    Direct Measurement Of Thicknesses, Volumes Or Compositions Of Nanomaterials By Quantitative Atomic Number Contrast In High-angle Annular Dark-field Scanning Transmission Electron Microscopy

    The sizes, shapes, volumes and compositions of nanoparticles are very important parameters determining many of their properties. Efforts to measure these parameters for individual nanoparticles and to obtain reliable statistics for a large number of nanoparticles require a fast and reliable method for 3-D characterization. In this dissertation, a direct measurement method for thicknesses, volumes or compositions of nanomaterials by quantitative atomic number contrast in High-Angle Annular Dark-Field (HAADF) Scanning Transmission Electron Microscopy (STEM) is presented. A HAADF detector collects electrons scattered incoherently to high angles. The HAADF signal intensity is in first-order approximation proportional to the sample thickness and increases with atomic number. However, for larger sample thicknesses this approach fails. A simple description for the thickness dependence of the HAADFSTEM contrast has been developed in this dissertation. A new method for the calibration of the sensitivity of the HAADF detector for a FEI F30 transmission electron microscope (TEM) is developed in this dissertation. A nearly linear relationship of the HAADF signal with the electron current is confirmed. Cross sections of multilayered samples provided by TriQuint Semiconductors in Apopka, FL, for contrast calibration were obtained by focused ion-beam (FIB) preparation yielding data on the interaction cross section per atom. iv To obtain an absolute intensity calibration of the HAADF-STEM intensity, Convergent Beam Electron Diffraction (CBED) was performed on Si single crystals. However, for samples prepared by the focused ion beam technique, CBED often significantly underestimates the sample thickness. Multislice simulations from Dr. Kirkland’s C codes are used for comparison with experimental results. TEM offers high lateral resolution, but contains little or no information on the thickness of samples. Thickness maps in energy-filtered TEM (EFTEM), CBED and tilt series are so far the only methods to determine thicknesses of particles in TEM. In this work I have introduced the use of wedge-shaped multilayer samples prepared by FIB for the calibration of HAADF-STEM contrasts. This method yields quantitative contrast data as a function of sample thickness. A database with several pure elements and compounds has been compiled, containing experimental data on the fraction of electrons scattered onto the HAADF detector for each nanometer of sample thickness. The use of thick samples reveals an increased signal at the interfaces of high- and low-density materials. This effect can be explained by the transfer of scattered electrons from the high density material across the interface into the less-absorbing low-density material. The calibrations were used to determine concentration gradients in nanoscale Fe-Pt multilayers as well as thicknesses and volumes of individual Au-Fe, Pt, and Ag nanoparticles. Volumes of nanoparticles with known composition can be determined with accuracy better than 15%.     Longitudinal trends in prostate cancer incidence, mortality, and survival of patients from two Shanghai city districts: a retrospective population-based cohort study, 2000-2009.

    BackgroundProstate cancer is the fifth most common cancer affecting men of all ages in China, but robust surveillance data on its occurrence and outcome is lacking. The specific objective of this retrospective study was to analyze the longitudinal trends of prostate cancer incidence, mortality, and survival in Shanghai from 2000 to 2009.MethodsA retrospective population-based cohort study was performed using data from a central district (Putuo) and a suburban district (Jiading) of Shanghai. Records of all prostate cancer cases reported to the Shanghai Cancer Registry from 2000 to 2009 for the two districts were reviewed. Prostate cancer outcomes were ascertained by matching cases with individual mortality data (up to 2010) from the National Death Register. The Cox proportional hazards model was used to analyze factors associated with prostate cancer survival.ResultsA total of 1022 prostate cancer cases were diagnosed from 2000 to 2009. The average age of patients was 75 years. A rapid increase in incidence occurred during the study period. Compared with the year 2000, 2009 incidence was 3.28 times higher in Putuo and 5.33 times higher in Jiading. Prostate cancer mortality declined from 4.45 per 105 individuals per year in 2000 to 1.94 per 105 in 2009 in Putuo and from 5.45 per 105 to 3.5 per 105 in Jiading during the same period. One-year and 5-year prostate cancer survival rates were 95% and 56% in Putuo, and 88% and 51% in Jiading, respectively. Staging of disease, Karnofsky Performance Scale Index, and selection of chemotherapy were three independent factors influencing the survival of prostate cancer patients.ConclusionsThe prostate cancer incidence increased rapidly from 2000 to 2009, and prostate cancer survival rates decreased in urban and suburban Chinese populations. Early detection and prompt prostate cancer treatment is important for improving health and for increasing survival rates of the Shanghai male population

We show that τ(Spn)=pn−1(p−2)\tau(S_p^n)=p^{n-1}(p-2) for the Sierpi\'{n}ski graph SpnS_p^n with p≥2p\geq 2 and n≥1n\geq 1. The generalized Sierpi\'{n}ski triangle graph Spn^\hat{S_p^n} is obtained by contracting all non-clique edges from the Sierpi\'{n}ski graph Spn+1S_p^{n+1}.     MicroRNA expression profiling in human acute organophosphorus poisoning and functional analysis of dysregulated miRNAs

    Objective: Acute organophosphorus(OP) pesticide poisoning is associated with dysfunctions in multiple organs, especially skeletal muscles, the nervous system and the heart. However, little is known about the specific microRNA (miRNA) changes that control the pathophysiological processes of acute OP poisoning damage. We aimed to explore miRNA expression profiles and gain insight into molecular mechanisms of OP toxic effects.Methods: MicroRNA expression was analyzed by TaqMan Human MicroRNA Array analysis and subsequent validated with quantitive PCR. The targets of the significantly different miRNAs were predicted with miRNA prediction databases, and pathway analysis of the predicted target genes was performed using bioinformatics resources.Results: 37 miRNAs were significantly different in the sera of poisoned patients compared to the healthy controls, including 29 miRNAs that were up-regulated and 8 miRNAs that were down-regulated. Functional analysis indicated that many pathways potentially regulated by these miRNAs are involved in skeletal muscle, nervous system and heart disorders.Conclusion: This study mapped changes in the serum miRNA expression profiles of poisoning patients and predicted functional links between miRNAs and their target genes in the regulation of acute OP poisoning. Our findings are an important resource for further understanding the role of these miRNAs in the regulation of OP-induced injury.Keywords: MicroRNA, expression profiles, human, acute organophosphorus pesticide poisoning, signaling pathways
