indicate that there are multiple strategies for document triage, each involving different styles of reading, interacting, and organizing. Common strategies include: 1) focused reading early in the task, relegating the organizing until later in the process; 2) skimming performed in tandem with organizing, which relies on gaining an incremental understanding of the topic; and 3) metadata-based organizing, a strategy that stresses working with document surrogates to minimize the time spent reading. The findings suggest ways applications may better support the intertwined nature of the browsing, reading, and organizing activities in document triage

    Transmission of BSA during Cross-flow Microfiltration: Influence of pH and Salt Concentration

    In many downstream processes microfiltration (MF) is used as the first step to separate the micro-organisms from the product-containing broth. The transmission of proteins through these MF membranes is of great importance, especially when a protein is the desired product. Usually, the largest fraction of the proteins retained in the membrane during the separation of micro-organisms is retained in the filter cake of micro-organisms that is built up on the membrane surface. However, this is not always the case. In some cases the transmission of proteins can decrease below 100% even though there are no micro-organisms present in the broth. In the present study the transmission of BSA through two different MF membranes (a nylon membrane, Pall Filtron Ultipore, Nylon66, 0.2 m, and a membrane made of polyether sulphone, Pall Filtron, Omega, 0.16 m) was investigated. The transmission of BSA was highest for the membrane that was said to be a low protein binding membrane (Omega) and it was affected by the pH in the solution. At pH 5, which is close to the IEP of the protein, the transmission was 100%, while at pH 3 and 7 the transmission was much lower (sometimes as low as 40%). However, when the ionic strength in the solution was increased the transmission of BSA through both MF membranes increased dramatically (at pH 3 and 7). This shows that the electrostatic interactions between the protein and the filter cake (made up of protein aggregates) and between the protein and the membrane affect the transmission. The increase in transmission when the protein is close to its IEP (at pH 5), is believed to be due to the lack of electrostatic repulsion between the proteins in the solution and filter cake of protein aggregates formed on the membrane. At high ionic strength the charged protein molecules are shielded from each other and from the membrane and the filter cake, by the ions in the solution. Due to this shielding effect the protein molecules act as if they were uncharged and the transmission of BSA is thus increased
