    From symplectic cohomology to Lagrangian enumerative geometry

    We build a bridge between Floer theory on open symplectic manifolds and the enumerative geometry of holomorphic disks inside their Fano compactifications, by detecting elements in symplectic cohomology which are mirror to Landau-Ginzburg potentials. We also treat the higher Maslov index versions of LG potentials. We discover a relation between higher disk potentials and symplectic cohomology rings of anticanonical divisor complements (themselves related to closed-string Gromov-Witten invariants), and explore several other applications to the geometry of Liouville domains.Comment: 47 pages, 13 figures; v2: reference fixes, minor correction

    Construction of Integrated Genetic Map between Various Existing DNA Markers and Newly Developed P450-related PBA Markers in Diploid Potato (Solanum tuberosum)

    We have generated estimates of the genomic distribution of newly developed molecular markers, which are P450-based analogues (PBAs), in diploid potato (Solanum tuberosum, 2n=2x=24) based on functional genomics. A total of 401 markers, including 111 SSR, 33 RFLP, 87 RFLP-STS, 45 CAPS, 94 RAPD, 15 PBA, 9 AFLP, 3 RGL and 4 ISSR markers, were employed in a parental polymorphism survey. A total of 127 out of 401 markers (172 loci) displayed polymorphisms between parents and were confirmed to show segregation in partial progenies. These selected 127 markers were then tested for their possible use for a whole mapping population. The subsequent results of genetic mapping of the PBAs revealed that they were distributed on at least 8 chromosomes, suggesting that they have a significant potential not only as tools for assessing genetic diversity but also as effective markers to construct more detailed genetic maps of potato, in conjunction with existing identified genetic loci. To integrate the information from existing maps into our mapping study, we performed a comparative analysis between two representative maps (RFLP and SSR-based) with our PBA map. Based on the subsequent results, we predict that our map will be useful as a bridge between the existing genetic maps of potato and will enable to integrate information about different markers

    Comparison of energy metabolism in Insulin-Dependent and Non-Insulin-Dependent diabetes mellitus

    To compare the metabolic consequences of insulin-dependent diabetes mellitus (IDDM) and non-insulin-dependent diabetes mellitus (NIDDM), glycemic control and energy metabolism were evaluated in 18 children displaying IDDM and 19 NIDDM adult patients. With rising concentrations of fasting blood glucose (FBG), hemoglobin A1C and free fatty acid, the percentage of the ratio of resting energy expenditure (REE) to predicted REE expressed as %REE increased and the respiratory quotient (RQ) decreased. The linear regression between RQ and FBG showed the same gradient in IDDM and NIDDM although the RQ in IDDM was always 0.07 lower than that in NIDDM given various FBG concentrations. Those patients whose RQ values were less than 0.7, indicating ketone body production, included 8 (44%) IDDM and 2 (11%) NIDDM patients. These results may explain the relatively greater manifestation of ketoacidosis in IDDM

    Use of Rice SSR Markers as RAPD Markers for Genetic Diversity Analysis in Zingiberaceae

    Species of Zingiberaceae display a diversity in habitat, ethnobotanical use and morphology. However, little is known about the genetic relationships among taxa and genetic diversity, primarily due to the lack of suitable molecular markers. We tested the cross-amplification potential of microsatellite markers among taxa to identify a larger number of genetic markers. To assess the applicability of rice microsatellite markers to the Zingiberaceae, we tested 12 microsatellite markers for 14 genotypes from three genera of this family: Zingiber, Alpinia and Curcuma. The origin of the genotypes was diverse, covering eight Asian countries. Four microsatellite primer sets failed to amplify fragments in all genotypes studied, whereas the other primer sets amplified all the genotypes. Among the 141 bands, that could be scored, 140 (99.5%) were polymorphic. On the average, each microsatellite primer set amplified 17.6 DNA fragments. In general, amplified fragments were larger than the original rice fragments including the microsatellite region, although in some cases, the amplified bands were similar in size. Though sequence analysis of these bands confirmed the absence of target repeat motif, amplification of a large number of polymorphic bands provided the basis to perform an analysis of genetic diversity. Primers could generate enough polymorphism for possible use in diversity studies, based on provisional multivariate analyses such as cluster analysis and principal component analysis (PCA). The whole set of genotypes based on molecular data was classified into four clusters after cluster analysis. Genotypes from the Curcuma and Alpinia genera were grouped into clusters I and II, respectively. Clusters III and IV comprised genotypes from the genus Zingiber. PCA led to a similar classification. The high polymorphism documented in the present study indicated that the rice microsatellite primers were useful for genetic diversity studies among genera in the family Zingiberaceae

    Hypocretin/orexin and nociceptin/orphanin FQ coordinately regulate analgesia in a mouse model of stress-induced analgesia

    金沢大学医薬保健研究域医学系Stress-induced analgesia (SIA) is a key component of the defensive behavioral "fight-or-flight" response. Although the neural substrates of SIA are incompletely understood, previous studies have implicated the hypocretin/orexin (Hcrt) and nociceptin/orphanin FQ (N/OFQ) peptidergic systems in the regulation of SIA. Using immunohistochemistry in brain tissue from wild-type mice, we identified N/OFQ-containing fibers forming synaptic contacts with Hcrt neurons at both the light and electron microscopic levels. Patch clamp recordings in GFP-tagged mouse Hcrt neurons revealed that N/OFQ hyperpolarized, decreased input resistance, and blocked the firing of action potentials in Hcrt neurons. N/OFQ postsynaptic effects were consistent with opening of a G protein-regulated inwardly rectifying K+ (GIRK) channel. N/OFQ also modulated presynaptic release of GABA and glutamate onto Hcrt neurons in mouse hypothalamic slices. Orexin/ataxin-3 mice, in which the Hcrt neurons degenerate, did not exhibit SIA, although analgesia was induced by i.c.v. administration of Hcrt-1. N/OFQ blocked SIA in wild-type mice, while coadministration of Hcrt-1 overcame N/OFQ inhibition of SIA. These results establish what is, to our knowledge, a novel interaction between the N/OFQ and Hcrt systems in which the corticotropin-releasing factor and N/OFQ systems coordinately modulate the Hcrt neurons to regulate SIA

    Japanese Lung Cancer Society Guidelines for Stage IV NSCLC With EGFR Mutations

    Patients with NSCLC in East Asia, including Japan, frequently contain EGFR mutations. In 2018, we published the latest full clinical practice guidelines on the basis of those provided by the Japanese Lung Cancer Society Guidelines Committee. The purpose of this study was to update those recommendations, especially for the treatment of metastatic or recurrent EGFR-mutated NSCLC. We conducted a literature search of systematic reviews of randomized controlled and nonrandomized trials published between 2018 and 2019 that multiple physicians had reviewed independently. On the basis of those studies and the advice from the Japanese Society of Lung Cancer Expert Panel, we developed updated guidelines according to the Grading of Recommendations, Assessment, Development, and Evaluation system. We also evaluated the benefits of overall and progression-free survival, end points, toxicities, and patients’ reported outcomes. For patients with NSCLC harboring EGFR-activating mutations, the use of EGFR tyrosine kinase inhibitors (EGFR TKIs), especially osimertinib, had the best recommendation as to first-line treatment. We also recommended the combination of EGFR TKI with other agents (platinum-based chemotherapy or antiangiogenic agents); however, it can lead to toxicity. In the presence of EGFR uncommon mutations, except for an exon 20 insertion, we also recommended the EGFR TKI treatment. However, we could not provide recommendations for the treatment of EGFR mutations with immune checkpoint inhibitors, including monotherapy, and its combination with cytotoxic chemotherapy, because of the limited evidence present in the literature. The 2020 Japanese Lung Cancer Society Guidelines can help community-based physicians to determine the most appropriate treatments and adequately provide medical care to their patients

    Hospital and clinic cooperation for the treatment of rheumatoid arthritis in Okayama Prefecture, Japan

    Objective: To survey the current status and problems of cooperation between clinics and hospitals in Okayama Prefecture, Japan for the treatment of rheumatoid arthritis (RA).  Methods: We distributed a questionnaire to 300 of the 983 Okayama Prefecture clinics that had either an internal medicine or orthopedic surgery department, from December 2013 to February 2014. The questionnaire covered practice pattern for RA treatment in clinics, current status of the hospital and clinic cooperation, and acceptance of the biologic therapy.  Results: One hundred clinics responded to the questionnaire. Seventy percent of the clinics reported making referrals to rheumatologists before the initiation of RA treatment, and half of the other 30% of the clinics administered methotrexate as the first-line treatment for RA by their own decision. Sixty-six clinics cooperated with flagship hospitals, conducting medical and laboratory examinations, providing prescriptions, and treating common diseases of patients. These clinics expected the cooperating rheumatologists to follow-up patients every 3 to 6 months and to make the diagnosis, make decisions regarding RA treatment changes, and perform surgery. Seventy-one percent of the clinics responded that cooperation with a hospital is possible even for patients who are administered biologics. As reasons for no cooperation with the flagship hospitals, clinics noted the lack of information about rheumatologists in the area and recent trends in the management of RA.  Conclusion: The current study reported, for the first time, the actual conditions of management of RA in clinics, as well as future problems of hospital and clinic cooperation in Okayama Prefecture

    看護基礎教育に関わる教員および実習指導者の意識

    本研究の目的は、看護基礎教育において協働する大学と実習施設の双方向からの実習指導を通した看護教育への意識を調査し、臨地実習での学生をどのように捉え、どのように教育を行っているのかを明らかにすることである。調査対象はA大学看護学部専任教員およびA大学の実習の一部を受け入れている実習施設であるB病院にて学生の実習指導を担当している看護師とし、自作の自記式アンケート調査を実施した。調査内容は、どのような看護師になってほしいと思って教育しているか、指導の中で意識していることなどである。10名の教員および52名の看護師から回答を得た。その内容を質的帰納的方法で分析した結果、教育の方向性に大きな違いはなく、それぞれに学生の様子および特徴を捉え、教育において学生を尊重している様子が伺えた
