    Relationship Between Foreign Film Exposure and Ethnocentrism

    This study looked at the relationship between foreign film exposure and ethnocentrism by both considering individual differences factors and motives to watch foreign films, based on the uses and gratifications theoretical model. The individual differences factors include cosmopoliteness, access to foreign films and environmental ethnic diversity. The uses and gratifications theory posits that social and psychological factors influence individuals\u27 motives to use media and ultimately lead to different media effects. Accordingly, this research investigated how individual differences related to motives to view foreign films and how foreign film exposure affected people\u27s attitudes to other cultures. A pilot study was conducted first to check whether there was enough variance in foreign film exposure for American college student viewers, considering the limited release of foreign movies in the American market. Finally an online survey was conducted at an urban college in the US, where 205 undergraduate students participated. The individual difference factors cosmopoliteness and access were found to significantly and positively correlate with motives, while no significant relationship was found between environmental ethnic diversity and motives. These results indicate that individual differences in diversity of communication network and media content, and interests in other cultures, as well as accessibility of foreign films, do influence motivations to watch foreign movies. Also, motives and foreign film exposure were found to be related significantly, suggesting that different uses of foreign films lead to varying levels of exposure. Most interestingly, a negative relationship between foreign film exposure and ethnocentrism was significantly supported by the study. However, additional work needs to examine how foreign film exposure may reduce ethnocentrism. Furthermore, environmental ethnic diversity is not related to ethnocentrism either, suggesting physical integration with ethnic minorities cannot guarantee act

    MicroRNA-140-5p inhibits cellular proliferation, migration and invasion by downregulating AKT/STAT3/NF-κB pathway in breast carcinoma cells

    MicroRNA-140-5p (miR-140-5p) plays a pivotal role in human cancers. However, its role and molecular mechanisms in breast carcinoma are not fully explored. Using miR-140-5p transfected breast cancer cell line MDA-MB-231, several in vitro experiments were performed and described in this paper. They consist of the cell proliferation assay, wound healing assay, transwell assay, colony formation assays and qRTPCR. Expression levels of target proteins were determined using western blotting. In addition, experiments on animal models were performed to study the possible role of miR-140-5p in tumorigenesis of breast carcinoma cells. The induction of experimental breast tumor in mice model was achieved through the incorporation of MDA-MB-231 tumor cells subcutaneously into the middle left side of the mice. The results showed that miR-140-5p up-regulation significantly suppresses proliferation, cellular invasion and migration of breast carcinoma cells. Furthermore, miR-140-5p up-regulation stops breast cancer cells at G0/G1 phase. The results of the animal model indicated that up-regulation of miR-140-5p suppresses its tumorigenic ability. Moreover, we also found that miR-140-5p up-regulation reduces the phosphorylation level of STAT3, p65, and AKT. In addition, miR-140-5p overexpression significantly decreases CDK2 expression while increasing E-cadherin expression level. These data revealed that miR-140-5p suppressed tumor progression of breast carcinoma cells through inhibition of the AKT/STAT3/NF-κB pathway. Taken the present study results together, we can conclude that miR-140-5p may act as a novel target in microRNA-targeting anticancer strategy for the treatment of breast cancer

    High-sensitive and rapid detection of Mycobacterium tuberculosis infection by IFN-γ release assay among HIV-infected individuals in BCG-vaccinated area

    <p>Abstract</p> <p>Background</p> <p>An accurate test for <it>Mycobacterium tuberculosis </it>infection is urgently needed in immunosuppressed populations. The aim of this study was to investigate the diagnostic power of enzyme-linked immunospot (ELISPOT)-based IFN-γ release assay in detecting active and latent tuberculosis in HIV-infected population in <it>bacillus Calmette-Guerin </it>(BCG)-vaccinated area. A total of 100 HIV-infected individuals including 32 active tuberculosis patients were recruited. An ELISPOT-based IFN-γ release assay, T-SPOT.TB, was used to evaluate the <it>M. tuberculosis </it>ESAT-6 and CFP-10 specific IFN-γ response. Tuberculin skin test (TST) was performed for all recruited subjects.</p> <p>Results</p> <p>The subjects were divided into group HIV+ATB (HIV-infected individuals with active tuberculosis, n = 32), group HIV+LTB (HIV-infected individuals with positive results of T-SPOT.TB assay, n = 46) and group HIV only (HIV-infected individuals with negative results of T-SPOT.TB assay and without evidence of tuberculosis infection, n = 22). In group HIV+ATB and HIV+LTB, T-SPOT.TB positive rate in subjects with TST <5 mm were 50% (16/32) and 41.3% (19/46), respectively. Individuals in group HIV+ATB and HIV+LTB with CD4+ T cells <500/μl, T-SPOT.TB showed a higher sensitivity than TST (64.5% vs. 22.6% and 62.2% vs. 29.7%, respectively, both <it>P </it>< 0.0001). In addition, the sensitivity of T-SPOT.TB assay in group HIV+ATB increased to >85% in patients with TB treatment for less than 1 month and CD4+ T cells ≥200/μl, while for patients treated for more than 3 months and CD4+ T cells <200/μl, the sensitivity was decreased to only 33.3%. Furthermore, the results could be generated by T-SPOT.TB assay within 24 hours, which was more rapid than TST with 48–72 hours.</p> <p>Conclusion</p> <p>ELISPOT-based IFN-γ release assay is more sensitive and rapid for the diagnosis of TB infection in Chinese HIV-infected individuals with history of BCG vaccination, and could be an effective tool for guiding preventive treatment with isoniazid in latently infected people and for TB control in China.</p

    GnRHa/Stanozolol Combined Therapy Maintains Normal Bone Growth in Central Precocious Puberty

    BackgroundGonadotropin-releasing hormone agonist (GnRHa) is the gold standard in the treatment of Central Precocious Puberty (CPP) with progressive puberty and accelerative growth. However, GnRHa treatment is reported to result in growth deceleration and prevents growth plate development which leads to a reduction in height velocity. Stanozolol (ST) has been used to stimulate growth in patients with delayed growth and puberty, nevertheless, the effects and mechanisms of ST on CPP with GnRHa treatment are currently unclear.Methods and ResultsIn the current study, we recorded the following vital observations that provided insights into ST induced chondrogenic differentiation and the maintenance of normal growth plate development: (1) ST efficiently prevented growth deceleration and maintained normal growth plate development in rats undergoing GnRHa treatment; (2) ST suppressed the inhibitory effect of GnRHa to promote chondrogenic differentiation; (3) ST induced chondrogenic differentiation through the activation of the JNK/c-Jun/Sox9 signaling pathway; (4) ST promoted chondrogenic differentiation and growth plate development through the JNK/Sox9 signaling pathway in vivo.ConclusionsST mitigated the inhibitory effects of GnRHa and promoted growth plate development in rats. ST induced the differentiation of chondrocytes and maintained normal growth plate development through the activation of JNK/c-Jun/Sox9 signaling. These novel findings indicated that ST could be a potential agent for maintaining normal bone growth in cases of CPP undergoing GnRHa treatment

    Comparison between the electrical conductivities of La0.8Sr0.2Ga0.8Mg0.2O3 and La0.8Sr0.2Ga0.8Mg0.15Co0.05O3

    La0.8Sr0.2Ga0.8Mg0.2O3(LSGM8282) and La0.8Sr0.2Ga0.8Mg0.15Co0.05O3 (LSGMC5) are two kinds of excellent electrolytes for intermediate temperature solid oxide fuel cells (ITSOFCs). LSGM8282 and LSGMC5 were prepared using solid state reaction in this study. Total electrical conductivities and electronic conductivities of LSGM8282 and LSGMC5 were comparatively studied using four-probe AC impedance spectrometry and Hebb-Wagner polarization. The results showed that the total electrical conductivity of LSGM8282 had no dependency on oxygen partial pressures. However, the total electrical conductivity of LSGMC5 increased with the decrease in oxygen partial pressures under high oxygen partial pressures. Within the temperature range of 973 similar to 1173 K, the orders of dependency of the electron conduction and hole conduction of LSGM8282 on the oxygen partial pressure was -1/4 and 1/4, respectively. Within the temperature range of 1073 similar to 1173 K, the orders of dependency of the electron conduction and hole conduction of LSGMC5 on the oxygen portial pressure was -1/4 and about 1/8, respectively. The oxygen ion conductivity of LSGM8282 had no dependency on oxygen partial pressures, while that of LSGMC5 increased with the decrease in oxygen partial pressures under high oxygen partial pressures

    Two Antimycin A Analogues from Marine-Derived Actinomycete Streptomyces lusitanus

    Two new antimycin A analogues, antimycin B1 and B2 (1–2), were isolated from a spent broth of a marine-derived bacterium, Streptomyces lusitanus. The structures of 1 and 2 were established on the basis of spectroscopic analyses and chemical methods. The isolated compounds were tested for their anti-bacterial potency. Compound 1 was found to be inactive against the bacteria Bacillus subtilis, Staphyloccocus aureus, and Loktanella hongkongensis. Compound 2 showed antibacterial activities against S. aureus and L. hongkongensis with MIC values of 32.0 and 8.0 μg/mL, respectively

    CARD11 regulates the thymic Treg development in an NF-κB-independent manner

    IntroductionCARD11 is a lymphoid lineage-specific scaffold protein regulating the NF-κB activation downstream of the antigen receptor signal pathway. Defective CARD11 function results in abnormal development and differentiation of lymphocytes, especially thymic regulatory T cells (Treg).MethodIn this study, we used patients’ samples together with transgenic mouse models carrying pathogenic CARD11 mutations from patients to explore their effects on Treg development. Immunoblotting and a GFP receptor assay were used to evaluate the activation effect of CARD11 mutants on NF-κB signaling. Then the suppressive function of Tregs carrying distinct CARD11 mutations was measured by in vitro suppression assay. Finally, we applied the retroviral transduced bone marrow chimeras to rescue the Treg development in an NF-κB independent manner.Results and discussWe found CARD11 mutations causing hyper-activated NF-κB signals also gave rise to compromised Treg development in the thymus, similar to the phenotype in Card11 deficient mice. This observation challenges the previous view that CARD11 regulates Treg lineage dependent on the NF-kB activation. Mechanistic investigations reveal that the noncanonical function CARD11, which negatively regulates the AKT/ FOXO1 signal pathway, is responsible for regulating Treg generation. Moreover, primary immunodeficiency patients carrying CARD11 mutation, which autonomously activates NF-κB, also represented the reduced Treg population in their peripheral blood. Our results propose a new regulatory function of CARD11 and illuminate an NF-κB independent pathway for thymic Treg lineage commitment

    Validity and Reproducibility of a Revised Semiquantitative Food Frequency Questionnaire (SQFFQ) for Women of Age-group 12-44 Years in Chengdu

    To find a credible nutritional screening tool for evaluating relationship between nutritional status and diseases in Chengdu female residents, the reliability and validity of a revised semi-quantitative food frequency questionnaire (SQFFQ) were tested. The validity was assessed by comparing the SQFFQ with the \u2018standard\u2019 method of 3 days\u2019 dietary recall, and the reliability was assessed by comparing the first SQFFQ with the second SQFFQ at 4 weeks interval. Correlation analysis showed that, for reliability, the average correlation coefficient (CC) of 22 kinds of nutrients was 0.66 and reduced to 0.60 after adjusting for energy; the average of intra-class correlation coefficients (ICC) was 0.65. For validity, the average CC was 0.35 and remained stable after adjusting for CC of energy or nutrients. Validity of 17 nutrients in SQFFQ survey had correlation with result of 3 days\u2019 dietary recall. The results showed that the revised SQFFQ can be used for investigating the role of nutrients in development of disease in Chengdu female residents
