    IP3 receptor isoforms differently regulate ER-mitochondrial contacts and local calcium transfer

    Contact sites of endoplasmic reticulum (ER) and mitochondria locally convey calcium signals between the IP3 receptors (IP3R) and the mitochondrial calcium uniporter, and are central to cell survival. It remains unclear whether IP3Rs also have a structural role in contact formation and whether the different IP3R isoforms have redundant functions. Using an IP3R-deficient cell model rescued with each of the three IP3R isoforms and an array of super-resolution and ultrastructural approaches we demonstrate that IP3Rs are required for maintaining ER-mitochondrial contacts. This role is independent of calcium fluxes. We also show that, while each isoform can support contacts, type 2 IP3R is the most effective in delivering calcium to the mitochondria. Thus, these studies reveal a non-canonical, structural role for the IP3Rs and direct attention towards the type 2 IP3R that was previously neglected in the context of ER-mitochondrial calcium signaling

    Optimised insert design for improved single-molecule imaging and quantification through CRISPR-Cas9 mediated knock-in

The use of CRISPR-Cas9 genome editing to introduce endogenously expressed tags has the potential to address a number of the classical limitations of single molecule localisation microscopy. In this work we present the first systematic comparison of inserts introduced through CRISPR-knock in, with the aim of optimising this approach for single molecule imaging. We show that more highly monomeric and codon optimised variants of mEos result in improved expression at the TubA1B locus, despite the use of identical guides, homology templates, and selection strategies. We apply this approach to target the G protein-coupled receptor (GPCR) CXCR4 and show a further insert dependent effect on expression and protein function. Finally, we show that compared to over-expressed CXCR4, endogenously labelled samples allow for accurate single molecule quantification on ligand treatment. This suggests that despite the complications evident in CRISPR mediated labelling, the development of CRISPR-PALM has substantial quantitative benefits

    Discrepant perceptions of communication, teamwork and situation awareness among surgical team members

    Objective To assess surgical team members’ differences in perception of non-technical skills. Design Questionnaire design. Setting Operating theatres (OTs) at one university hospital, three teaching hospitals and one general hospital in the Netherlands. Participants Sixty-six surgeons, 97 OT nurses, 18 anaesthetists and 40 nurse anaesthetists. Methods All surgical team members, of five hospitals, were asked to complete a questionnaire and state their opinion on the current state of communication, teamwork and situation awareness at the OT. Results Ratings for ‘communication’ were significantly different, particularly between surgeons and all other team members (P ? 0.001). The ratings for ‘teamwork’ differed significantly between all team members (P ? 0.005). Within ‘situation awareness’ significant differences were mainly observed for ‘gathering information’ between surgeons and other team members (P < 0.001). Finally, 72–90% of anaesthetists, OT nurses and nurse anaesthetists rated routine team briefings and debriefings as inadequate. Conclusions This study shows discrepancies on many aspects in perception between surgeons and other surgical team members concerning communication, teamwork and situation awareness. Future research needs to ascertain whether these discrepancies are linked to greater risk of adverse events or to process as well as systems failures.     Soil management options to reduce runoff and erosion on a hardsetting Alfisol in the semi-arid tropics

    Improved farming systems are needed to enhance productivity and reduce degradation on hardsetting Alfisols in the semi-arid tropics. A long-term experiment was started in July 1988 at ICRISAT, Hyderabad, India, to evaluate practices to improve infiltration and reduce erosion by stimulating biological activity and protecting soil in the rainy season. This paper outlines the rationale for the experiment, describes it, and summarises early results. Fifteen treatments in a randomised block design were applied to plots 28.5 m long by 5 m wide on a 2% slope. Nine annual treatments made up a factorial sub-set: tillage by tined implement to three depths (0, 10 and 20 cm) combined with three mulch treatments (no mulch, farmyard manure (FYM) at 15 t ha−1, and rice straw at 5 t ha−1). Pearl millet (Pennisetum glaucum) was sown in the factorial sub-set in July 1988. Six perennial species ley treatments (combinations of perennial pigeonpea (Cajanus cajan), verano stylo (Stylosanthes hamata), and buffel grass (Cenchrus ciliaris)) completed the randomised block design. All treatments were replicated three times making 45 plots in total. Plots were instrumented to measure runoff and soil loss. Millet straw yield was reduced by tillage to 20 cm and grain yield was significantly reduced by tillage to both 10 and 20 cm. Mulches had no effect on millet straw yield but FYM significantly increased millet grain yield. The proportion of rainfall running off plots ranged from 15.8 to 39.1%. Perennial species treatments tended to have higher runoff than treatments under millet, but this was not consistent as zero tillage without mulch, and tillage to 10 cm either with mulch or with FYM mulch, also had relatively high runoff. Rice straw mulch significantly reduced runoff by comparison with FYM and no mulch. Soil was lost mainly as suspended load, indicating that raindrop detachment was the main erosion process. Effects of perennial species were variable and probably reflected the degree of surface protection provided in this establishment phase. Tillage significantly increased suspended load concentration in the first major runoff event but not in an event later in the season. Rice straw mulch significantly reduced bed load concentration in the first event and reduced suspended load in the later event. The ability of straw mulch to reduce runoff volume and sediment concentration suggests mulch-based systems may be able to contribute to sustainability of farming systems in the semi-arid tropics. The lack of beneficial effects from tillage suggests a limited role for tillage-based systems. Results from subsequent years are needed to form definite conclusions and to show effects of slower biologically-induced changes in the soil

    Narrative Exposure Therapy as a treatment for child war survivors with posttraumatic stress disorder: Two case reports and a pilot study in an African refugee settlement

    BACKGROUND: Little data exists on the effectiveness of psychological interventions for children with posttraumatic stress disorder (PTSD) that has resulted from exposure to war or conflict-related violence, especially in non-industrialized countries. We created and evaluated the efficacy of KIDNET, a child-friendly version of Narrative Exposure Therapy (NET), as a short-term treatment for children. METHODS: Six Somali children suffering from PTSD aged 12–17 years resident in a refugee settlement in Uganda were treated with four to six individual sessions of KIDNET by expert clinicians. Symptoms of PTSD and depression were assessed pre-treatment, post-treatment and at nine months follow-up using the CIDI Sections K and E. RESULTS: Important symptom reduction was evident immediately after treatment and treatment outcomes were sustained at the 9-month follow-up. All patients completed therapy, reported functioning gains and could be helped to reconstruct their traumatic experiences into a narrative with the use of illustrative material. CONCLUSIONS: NET may be safe and effective to treat children with war related PTSD in the setting of refugee settlements in developing countries

    Mercury DPM: fast, flexible particle simulations in complex geometries part II: applications

    MercuryDPM is a particle-simulation software developed open-source by a global network of researchers. It was designed ​ab initio to simulate realistic geometries and materials, thus it contains several unique features not found in any other particle simulation software. These features have been discussed in a companion paper published in the DEM7 conference proceedings; here we present several challenging setups implemented in MercuryDPM ​ . Via these setups, we demonstrate the unique capability of the code to simulate and analyse highly complex geotechnical and industrial applications.These tups implemented include complex geometries such as (i) a screw conveyor, (ii) steady-state inflow conditions for chute flows, (iii) a confined conveyor belt to simulate a steady-state breaking wave, and(iii)aquasi-2D cylindrical slice to efficiently study shear flows.​MercuryDPM is also parallel, which we showcase via a multi-million particle simulations of a rotating drum. We further demonstrate how to simulate complex particle interactions, including: (i)deformable, charged clay particles; and (ii) liquid bridges and liquid migration in wet particulates, (iii) non-spherical particles implemented via superquadrics. Finally, we show how to analyse and complex systems using the unique micro-macro mapping (coarse-graining) tool MercuryCG

    Simple mindreading abilities predict complex theory of mind: developmental delay in autism spectrum disorders

    Theory of Mind (ToM) is impaired in individuals with Autism Spectrum Disorders (ASD). The aims of this study were to: i) examine the developmental trajectories of ToM abilities in two different mentalizing tasks in children with ASD compared to TD children; and ii) to assess if a ToM simple test known as Eyes-test could predict performance on the more advanced ToM task, i.e. Comic Strip test. Based on a sample of 37 children with ASD and 55 TD children, our results revealed slower development at varying rates in all ToM measures in children with ASD, with delayed onset compared to TD children. These results could stimulate new treatments for social abilities, which would lessen the social deficit in ASD
