    The Status of Pro-social Tendency of Left-Behind Adolescents in China: How Family Function and Self-Esteem Affect Pro-social Tendencies

    In China, adolescents are frequently left behind. To date, few studies have focused on the pro-social tendencies of left-behind adolescents and the relationship of family function, self-esteem, and pro-social tendency is yet to be examined. This study, therefore, aims to understand the status of pro-social tendency of left-behind adolescents and to explore the mediating and moderating roles of self-esteem in the relationship between family function and pro-social tendency. A large, school-based survey was conducted in three Chinese provinces. An analysis of covariance was first used to identify the differences in pro-social tendency between adolescents who were and were not left behind. We then analyzed the variance within left-behind adolescents using demographics, left-behind type, years of being left-behind, and caregiver related characteristics. A structural equation model was used to analyze the relationship of family function, self-esteem, and pro-social tendency, with bootstrapping used to explore the mediating role of self-esteem. Additionally, an ordinary least squares regression was used to examine the moderating effect of self-esteem. The results showed that the pro-social tendency of left-behind adolescents was lower than in non-left-behind adolescents (F = 15.11, p = 0.0001). Family function was positive related to pro-social tendency (r = 0.259), which had not only a direct effect on pro-social tendency (β = 0.254), but also an indirect effect through self-esteem (β = 0.071, bias-corrected 95% CI: 0.051:0.090; percentile 95% CI: 0.053:0.092). Additionally, 21.85% of the total effect of family function on pro-social tendency was mediated by self-esteem. Furthermore, self-esteem negatively moderated the relationship between family function and pro-social tendency (β = -0.208, p < 0.0001), such that the effect of family function on pro-social tendency became weaker as self-esteem increased. The current study verified the negative effect of being left behind on the social development of adolescents and contributed to the understanding of the importance of self-esteem in the relationship between family function and pro-social tendency. Interventions aimed at enhancing self-esteem should be developed and implemented in left-behind adolescents to promote wellness in the entirety of psychological and social outcomes

    Free energy landscape for the binding process of Huperzine A to acetylcholinesterase

    Drug-target residence time (t = 1/koff, where koff is the dissociation rate constant) has become an important index in discovering betteror best-in-class drugs. However, little effort has been dedicated to developing computational methods that can accurately predict this kinetic parameter or related parameters, koff and activation free energy of dissociation (ΔG≠ off). In this paper, energy landscape theory that has been developed to understand protein folding and function is extended to develop a generally applicable computational framework that is able to construct a complete ligand-target binding free energy landscape. This enables both the binding affinity and the binding kinetics to be accurately estimated.We applied this method to simulate the binding event of the anti-Alzheimer’s disease drug (−)−Huperzine A to its target acetylcholinesterase (AChE). The computational results are in excellent agreement with our concurrent experimental measurements. All of the predicted values of binding free energy and activation free energies of association and dissociation deviate from the experimental data only by less than 1 kcal/ mol. The method also provides atomic resolution information for the (−)−Huperzine A binding pathway, which may be useful in designing more potent AChE inhibitors. We expect thismethodology to be widely applicable to drug discovery and development

    Review and development of surrounding rock control technology for gob-side entry retaining in China

    The research and application of gob-side entry retaining (GER) technology in Chinese coal mines has been over 60 years. Two major technical types including GER with filling and GER with roof cutting were developed. However, due to the complex mining and geological conditions of the mined-out coal seam in different mining areas, as well as the strong mining pressure behaviours in the retained roadway, the promotion and application of GER technology presented ups and downs. Firstly, the achievements and key technological advances in the main GER types in China, the principle of surrounding rock stability, road-in support technology, roadside support technology, adaptability evaluation and surrounding rock stability monitoring are summarized, and the application applicability of different technologies at this stage is analyzed. Then, the difficulties and challenges faced by current GER technology are summarized: there is still no systematic theory for GER with strong mining pressure; there are still shortcomings in the theoretical understanding of the interaction mechanism between the surrounding rock and the support body for GER with filling; the mechanical and deformation characteristics of the filling materials for the roadside support body are not yet suitable for GER in deep working faces or with strong mining pressure; the mechanism and control technology of floor heave for GER are not yet perfect; the research on stability control of filling body for GER under strong dynamic load or rock burst is still in a blank. Finally, concerned with such difficulties and challenges, several reserve technologies have been proposed: the coordinated control of controlled roof cutting and filling for GER in fully mechanized caving/ full-seam mining in the thick coal seam, and the GER technology with additive modified high-water materials in working faces with strong mining pressure; finally, a set of intelligent inversion workflow for rock mechanical parameters used in GER numerical simulations for GER is established, and an intelligent optimization design method for GER support parameters is proposed

    Antibiotic affects the gut microbiota composition and expression of genes related to lipid metabolism and myofiber types in skeletal muscle of piglets

    Background: Early-life antibiotic administration is known to affect gut microbiota and host adiposity, but the effects of antibiotic exposure on skeletal muscle properties remain unknown. The present study evaluated the changes in skeletal muscle properties including myofiber characteristics and composition, as well as intramuscular fat (IMF) content in skeletal muscle of piglets when exposed to a tylosin-containing diet. Results: A total of 18 piglets (28 days of age) were randomly allocated into two groups: control basal diet (Control) and Control + 100 mg tylosin phosphate/kg of feed (Antibiotic). The trial lasted for 39 days. High-throughput amplicon sequencing revealed that no significant difference in initial gut microbiota composition was existed between Control and Antibiotic groups. Antibiotic administration increased body weight and growth rate and decreased feed to gain ratio of pigs (P < 0.05). The carcass lean and fat volumes of pigs were increased by the tylosin administration (P < 0.05). Antibiotic treatment increased myofiber density and the expression of genes related to type I and type IIb myofibers in longissimus muscle (P < 0.05). The IMF content in longissimus muscle was increased by antibiotic exposure (P < 0.05). Antibiotic administration increased expression of genes related to fatty acid uptake and de novo synthesis, and decreased expression of genes related to triglyceride hydrolysis (P < 0.05). Tylosin administration affected taxonomic distribution and beta diversity of the caecal and colonic microbiota of piglets. Conclusion: These results confirm that the growth performance, myofiber composition and muscle lipid metabolism are affected by antibiotic administration, which may be associated with an altered gut microbiota, suggesting that the gut microbiota could be served as a potential target for modulating skeletal muscle properties of host

    Short-chain fructo-oligosaccharides supplementation to suckling piglets : assessment of pre- and post-weaning performance and gut health

    Farmers face difficulties in redeeming their investment in larger litter sizes since this comes with larger litter heterogenicity, lower litter resilience and risk of higher mortality. Dietary oligosaccharides, given to the sow, proved beneficial for the offspring's performance. However, giving oligosaccharides to the suckling piglet is poorly explored. Therefore, this field trial studied the effect of dietary short-chain fructo-oligosaccharides (scFOS; 1g/day; drenched) supplementation to low (LBW, lower quartile), normal (NBW, two intermediate quartiles) and high (HBW, upper quartile) birth weight piglets from birth until 7 or 21 days of age. Performance parameters, gut microbiome and short-chain fatty acids profile of feces and digesta were assessed at birth (d 0), d 7, weaning (d 21.5) and 2 weeks post-weaning (d 36.5). Additional parameters reflecting gut health (intestinal integrity and morphology, mucosal immune system) were analysed at d 36.5. Most parameters changed with age or differed with the piglet's birth weight. Drenching with scFOS increased body weight by 1 kg in NBW suckling piglets and reduced the post-weaning mortality rate by a 100%. No clear difference in the IgG level, the microbiota composition and fermentative activity between the treatment groups was observed. Additionnally, intestinal integrity, determined by measuring intestinal permeability and regenerative capacity, was similar between the treatment groups. Also, intestinal architecture (villus lenght, crypt depth) was not affected by scFOS supplementation. The density of intra-epithelial lymphocytes and the expression profiles (real-time qPCR) for immune system-related genes (IL-10, IL-1ss, IL-6, TNF alpha and IFN gamma) were used to assess mucosal immunity. Only IFN gamma expression, was upregulated in piglets that received scFOS for 7 days. The improved body weight and the reduced post-weaning mortality seen in piglets supplemented with scFOS support the view that scFOS positively impact piglet's health and resilience. However, the modes of action for these effects are not yet fully elucidated and its potential to improve other performance parameters needs further investigation

    Meta-analysis of 8q24 for seven cancers reveals a locus between NOV and ENPP2 associated with cancer development.

    BACKGROUND: Human chromosomal region 8q24 contains several genes which could be functionally related to cancer, including the proto-oncogene c-MYC. However, the abundance of associations around 128 Mb on chromosome 8 could mask the appearance of a weaker, but important, association elsewhere on 8q24. METHODS: In this study, we completed a meta-analysis of results from nine genome-wide association studies for seven types of solid-tumor cancers (breast, prostate, pancreatic, lung, ovarian, colon, and glioma) to identify additional associations that were not apparent in any individual study. RESULTS: Fifteen SNPs in the 8q24 region had meta-analysis p-values < 1E-04. In particular, the region consisting of 120,576,000-120,627,000 bp contained 7 SNPs with p-values < 1.0E-4, including rs6993464 (p = 1.25E-07). This association lies in the region between two genes, NOV and ENPP2, which have been shown to play a role in tumor development and motility. An additional region consisting of 5 markers from 128,478,000 bp - 128,524,000 (around gene POU5F1B) had p-values < 1E-04, including rs6983267, which had the smallest p-value (p = 6.34E-08). This result replicates previous reports of association between rs6983267 and prostate and colon cancer. CONCLUSIONS: Further research in this area is warranted as these results demonstrate that the chromosomal region 8q24 may contain a locus that influences general cancer susceptibility between 120,576 and 120,630 kb.RIGHTS : This article is licensed under the BioMed Central licence at http://www.biomedcentral.com/about/license which is similar to the 'Creative Commons Attribution Licence'.
