    Targeted next-generation sequencing of dedifferentiated chondrosarcoma in the skull base reveals combined TP53 and PTEN mutations with increased proliferation index, an implication for pathogenesis

    Dedifferentiated chondrosarcoma (DDCS) is a rare disease with a dismal prognosis. DDCS consists of two morphologically distinct components: the cartilaginous and noncartilaginous components. Whether the two components originate from the same progenitor cells has been controversial. Recurrent DDCS commonly displays increased proliferation compared with the primary tumor. However, there is no conclusive explanation for this mechanism. In this paper, we present two DDCSs in the sellar region. Patient 1 exclusively exhibited a noncartilaginous component with a TP53 frameshift mutation in the pathological specimens from the first surgery. The tumor recurred after radiation therapy with an exceedingly increased proliferation index. Targeted next-generation sequencing (NGS) revealed the presence of both a TP53 mutation and a PTEN deletion in the cartilaginous and the noncartilaginous components of the recurrent tumor. Fluorescence in situ hybridization and immunostaining confirmed reduced DNA copy number and protein levels of the PTEN gene as a result of the PTEN deletion. Patient 2 exhibited both cartilaginous and noncartilaginous components in the surgical specimens. Targeted NGS of cells from both components showed neither TP53 nor PTEN mutations, making Patient 2 a naïve TP53 and PTEN control for comparison. In conclusion, additional PTEN loss in the background of the TP53 mutation could be the cause of increased proliferation capacity in the recurrent tumor

    Expression and prognostic significance of Golgiglycoprotein73 (GP73) with Epithelial-mesenchymal transition (EMT) related molecules in Hepatocellular Carcinoma (HCC)

    BACKGROUND: Hepatocellular carcinoma (HCC) is the fifth most common cancer and the third cause of cancer-related deaths, worldwide. It is essential to develop an effective prognostic biomarker and determine the mechanisms underlying HCC invasion and metastasis. AIMS: This study aimed to investigate the expression of Golgi glycoprotein73 (GP73) and Epithelial-mesenchymal transition (EMT) molecules such as E-cadherin and Vimentin in HCC. We also evaluated the prognostic value of GP73 in HCC. METHODS: Immunohistochemistry (IHC) was used to determine the expression of GP73 and EMT molecules in 75 HCC specimens and the corresponding paracarcinomatous liver (PCL) tissues. Spearman’s correlation test was used to analyze the correlation of GP73 and EMT molecules. Clinicopathological features of the HCC patients were also analyzed. Univariate survival analysis was performed by the Kaplan–Meier method and differences among the groups were analyzed by the Log-rank test. RESULTS: GP73 expression in HCC was higher compared with PCL tissues (χ( 2 ) = 73.60, P < 0.05). EMT molecules were also detected in HCC and PCL tissues. GP73 was negatively correlated with E-cadherin (r = − 0.49, P < 0.05), but positively correlated with Vimentin (r = 0.46, P < 0.05) in HCC. GP73 was correlated with the clinicopathological features including Edmondson grade, vascular invasion and TNM stage (P < 0.05), which was also associated with overall survival (OS) (P < 0.05). CONCLUSIONS: GP73 was negatively with E-cadherin and positively correlated with Vimentin. It might be associated with aggressive behavior of HCC and had influence on patients’ OS. Further research is needed to determine the potential of GP73.     GPS Scintillations and TEC Variations in Association With a Polar Cap Arc

    A unique example of a polar cap arc producing clear amplitude and phase scintillations in GPS L-band signals is presented using observations from an all-sky imager and a GPS receiver and a digital ionosonde at Resolute Bay and the SuperDARN Inuvik radar. During the southward interplanetary magnetic field (IMF) condition, the polar cap arc moved quickly from the dusk-side to the midnight auroral oval at a speed of ∼700 m/s, as revealed by all-sky 557.7 and 630.0 nm images. When it intersected the raypath of GPS signals, both amplitude and phase scintillations appeared, which is very different from previous results. Moreover, the scintillations were precisely determined through power spectral analysis. We propose that the strong total electron content (TEC) enhancement (∼6 TECU) and flow shears in association with the polar cap arc under the southward IMF condition were creating the scintillations. It provides evidence for the existence of polar cap arc scintillations that may be harmful for satellite applications even through L-band signals.publishedVersio

    Surface-Based Regional Homogeneity in First-Episode, Drug-Naive Major Depression: A Resting-State fMRI Study

    Background. Previous volume-based regional homogeneity (ReHo) studies neglected the intersubject variability in cortical folding patterns. Recently, surface-based ReHo was developed to reduce the intersubject variability and to increase statistical power. The present study used this novel surface-based ReHo approach to explore the brain functional activity differences between first-episode, drug-naive MDD patients and healthy controls. Methods. Thirty-three first-episode, drug-naive MDD patients and 32 healthy controls participated in structural and resting-state fMRI scans. MDD patients were rated with a 17-item Hamilton Rating Scale for Depression prior to the scan. Results. In comparison with the healthy controls, MDD patients showed reduced surface-based ReHo in the left insula. There was no increase in surface-based ReHo in MDD patients. The surface-based ReHo value in the left insula was not significantly correlated with the clinical information or the depressive scores in the MDD group. Conclusions. The decreased surface-based ReHo in the left insula in MDD may lead to the abnormal top-down cortical-limbic regulation of emotional and cognitive information. The surface-based ReHo may be a useful index to explore the pathophysiological mechanism of MDD.</p

    No association of the insulin gene VNTR polymorphism with polycystic ovary syndrome in a Han Chinese population

    <p>Abstract</p> <p>Background</p> <p>Polycystic ovary syndrome (PCOS) is a common endocrine disorder associated with an increased risk of type II diabetes mellitus. The results of previous research about the association of the VNTR polymorphism in 5-prime flanking region of the insulin (INS) gene with PCOS have been inconsistent. The present study was to investigate the association of the INS-VNTR polymorphism with PCOS in a Han Chinese population.</p> <p>Methods</p> <p>The -23/HphI polymorphism as a surrogate marker of the INS-VNTR length polymorphism was genotyped by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) in 216 PCOS patients and 192 non-PCOS women as a control group. Allelic and genotypic frequencies were compared between patients and controls, and these results were analyzed in respect to clinical test data.</p> <p>Results</p> <p>No significant differences were observed between the cases and controls groups either in allele (P = 0.996) or genotype (P = 0.802) frequencies of INS-VNTR polymorphism; Regarding anthropometric data and hormone levels, there were no significant differences between INS-VNTR genotypes in the PCOS group, as well as in the non-PCOS group.</p> <p>Conclusion</p> <p>The present study demonstrated for the first time that the INS-VNTR polymorphism is not a key risk factor for sporadic PCOS in the Han Chinese women. Further studies are needed to give a global view of this polymorphism in pathogenesis of PCOS in a large-scale sample, family-based association design or well-defined subgroups of PCOS.</p

    Latent profile analysis of psychological needs thwarting in Chinese school teachers: longitudinal associations with problematic smartphone use, psychological distress, and perceived administrative support

    IntroductionIn light of the significant impact that teachers have on education quality and student growth, their mental health warrants special attention. With the increasing popularity of Information and Communication Technology (ICT) and the rise of online teaching during the pandemic, teachers have become a group prone to developing problematic smartphone use (PSU). Psychological need thwarting (PNT) has been shown to be closely related to PSU, psychological distress, and perceived administrative support. However, most previous studies have adopted a variable-centered approach, which may overlook the possibility that the three basic needs are not closely associated and could form distinct profiles. Therefore, this study aims to apply latent profile analysis to identify different PNT profiles and their associations with PSU, psychological distress, and perceived administrative support.MethodsA longitudinal survey was conducted using convenience and purposive sampling methods. The survey involved 1,642 primary and middle school teachers working in China over a two-month interval, with the first assessment in November 2021 (Time 1) and the second in January 2022 (Time 2).ResultsThe results indicate that a three-profile model, intricately based on the PNT data gathered at Time 1, is most optimal: Class 1 is labeled as ‘High autonomy-High competence and Moderate relatedness thwarting’, Class 2 as ‘High autonomy-High competence and High relatedness thwarting’, and Class 3 as ‘Low psychological needs thwarting’. Distinct associations were observed among the three profiles concerning PSU, psychological distress, and perceived administrative support. Specifically, in terms of PSU, the score of Class 2 was higher than Class 1, with that of Class 3 being the lowest at Time 1, while at Time 2 no significant difference was found between any two of these three groups; in terms of distress, the scores of the three profiles were arranged from high to low as Class 2, 1, and 3 at both time points; and in terms of perceived administrative support, the order was just the opposite, with 3, 1, and 2 from high to low at both Time 1 and Time 2.ConclusionNotably, the consistent ranking of the three classes in terms of psychological distress and administrative support suggests a lasting influence of PNT. Future studies should explore this enduring impact further by employing additional longitudinal data sets and examining potential mediators or moderators beyond the current study’s scope

    A flexible Cu-based catalyst system for the transformation of fructose to furanyl ethers as potential bio-fuels

    Abstract(#br)Biomass-derived furanyl ethers, such as 5-alkoxymethylfurfurals (AMFs) and 2,5-bis(alkoxymethyl)furans (BAMFs), can be employed as promising biofuels or additives. The development of multifunctional catalysts for the efficient production of furanyl ethers from sugars through 5-hydroxymethylfurfural (HMF) as an intermediate is highly desirable but challenging, because multiple reactions including dehydration, etherification and hydrogenation get involved and the side reaction of sugars and HMF to form humins is inevitable. In this contribution, we found that the introduction of CuO resulted in the generation of Lewis acid sites at the cost of Bronsted acid sites over CuO-USY catalysts through the formation of Al-O-Cu(II) species. The dispersity of CuO particles and the amount of Lewis acid sites could be manipulated by adjusting the loading of CuO. If 5 wt% CuO was supported on USY zeolite to give a CuO(5)-USY catalyst, CuO particles with a high dispersity (36.4%) afforded abundant Lewis acid sites (457.1 μ mol/g). Lewis acid over CuO(5)-USY greatly promoted the acid-catalyzed dehydration of fructose to HMF and HMF etherification to AMFs, resulting in a HMF yield up to 86.2% from fructose and AMFs yields greater than 90% from HMF. Interestingly, a combination of CuO(5)-USY and a small amount of metallic Cu powder was able to offer desirable BAMFs yields by the reductive etherification of HMF under hydrogen atmosphere. As a result, 5-methoxymethylfurfural (MMF) of 79.6% and 2,5-bis(methoxymethyl)furan (BMMF) yield of 74.5% were achieved from fructose through HMF as an intermediate in the presence of CuO(5)-USY alone or with metallic Cu as a co-catalyst. Therefore, the above Cu-based catalyst system holds the promise to flexibly produce a family of AMFs or BAMFs from fructose via a facile two-step approach

    A simulation study on the measurement of D0-D0bar mixing parameter y at BES-III

    We established a method on measuring the \dzdzb mixing parameter yy for BESIII experiment at the BEPCII e+ee^+e^- collider. In this method, the doubly tagged ψ(3770)D0D0\psi(3770) \to D^0 \overline{D^0} events, with one DD decays to CP-eigenstates and the other DD decays semileptonically, are used to reconstruct the signals. Since this analysis requires good e/πe/\pi separation, a likelihood approach, which combines the dE/dxdE/dx, time of flight and the electromagnetic shower detectors information, is used for particle identification. We estimate the sensitivity of the measurement of yy to be 0.007 based on a 20fb120fb^{-1} fully simulated MC sample.Comment: 6 pages, 7 figure
