Naringenin suppresses neutrophil infiltration into adipose tissue in high-fat diet-induced obese mice
Recruitment of immune cells to adipose tissue is altered dramatically in obesity, which results in chronic inflammation ofthe adipose tissue that leads to metabolic disorders, such as insulin resistance and type 2 diabetes mellitus. The regulationof immune cell infiltration into adipose tissue has prophylactic and therapeutic implications for obesity-related diseases. Wepreviously showed that naringenin, a citrus flavonoid, suppressed macrophage infiltration into adipose tissue by inhibitingmonocyte chemoattractant protein-1 (MCP-1) expression in the progression phase to high-fat diet (HFD)-induced obesity.In the current study, we evaluated the effects of naringenin on neutrophil infiltration into adipose tissue, because neutrophilsalso infiltrate into adipose tissue in the progression phase to obesity. Naringenin suppressed neutrophil infiltration into adiposetissue induced by the short-term (2 weeks) feeding of a HFD to mice. Naringenin tended to inhibit the HFD-inducedexpression of several chemokines, including MCP-1 and MCP-3, in adipose tissue. Naringenin also inhibited MCP-3 expressionin 3T3-L1 adipocytes and a co-culture of 3T3-L1 adipocytes and RAW264 macrophages. However, naringenin did notaffect the expression of macrophage inflammatory protein-2 (MIP-2), an important chemokine for neutrophil migration andactivation, in macrophages or in a co-culture of adipocytes and macrophages. Nach der japanischen Weltanschauung ist die Natur das Leben nach dem Tode (die Heimat der Seele) und auch die Welt der Götter (anima). Wenn man die Bedeutsamkeit der japanischen „Pflege der Toten“ (Totenmesse) und der Pflege der Natur (Götter) durch das traditionelle Ritual unter ökologischem Gesichtspunkt neu bewertet, wird das zur Folge haben, ein vom modernen europäischen Prinzip der „Herrschaft“ und „Bedürfnisbefriedigung“ distanziertes Ethos in der Gesellschaft fördern zu helfen
Microscopic characterization of the C–F bonds in fluorine–graphite intercalation compounds
The structures of fluorine–graphite intercalation compounds (F-GICs, C₂.₈F and C₃.₅F) have been analyzed by high-resolution transmission electron microscopy (TEM). Cross-sectional TEM images of the F-GICs indicate that the interlayer distance increases by insertion of fluorine with randomly buckled carbon layers. Such a structure can form by alternation in the bond angle at a carbon atom covalently bonded with fluorine. Electron energy loss spectroscopy combined with TEM indicates that the π-orbital network over the graphitic carbon layer reduces with fluorination. The C–F bond is essentially covalent
Bifidobacterium response to lactulose ingestion in the gut relies on a solute-binding protein-dependent ABC transporter
ビフィズス菌がラクチュロースを利用する仕組みを解明 --ビフィズス菌の増殖作用の予測への活用も--. 京都大学プレスリリース. 2021-05-24.This study aims to understand the mechanistic basis underlying the response of Bifidobacterium to lactulose ingestion in guts of healthy Japanese subjects, with specific focus on a lactulose transporter. An in vitro assay using mutant strains of Bifidobacterium longum subsp. longum 105-A shows that a solute-binding protein with locus tag number BL105A_0502 (termed LT-SBP) is primarily involved in lactulose uptake. By quantifying faecal abundance of LT-SBP orthologues, which is defined by phylogenetic analysis, we find that subjects with 10⁷ to 10⁹ copies of the genes per gram of faeces before lactulose ingestion show a marked increase in Bifidobacterium after ingestion, suggesting the presence of thresholds between responders and non-responders to lactulose. These results help predict the prebiotics-responder and non-responder status and provide an insight into clinical interventions that test the efficacy of prebiotics
Ventricular pacing inhibition by oversensing due to diaphragmatic myopotential during deep inspiration
AbstractAn 80-year-old man, who had dilated cardiomyopathy with right ventricular (RV) dilatation, underwent implantable cardioverter defibrillator (ICD) implantation for advanced atrioventricular block and primary prevention of sudden cardiac death. Tined and screw-in leads were placed on the right atrial appendage and RV apex, respectively. Ventricular pacing inhibition was detected after surgery due to oversensing by diaphragmatic myopotential occurring only during deep inspiration. We performed re-surgery and switched the screw-in lead for a tined lead. The diaphragmatic myopotential decreased, thereby improving oversensing by diaphragmatic myopotential and ventricular pacing inhibition. It might be beneficial to use a tined lead when placing the ventricular lead at the RV apex for implantation of a pacemaker or ICD if oversensing of diaphragmatic myopotential is observed using a screw-in lead.<Learning objective: Oversensing due to diaphragmatic myopotential is rarely observed. However if it occurs, it becomes a critical problem, which causes pacemaker inhibition or inappropriate ICD shock. However, the method of preventing this problem is unknown. In this case, we demonstrated that a tined-lead may be useful for the prevention of oversensing by diaphragmatic myopotential.
Clinical Guides for aHUS
Atypical hemolytic uremic syndrome (aHUS) is a rare disease characterized by the triad of microangiopathic hemolytic anemia, thrombocytopenia, and acute kidney injury. In 2013, we developed diagnostic criteria to enable early diagnosis and timely initiation of appropriate treatment for aHUS. Recent clinical and molecular findings have resulted in several proposed classifications and definitions of thrombotic microangiopathy and aHUS. Based on recent advances in this field and the emerging international consensus to exclude secondary TMAs from the definition of aHUS, we have redefined aHUS and proposed diagnostic algorithms, differential diagnosis, and therapeutic strategies for aHUS
The Japanese Society of Pathology Guidelines on the handling of pathological tissue samples for genomic research: Standard operating procedures based on empirical analyses
Genome research using appropriately collected pathological tissue samples is expected to yield breakthroughs in the development of biomarkers and identification of therapeutic targets for diseases such as cancers. In this connection, the Japanese Society of Pathology (JSP) has developed “The JSP Guidelines on the Handling of Pathological Tissue Samples for Genomic Research” based on an abundance of data from empirical analyses of tissue samples collected and stored under various conditions. Tissue samples should be collected from appropriate sites within surgically resected specimens, without disturbing the features on which pathological diagnosis is based, while avoiding bleeding or necrotic foci. They should be collected as soon as possible after resection: at the latest within about 3 h of storage at 4°C. Preferably, snap‐frozen samples should be stored in liquid nitrogen (about −180°C) until use. When intending to use genomic DNA extracted from formalin‐fixed paraffin‐embedded tissue, 10% neutral buffered formalin should be used. Insufficient fixation and overfixation must both be avoided. We hope that pathologists, clinicians, clinical laboratory technicians and biobank operators will come to master the handling of pathological tissue samples based on the standard operating procedures in these Guidelines to yield results that will assist in the realization of genomic medicine
