    Local Mammary Glucose Supply Regulates Availability and Intracellular Metabolic Pathways of Glucose in the Mammary Gland of Lactating Dairy Goats Under Malnutrition of Energy

    As glucose is the regulator of both the milk yield and mammary oxidative status, glucose supply is considered to play important nutritional and physiological role on mammary gland (MG) metabolism. However, inconsistent results were observed from different infusion methods to evaluate the effect of glucose on MG glucose metabolism. Thus, precise method should be developed to learn how availability and intracellular metabolic pathways of glucose in the MG are altered by the direct mammary glucose supply. In addition, limited information is available on the role of mammary glucose supply in milk synthesis in lactating ruminants under an energy-deficient diet. Direct glucose supply to the MG was implemented in the current study through the external pudendal artery infusion under an energy-deficient diet. Six doses of glucose (0, 20, 40, 60, 80, and 100 g/d) were infused through the external pudendal arteries, which is the main artery to the MG, to six lactating goats fed with basal diet meeting 81% energy requirement in a 6 × 6 Latin square design. Milk and lactose yields were both quadratically increased with increased glucose infusion, whereas the milk yield changed inconsistently with the increased energy balance (EB), indicating local glucose supply, rather than EB, improved milk production. Glucose fluxes in the MG were significantly increased and correlated with mammary plasma flow. However, the ratio of lactose yield to glucose absorbed by the MG was significantly decreased. The increased glucose fluxes in the MG and changed glucose-related metabolites in milk indicated that the glucose availability and intracellular metabolic pathways was regulated by local mammary glucose. Acute glycolysis consumed the superfluous glucose and induced accumulation of oxygen radicals in the MG during over-supplied glucose conditions.     Polarization-sensitive optical projection tomography for muscle fiber imaging

    Optical projection tomography (OPT) is a tool used for three-dimensional imaging of millimeter-scale biological samples, with the advantage of exhibiting isotropic resolution typically in the micron range. OPT can be divided into two types: transmission OPT (tOPT) and emission OPT (eOPT). Compared with eOPT, tOPT discriminates different tissues based on their absorption coefficient, either intrinsic or after specific staining. However, it fails to distinguish muscle fibers whose absorption coefficients are similar to surrounding tissues. To circumvent this problem, in this article we demonstrate a polarization sensitive OPT system which improves the detection and 3D imaging of muscle fibers by using polarized light. We also developed image acquisition and processing protocols that, together with the system, enable the clear visualization of muscles. Experimental results show that the muscle fibers of diaphragm and stomach, difficult to be distinguished in regular tOPT, were clearly displayed in our system, proving its potential use. Moreover, polarization sensitive OPT was fused with tOPT to investigate the stomach tissue comprehensively.     Molecules in the peculiar age-defying source IRAS 19312+1950

    Context. IRAS 19312+1950 is an isolated infrared source that exhibits a characteristic quasi-point-symmetric morphology in the near- and mid-infrared images and is also very bright in molecular radio lines. Because of its unique observational characteristics, various observational studies have been conducted and several hypotheses have been proposed regarding its origin, which is still unclear. So far, it has been suggested that it could be a peculiar evolved star, a young stellar object, or even a red nova remnant. Regardless of which type of object it is ultimately classified as, IRAS 19312+1950 is exceptionally bright in the infrared and molecular radio lines and therefore will undoubtedly be crucial as a prototype of this kind of object having a peculiar nature or unusual evolutionary phase. Aims. This study aims to reveal the molecular composition of the central part of IRAS 19312+1950 by performing an unbiased molecular radio line survey and discussing the origin of the object from a molecular chemical point of view. Methods. We carried out a spectral line survey with the IRAM 30 m telescope towards the center of IRAS 19312+1950 in the 3 and 1.3 mm windows. Results. In total, 28 transition lines of 22 molecular species and those isotopologues are detected towards IRAS 19312+1950, some of which exhibit a broad and a narrow components. Seventeen thermal lines and 1 maser line are newly detected. The molecular species of C17^{17}O, 30^{30}SiO, HN13^{13}C, HC18^{18}O+^{+}, H2_{2}CO, and cc-C3_{3}H2_{2} are detected for the first time in this object. Conclusions. Our results, in combination with previous studies, favor the hypothesis that IRAS 19312+1950 might be a red nova remnant, in which the progenitors that merged to become a red nova may have contained at least two evolved stars with oxygen-rich and carbon-rich chemistry, respectively.Comment: 30 pages, 24 figures, accepted for publication in A&

    AI safety of film capacitors.

    With a large number of film capacitors being deployed in critical locations in electrical and electronic systems, artificial intelligence (AI) technology is also expected to address the problems encountered in this process. According to our findings, AI applications can cover the entire lifecycle of film capacitors. However, the AI safety hazards in these applications have not received the attention they deserve. To meet this, the authors argue, with specific examples, risks that flawed, erratic, and unethical AI can introduce in the design, operation, and evaluation of film capacitors. Human-AI common impact and more multi-dimensional evaluation for AI are proposed to better cope with unknown, ambiguity, and known risks brought from AI in film capacitors now and in the future

    Evaluation of human enterovirus 71 and coxsackievirus A16 specific immunoglobulin M antibodies for diagnosis of hand-foot-and-mouth disease

    <p>Abstract</p> <p>Background</p> <p>Hand-foot-and-mouth disease (HFMD) is caused mainly by the human enterovirus type 71 (HEV71) and the Coxsackievirus A group type 16 (CVA16). Large outbreaks of disease have occurred frequently in the Asia-Pacific region. Reliable methods are needed for diagnosis of HFMD in childen. IgM-capture ELISA, with its notable advantages of convenience and low cost, provides a potentially frontline assay. We aimed to evaluate the newly developed IgM-capture ELISAs for HEV71 and CVA16 in the diagnosis of HFMD, and to measure the kinetics of IgM over the course of HEV71 or CVA16 infections.</p> <p>Results</p> <p>We mapped, for the first time, the kinetics of IgM in HEV71 and CVA16 infection. HEV71- and CVA16-IgM were both detectable in some patients on day 1 of illness, and in 100% of patients by day 5 (HEV71) and day 8 (CVA16) respectively; both IgMs persisted for several weeks. The IgM detection rates were 90.2% (138 of 153 sera) and 68.0% (66 of 97 sera) for HEV71 and CVA16 infections, respectively, during the first 7 days of diseases. During the first 90 days after onset these values were 93.6% (233 of 249 sera) and 72.8% (91 of 125 sera) for HEV71 and CVA16 infections, respectively. Some cross-reactivity was observed between HEV71- and CVA16-IgM ELISAs. HEV71-IgM was positive in 38 of 122 (31.1%) CVA16 infections, 14 of 49 (28.6%) other enteroviral infections and 2 of 105 (1.9%) for other respiratory virus infected sera. Similarly, CVA16-IgM was apparently positive in 58 of 211 (27.5%) HEV71 infections, 16 of 48 (33.3%) other enterovirus infections and 3 of 105 (2.9%) other respiratory virus infected sera. Nevertheless, the ELISA yielded the higher OD<sub>450 </sub>value of main antibody than that of cross-reaction antibody, successfully identifying the enteroviral infection in 96.6% (HEV71) and 91.7% (CVA16) cases. When blood and rectal swabs were collected on the same day, the data showed that the agreement between IgM-capture ELISA and real-time RT-PCR in HEV71 was high (Kappa value = 0.729) while CVA16 somewhat lower (Kappa value = 0.300).</p> <p>Conclusions</p> <p>HEV71- and CVA16-IgM ELISAs can be deployed successfully as a convenient and cost-effective diagnostic tool for HFMD in clinical laboratories.</p

    Data sonification of film capacitors.

    Film capacitors are playing an increasingly important role in power-related fields, driven by the continuous development of dielectric materials and practical needs. Long-term accumulation has also led to an increasing wealth of data related to film capacitors. Sonification opens up a new way for people to make good use of data from film capacitors. A framework for sonifying film capacitors data based on TwoTone is presented. Based on the analysis and discussion, it is clear that the sonification results can easily represent the monotonic variation pattern of film capacitors data. What's more, the sonification results increase the possibility that people pay attention to the changing trend of film capacitors data when there is no significant difference in the visual perception of the data. In addition to providing a new way of music generation of electrical equipment, the method proposed is expected to contribute to theory reference in typical scenarios, such as factory calibration of film capacitors, monitoring of film capacitor operation status, and presentation of statistical data of film capacitors' dielectric materials, which will help us to better understand the distribution characteristics of polymer films

    The role of c-reactive protein and fibrinogen in the development of intracerebral hemorrhage: A mendelian randomization study in European population

    Background: The causal association of C-reactive protein (CRP) and fibrinogen on intracerebral hemorrhage (ICH) remains uncertain. We investigated the causal associations of CRP and fibrinogen with ICH using two-sample Mendelian randomization. Method: We used single-nucleotide polymorphisms associated with CRP and fibrinogen as instrumental variables. The summary data on ICH were obtained from the International Stroke Genetics Consortium (1,545 cases and 1,481 controls). Two-sample Mendelian randomization estimates were performed to assess with inverse-variance weighted and sensitive analyses methods including the weighted median, the penalized weighted median, pleiotropy residual sum and outlier (MR-PRESSO) approaches. MR-Egger regression was used to explore the pleiotropy. Results: The MR analyses indicated that genetically predicted CRP concentration was not associated with ICH, with an odds ratio (OR) of 1.263 (95% CI = 0.935–1.704, p = 0.127). Besides, genetically predicted fibrinogen concentration was not associated with an increased risk of ICH, with an OR of 0.879 (95% CI = 0.060–18.281; p = 0.933). No evidence of pleiotropic bias was detected by MR-Egger. The findings were overall robust in sensitivity analyses. Conclusions: Our findings did not support that CRP and fibrinogen are causally associated with the risk of ICH
