    Chemical Composition, Antioxidant, and Antimicrobial Activities of Lichen Umbilicaria cylindrica (L.) Delise (Umbilicariaceae)

    The phytochemical analysis of methanol and chloroform extracts of Umbilicaria cylindrica was determined by HPLC-UV method. The predominant phenolic compound in both extracts was depsidone, salazinic acid (1). Besides salazinic acid, the tested extracts of U. cylindrica contain norstictic acid (2), methyl-β-orcinol carboxylate (3), ethyl haematommate (4), atranorin (5), and usnic acid (6), in different amounts and relations. The lichen extracts showed comparable and strong antioxidant activity, exhibited higher DPPH and hydroxyl radical scavengings, chelating activity, and inhibitory activity towards lipid peroxidation. The lichen extracts demonstrated important antimicrobial activity against eight strains with MIC values from 15.62 to 62.50 μg/mL. This is the first report of the detail chemical composition and antioxidant activity of the lichen Umbilicaria cylindrica, and the results suggest that this lichen can be used as a new source of the natural antioxidants and the substances with antimicrobial features

    Innovating Nature-based Solutions: learnings from the EU Horizon 2020 RECONECT project

We developed and applied the methodology to scout NbS-related innovations that are at a certain TRL and had the potential for commercial exploitation, and then included tailored activities to help these solutions to reach at least CRI-1 (i.e. have the business model outlined, initial market research done, business case formulated). The tools and technologies were assessed through a lens of innovation, explication, and upscaling. As a result, novel guidelines tailored for NbS innovations were established to introduce a participatory approach in TRL and CRI evaluations to support commercial exploitation for upscaling NbS into mainstream society through market uptake of H2020 frameworks.Horizon Europe 2020 Programm

Nature-based solutions (NBSs) seem to be a suitable response, integrating the hydrology, geomorphology, hydraulic, and ecological dynamics. While there are some methods and tools for suitability mapping of small-scale NBSs, literature concerning the spatial allocation of large-scale NBSs is still lacking. The present work aims to develop new toolboxes and enhance an existing methodology by developing spatial analysis tools within a geographic information system (GIS) environment to allocate large-scale NBSs based on a multi-criteria algorithm. The methodologies combine machine learning spatial data processing techniques and hydrodynamic modelling for allocation of large-scale NBSs. The case studies concern selected areas in the Netherlands, Serbia, and Bolivia, focusing on three large-scale NBS: rainwater harvesting, wetland restoration, and natural riverbank stabilisation. Information available from the EC H2020 RECONECT project as well as other available data for the specific study areas was used. The research highlights the significance of incorporating machine learning, GIS, and remote sensing techniques for the suitable allocation of large-scale NBSs.     Singularities of nn-fold integrals of the Ising class and the theory of elliptic curves

    We introduce some multiple integrals that are expected to have the same singularities as the singularities of the n n-particle contributions χ(n)\chi^{(n)} to the susceptibility of the square lattice Ising model. We find the Fuchsian linear differential equation satisfied by these multiple integrals for n=1,2,3,4 n=1, 2, 3, 4 and only modulo some primes for n=5 n=5 and 6 6, thus providing a large set of (possible) new singularities of the χ(n)\chi^{(n)}. We discuss the singularity structure for these multiple integrals by solving the Landau conditions. We find that the singularities of the associated ODEs identify (up to n=6n= 6) with the leading pinch Landau singularities. The second remarkable obtained feature is that the singularities of the ODEs associated with the multiple integrals reduce to the singularities of the ODEs associated with a {\em finite number of one dimensional integrals}. Among the singularities found, we underline the fact that the quadratic polynomial condition 1+3w+4w2=0 1+3 w +4 w^2 = 0, that occurs in the linear differential equation of χ(3) \chi^{(3)}, actually corresponds to a remarkable property of selected elliptic curves, namely the occurrence of complex multiplication. The interpretation of complex multiplication for elliptic curves as complex fixed points of the selected generators of the renormalization group, namely isogenies of elliptic curves, is sketched. Most of the other singularities occurring in our multiple integrals are not related to complex multiplication situations, suggesting an interpretation in terms of (motivic) mathematical structures beyond the theory of elliptic curves.Comment: 39 pages, 7 figure

    Differences in genomic abnormalities among African individuals with monoclonal gammopathies using calculated ancestry

    Multiple myeloma (MM) is two- to three-fold more common in African Americans (AAs) compared to European Americans (EAs). This striking disparity, one of the highest of any cancer, may be due to underlying genetic predisposition between these groups. There are multiple unique cytogenetic subtypes of MM, and it is likely that the disparity is associated with only certain subtypes. Previous efforts to understand this disparity have relied on self-reported race rather than genetic ancestry, which may result in bias. To mitigate these difficulties, we studied 881 patients with monoclonal gammopathies who had undergone uniform testing to identify primary cytogenetic abnormalities. DNA from bone marrow samples was genotyped on the Precision Medicine Research Array and biogeographical ancestry was quantitatively assessed using the Geographic Population Structure Origins tool. The probability of having one of three specific subtypes, namely t(11;14), t(14;16), or t(14;20) was significantly higher in the 120 individuals with highest African ancestry (≥80%) compared with the 235 individuals with lowest African ancestry (<0.1%) (51% vs. 33%, respectively, p value = 0.008). Using quantitatively measured African ancestry, we demonstrate a major proportion of the racial disparity in MM is driven by disparity in the occurrence of the t(11;14), t(14;16), and t(14;20) types of MM

    Whole-genome sequencing of multiple myeloma reveals oncogenic pathways are targeted somatically through multiple mechanisms.

    Multiple myeloma (MM) is a biologically heterogeneous malignancy, however, the mechanisms underlying this complexity are incompletely understood. We report an analysis of the whole-genome sequencing of 765 MM patients from CoMMpass. By employing promoter capture Hi-C in naïve B-cells, we identify cis-regulatory elements (CREs) that represent a highly enriched subset of the non-coding genome in which to search for driver mutations. We identify regulatory regions whose mutation significantly alters the expression of genes as candidate non-coding drivers, including copy number variation (CNV) at CREs of MYC and single-nucleotide variants (SNVs) in a PAX5 enhancer. To better inform the interplay between non-coding driver mutations with other driver mechanisms, and their respective roles in oncogenic pathways, we extended our analysis identifying coding drivers in 40 genes, including 11 novel candidates. We demonstrate the same pathways can be targeted by coding and non-coding mutations; exemplified by IRF4 and PRDM1, along with BCL6 and PAX5, genes that are central to plasma cell differentiation. This study reveals new insights into the complex genetic alterations driving MM development and an enhanced understanding of oncogenic pathways

    Pulmonary valve papillary fibroelastoma diagnosed by echocardiography: a case report

    Papillary fibroelastomas (PFEs) are rare cardiac valve tumours with reported incidence of <0.03% according to autopsy studies. Among them, pulmonary valve PFEs are extremely rare. With wider use of echocardiography, they are being increasingly recognized premortem. We describe a case of a 32-year-old Caucasian woman with PFE of pulmonic valve diagnosed by echocardiography. The patient underwent surgery due to high mobility of the tumour and high risk of embolic complications. The surgery was done with complete tumour resection and total preservation of valve function. This case report discusses diagnostics of PFEs, their characteristic echocardiographic and histological features, and possible complications and suggests treatment options in this rare cardiac tumour
