Active Stat3 is required for survival of human squamous cell carcinoma cells in serum-free conditions
BACKGROUND: Squamous cell carcinoma (SCC) of the skin is the most aggressive form of non-melanoma skin cancer (NMSC), and is the single most commonly diagnosed cancer in the U.S., with over one million new cases reported each year. Recent studies have revealed an oncogenic role of activated signal transducer and activator of transcription 3 (Stat3) in many human tumors, especially in those of epithelial origin, including skin SCC. Stat3 is a mediator of numerous growth factor and cytokine signaling pathways, all of which activate it through phosphorylation of tyrosine 705. RESULTS: To further address the role of Stat3 in skin SCC tumorigenesis, we have analyzed a panel of human skin-derived cell lines ranging from normal human epidermal keratinocytes (NHEK), to non-tumorigenic transformed skin cells (HaCaT), to highly tumorigenic cells (SRB1-m7 and SRB12-p9) and observed a positive correlation between Stat3 phosphorylation and SCC malignancy. We next determined the role of Stat3 activity in cell proliferation and viability under serum-free culture conditions. This was accomplished by suppressing Stat3 activity in the SRB12-p9 cells through stable expression of a dominant negative acting form of Stat3β, which contains a tyrosine 705 to phenylalanine mutation (S3DN). The S3DN cells behaved similar to parental SRB12-p9 cells when cultured in optimal growth conditions, in the presence of 10% fetal calf serum. However, unlike the SRB12-p9 cells, S3DN cells underwent apoptotic cell death when cultured in serum-free medium (SFM). This was evidenced by multiple criteria, including accumulation of sub-G1 particles, induced PARP cleavage, and acquisition of the characteristic morphological changes associated with apoptosis. CONCLUSION: This study provides direct evidence for a role for Stat3 in maintaining cell survival in the conditions of exogenous growth factor deprivation produced by culture in SFM. We also propose that delivery of the S3DN gene or protein to tumor cells could induce apoptosis and/or sensitize those cells to the apoptotic effects of cancer therapeutic agents, raising the possibility of using S3DN as an adjunct for treatment of skin SCC
In-situ strain tuning of the Dirac surface states in Bi2Se3 films
Elastic strain has the potential for a controlled manipulation of the band
gap and spin-polarized Dirac states of topological materials, which can lead to
pseudo-magnetic-field effects, helical flat bands and topological phase
transitions. However, practical realization of these exotic phenomena is
challenging and yet to be achieved. Here, we show that the Dirac surface states
of the topological insulator Bi2Se3 can be reversibly tuned by an externally
applied elastic strain. Performing in-situ x-ray diffraction and in-situ
angle-resolved photoemission spectroscopy measurements during tensile testing
of epitaxial Bi2Se3 films bonded onto a flexible substrate, we demonstrate
elastic strains of up to 2.1% and quantify the resulting reversible changes in
the topological surface state. Our study establishes the functional
relationship between the lattice and electronic structures of Bi2Se3 and, more
generally, demonstrates a new route toward momentum-resolved mapping of
strain-induced band structure changes
Presence of antioxidative agent, Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro- in newly isolated Streptomyces mangrovisoli sp. nov.
A novel Streptomyces, strain MUSC 149T was isolated from mangrove soil. A polyphasic approach was used to study the taxonomy of MUSC 149T, which shows a range of phylogenetic and chemotaxonomic properties consistent with those of the members of the genus Streptomyces. The diamino acid of the cell wall peptidoglycan was LL-diaminopimelic acid. The predominant menaquinones were identified as MK9(H8) and MK9(H6). Phylogenetic analysis indicated that closely related strains include Streptomyces rhizophilus NBRC 108885T (99.2 % sequence similarity), Streptomyces gramineus NBRC 107863T (98.7 %) and Streptomyces graminisoli NBRC 108883T (98.5 %). The DNA–DNA relatedness values between MUSC 149T and closely related type strains ranged from 12.4 ± 3.3 % to 27.3 ± 1.9 %. The DNA G + C content was determined to be 72.7 mol%. The extract of MUSC 149T exhibited strong antioxidant activity and chemical analysis reported identification of an antioxidant agent, Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-. These data showed that metabolites of MUSC 149T shall be useful as preventive agent against free-radical associated diseases. Based on the polyphasic study of MUSC 149T, the strain merits assignment to a novel species, for which the name Streptomyces mangrovisoli sp. nov. is proposed. The type strain is MUSC 149T (= MCCC 1K00699T = DSM 100438T)

Integrative conjugative elements of the ICEPan family play a potential role in Pantoea ananatis ecological diversification and antibiosis
Pantoea ananatis is a highly versatile enterobacterium isolated from diverse environmental sources. The ecological diversity of this species may be attributed, in part, to the acquisition of mobile genetic elements. One such element is an Integrative and Conjugative Element (ICE). By means of in silico analyses the ICE elements belonging to a novel family, ICEPan, were identified in the genome sequences of five P. ananatis strains and characterized. PCR screening showed that ICEPan is prevalent among P. ananatis strains isolated from different environmental sources and geographic locations. Members of the ICEPan family share a common origin with ICEs of other enterobacteria, as well as conjugative plasmids of Erwinia spp. Aside from core modules for ICEPan integration, maintenance and dissemination, the ICEPan contain extensive non-conserved islands coding for proteins that may contribute toward various phenotypes such as stress response and antibiosis, and the highly diverse ICEPan thus plays a major role in the diversification of P. ananatis. An island is furthermore integrated within an ICEPan DNA repair-encoding locus umuDC and we postulate its role in stress-induced dissemination and/or expression of the genes on this island
Small Conductance Calcium-Activated Potassium Current is Activated During Hypokalemia and Masks Short Term Cardiac Memory Induced by Ventricular Pacing.
Background: Hypokalemia increases the vulnerability to ventricular fibrillation (VF). We hypothesize that the apamin-sensitive small conductance calcium-activated potassium current (IKAS) is activated during hypokalemia and that IKAS blockade is proarrhythmic. Methods and Results: Optical mapping was performed in 23 Langendorff perfused rabbit ventricles with atrioventricular block and either right ventricular (RV) or left ventricular (LV) pacing during normokalemia or hypokalemia. Apamin prolonged the action potential duration (APD) measured to 80% repolarization (APD80) by 26 ms [95% confidence interval, CI, 14–37] during normokalemia and by 54 ms [CI, 40 to 68] during hypokalemia (P=0.01) at 1000 ms pacing cycle length (PCL). In hypokalemic ventricles, apamin increased the maximal slope of APD restitution, the PCL threshold of APD alternans, the PCL for wavebreak induction and the area of spatially discordant APD alternans. Apamin significantly facilitated the induction of sustained VF (from 3/9 hearts to 9/9 hearts, P=0.009). Short term cardiac memory was assessed by the slope of APD80 versus activation time. The slope increased from 0.01 [CI, −0.09 to 0.12] at baseline to 0.34 [CI, 0.23 to 0.44] after apamin (P<0.001) during RV pacing, and from 0.07 [CI, −0.05 to 0.20] to 0.54 [CI, 0.06 to 1.03] after apamin infusion (P=0.045) during LV pacing. Patch-clamp studies confirmed increased IKASin isolated rabbit ventricular myocytes during hypokalemia (P=0.038). Conclusions: Hypokalemia activates IKAS to shorten APD and maintain repolarization reserve at late activation sites during ventricular pacing. IKAS blockade prominently lengthens the APD at late activation sites and facilitates VF induction
Streptomyces antioxidans sp. nov., a Novel Mangrove Soil Actinobacterium with Antioxidative and Neuroprotective Potentials
A novel strain, Streptomyces antioxidans MUSC 164(T) was recovered from mangrove forest soil located at Tanjung Lumpur, Malaysia. The Gram-positive bacterium forms yellowish-white aerial and brilliant greenish yellow substrate mycelium on ISP 2 agar. A polyphasic approach was used to determine the taxonomy status of strain MUSC 164(T). The strain showed a spectrum of phylogenetic and chemotaxonomic properties consistent with those of the members of the genus Streptomyces. The cell wall peptidoglycan was determined to contain LL-diaminopimelic acid. The predominant menaquinones were identified as MK-9(H(6)) and MK-9(H(8)), while the identified polar lipids consisted of aminolipid, diphosphatidylglycerol, glycolipid, hydroxyphosphatidylethanolamine, phospholipid, phosphatidylinositol, phosphatidylethanolamine, phosphatidylglycerol and lipid. The cell wall sugars consist of galactose, glucose and ribose. The predominant cellular fatty acids (>10.0%) were identified as iso-C(15:)(0) (34.8%) and anteiso-C(15:)(0)(14.0%). Phylogenetic analysis identified that closely related strains for MUSC 164(T) as Streptomyces javensis NBRC 100777(T) (99.6% sequence similarity), Streptomyces yogyakartensis NBRC 100779(T) (99.6%) and Streptomyces violaceusniger NBRC 13459(T) (99.6%). The DNA–DNA relatedness values between MUSC 164(T) and closely related type strains ranged from 23.8 ± 0.3% to 53.1 ± 4.3%. BOX-PCR fingerprints comparison showed that MUSC 164(T) exhibits a unique DNA profile, with DNA G + C content determined to be 71.6 mol%. Based on the polyphasic study of MUSC 164(T), it is concluded that this strain represents a novel species, for which the name Streptomyces antioxidans sp. nov. is proposed. The type strain is MUSC 164(T) (=DSM 101523(T) = MCCC 1K01590(T)). The extract of MUSC 164(T) showed potent antioxidative and neuroprotective activities against hydrogen peroxide. The chemical analysis of the extract revealed that the strain produces pyrazines and phenolic-related compounds that could explain for the observed bioactivities
Gene expression regulation in the plant growth promoting Bacillus atrophaeus UCMB-5137 stimulated by maize root exudates
Despite successful use of Plant Growth Promoting Rhizobacteria (PGPR) in agriculture, little is known about
specific mechanisms of gene regulation facilitating the effective communication between bacteria and plants during
plant colonization. Active PGPR strain B. atrophaeus UCMB-5137 was studied in this research. RNA sequencing
profiles were generated in experiments where root exudate stimulations were used to mimic interactions between
bacteria and plants. It was found that the gene regulation in B. atrophaeus UCMB-5137 in response to the root
exudate stimuli differed from the reported gene regulation at similar conditions in B. amyloliquefaciens FZB42,
which was considered as a paradigm PGPR. This difference was explained by hypersensitivity of UCMB-5137 to
the root exudate stimuli impelling it to a sessile root colonization behavior through the CcpA-CodY-AbrB
regulation. It was found that the transcriptional factor DegU also could play an important role in gene regulations
during plant colonization. A significant stress caused by the root exudates on in vitro cultivated B. atrophaeus
UCMB-5137 was noticed and discussed. Multiple cases of conflicted gene regulations showed scantiness of our
knowledge on the regulatory network in Bacillus. Some of these conflicted regulations could be explained by
interference of non-coding RNA (ncRNA). Search through differential expressed intergenic regions revealed 49
putative loci of ncRNA regulated by the root exudate stimuli. Possible target mRNA were predicted and a general

Machine-Part cell formation through visual decipherable clustering of Self Organizing Map
Machine-part cell formation is used in cellular manufacturing in order to
process a large variety, quality, lower work in process levels, reducing
manufacturing lead-time and customer response time while retaining flexibility
for new products. This paper presents a new and novel approach for obtaining
machine cells and part families. In the cellular manufacturing the fundamental
problem is the formation of part families and machine cells. The present paper
deals with the Self Organising Map (SOM) method an unsupervised learning
algorithm in Artificial Intelligence, and has been used as a visually
decipherable clustering tool of machine-part cell formation. The objective of
the paper is to cluster the binary machine-part matrix through visually
decipherable cluster of SOM color-coding and labelling via the SOM map nodes in
such a way that the part families are processed in that machine cells. The
Umatrix, component plane, principal component projection, scatter plot and
histogram of SOM have been reported in the present work for the successful
visualization of the machine-part cell formation. Computational result with the
proposed algorithm on a set of group technology problems available in the
literature is also presented. The proposed SOM approach produced solutions with
a grouping efficacy that is at least as good as any results earlier reported in
the literature and improved the grouping efficacy for 70% of the problems and
