    Novel interface-selected waves and their influences on wave competitions

    The topic of interface effects in wave propagation has attracted great attention due to their theoretical significance and practical importance. In this paper we study nonlinear oscillatory systems consisting of two media separated by an interface, and find a novel phenomenon: interface can select a type of waves (ISWs). Under certain well defined parameter condition, these waves propagate in two different media with same frequency and same wave number; the interface of two media is transparent to these waves. The frequency and wave number of these interface-selected waves (ISWs) are predicted explicitly. Varying parameters from this parameter set, the wave numbers of two domains become different, and the difference increases from zero continuously as the distance between the given parameters and this parameter set increases from zero. It is found that ISWs can play crucial roles in practical problems of wave competitions, e.g., ISWs can suppress spirals and antispirals

    Studies on the formation and forming mechanism of the related substance E in potassium clavulanate production by HPLC-MS/MS

    O objetivo deste estudo foi investigar a formação da substância E e o respectivo mecanismo na produção de clavulanato de potássio. Confirmou-se a impureza com tempo de retenção de 11,1 min no produto final, clavulanato de potássio, como substância E, por meio de cromatografia líquida de alta eficiência, em conjunto com detecção por espectrometria de massas (CLAE-MS-MS). A análise da substância relacionada E durante a produção do ácido clavulânico mostrou que essa impureza pode ser formada tanto durante a fermentação quanto durante os processos de purificação, especialmente no estágio final de fermentação, filtração, concentração e procedimento de extração. O ácido clavulãnico foi o precursor da substância E. Estudos no mecanismo de sua formação mostraram que a substância E formou-se pela combinação do grupo imina da molécula do ácido clavulânico com o grupo carboxílico de outra molécula de ácido clavulânico, com a abertura do anel β-lactâmico. Resultados do teste ortogonal multifatorial confirmaram que a concentração do ácido clavulânico foi o fator dominante para acelerar a reação, enquanto a temperatura foi outro fator que contribuiu. O pH de 5,0 a 6,5 teve pouco impacto na geração da substância E.The objective of this study was to investigate the formation and forming mechanism of the related substance E in potassium clavulanate production. The impurity with retention time of 11.1 min in potassium clavulanate final product was confirmed as the related substance E by high performance liquid chromatography with tandem mass spectrometric detection (HPLC-MS/MS).The related substance E analysis during the production of clavulanic acid showed that this impurity could be formed during both the fermentation and purification processes, especially in the later fermentation stage, filtration concentration and back-extraction procedure. Clavulanic acid was the precursor of the related substance E. Studies on its forming mechanism showed that the related substance E was formed by the combination of the imino group of one molecule of clavulanic acid with the carboxyl group of another molecule of clavulanic acid with the opening of β-lactam ring. Results of a multi-factor orthogonal test confirmed that the concentration of clavulanic acid was the dominant factor to accelerate the reaction, while the temperature was another contributing factor. The pH 5.0-6.5 had little impact on the generation of the related substance E

    In vitro analysis of phosphorothioate modification of DNA reveals substrate recognition by a multiprotein complex

    A wide variety of prokaryotes possess DNA modifications consisting of sequence-specific phosphorothioates (PT) inserted by members of a five-gene cluster. Recent genome mapping studies revealed two unusual features of PT modifications: short consensus sequences and partial modification of a specific genomic site in a population of bacteria. To better understand the mechanism of target selection of PT modifications that underlies these features, we characterized the substrate recognition of the PT-modifying enzymes termed DptC, D and E in a cell extract system from Salmonella. The results revealed that double-stranded oligodeoxynucleotides underwent de novo PT modification in vitro, with the same modification pattern as in vivo, i. e., GpsAAC/GpsTTC motif. Unexpectedly, in these in vitro analyses we observed no significant effect on PT modification by sequences flanking GAAC/GTTC motif, while PT also occurred in the GAAC/GTTC motif that could not be modified in vivo. Hemi-PT DNA also served as substrate of the PT-modifying enzymes, but not single-stranded DNA. The PT-modifying enzymes were then found to function as a large protein complex, with all of three subunits in tetrameric conformations.     Pathological phenotypes and in vivo DNA cleavage by unrestrained activity of a phosphorothioate-based restriction system in Salmonella

    Prokaryotes protect their genomes from foreign DNA with a diversity of defence mechanisms, including a widespread restriction–modification (R–M) system involving phosphorothioate (PT) modification of the DNA backbone. Unlike classical R–M systems, highly partial PT modification of consensus motifs in bacterial genomes suggests an unusual mechanism of PT-dependent restriction. In Salmonella enterica, PT modification is mediated by four genes dptB–E, while restriction involves additional three genes dptF–H. Here, we performed a series of studies to characterize the PT-dependent restriction, and found that it presented several features distinct with traditional R–M systems. The presence of restriction genes in a PT-deficient mutant was not lethal, but instead resulted in several pathological phenotypes. Subsequent transcriptional profiling revealed the expression of > 600 genes was affected by restriction enzymes in cells lacking PT, including induction of bacteriophage, SOS response and DNA repair-related genes. These transcriptional responses are consistent with the observation that restriction enzymes caused extensive DNA cleavage in the absence of PT modifications in vivo. However, overexpression of restriction genes was lethal to the host in spite of the presence PT modifications.     Predicted Electronic Markers for Polytypes of LaOBiS\u3csub\u3e2\u3c/sub\u3e Examined via Angle-Resolved Photoemission Spectroscopy

    The natural periodic stacking of symmetry-inequivalent planes in layered compounds can lead to the formation of natural superlattices; albeit close in total energy, (thus in their thermodynamic stability), such polytype superlattices can exhibit different structural symmetries, thus have markedly different electronic properties which can in turn be used as “structural markers”. We illustrate this general principle on the layered LaOBiS2 compound where density-functional theory (DFT) calculations on the (BiS2)/(LaO)/(BiS2) polytype superlattices reveal both qualitatively and quantitatively distinct electronic structure markers associated with the Rashba physics, yet the total energies are only ∼ 0.1 meV apart. This opens the exciting possibility of identifying subtle structural features via electronic markers. We show that the pattern of removal of band degeneracies in different polytypes by the different forms of symmetry breaking leads to Rashba “minigaps” with characteristic Rashba parameters that can be determined from spectroscopy, thereby narrowing down the physically possible polytypes. By identifying these distinct DFT-predicted fingerprints via angle-resolved photoemission spectroscopy (ARPES) measurements on LaBiOS2 we found the dominant polytype with small amounts of mixtures of other polytypes. This conclusion, consistent with neutron scattering results, establishes ARPES detection of theoretically established electronic markers as a powerful tool to delineate energetically quasidegenerate polytypes

    Disconnection-mediated Twin/Twin-junction migration in FCC metals

    We present the results of novel, time-resolved, in situ HRTEM observations, molecular dynamics (MD) simulations, and disconnection theory that elucidate the mechanism by which the motion of grain boundaries (GBs) in polycrystalline materials are coupled through disconnection motion/reactions at/adjacent to GB triple junctions (TJs). We focus on TJs composed of a pair of coherent twin boundaries (CTBs) and a Σ9 GB in copper. As for all GBs, disconnection theory implies that multiple modes/local mechanisms for CTB migration are possible and that the mode selection is affected by the nature of the driving force for migration. While we observe (HRTEM and MD) CTB migration through the motion of pure steps driven by chemical potential jump, other experimental observations (and our simulations) show that stress-driven CTB migration occurs through the motion of disconnections with a non-zero Burgers vector; these are pure-step and twinning-partial CTB migration mechanisms. Our experimental observations and simulations demonstrate that the motion of a GB drags its delimiting TJ and may force the motion of the other GBs meeting at the TJ. Our experiments and simulations focus on two types of TJs composed of a pair of CTBs and a Σ9 GB; a 107° TJ readily migrates while a 70° TJ is immobile (experiment, simulation) in agreement with our disconnection theory even though the intrinsic mobilities of the constituent GBs do not depend on TJ-type. We also demonstrate that disconnections may be formed at TJs (chemical potential jump/stress driven) and at GB/free surface junctions (stress-driven)

    The north–south shift of the ridge location of the western Pacific subtropical high and its influence on the July precipitation in the Jianghuai region from 1978 to 2021

    The Jianghuai region is the area between the Yangtze River and the Huai River in China and is a densely populated agriculture region therefore, the economics and human activity there are significantly affected by the precipitation changes, particularly during the summer when extreme storms and droughts normally occur. It will be helpful if the summer precipitation changes can be predicted. The monthly ERA5 atmospheric reanalysis data from 1978 to 2021 are used in this study to investigate the relationship between the ridge latitudinal location of the western Pacific subtropical high (WPSH) and the precipitation in July over the Jianghuai region. The results show that the WPSH ridge location has an important impact on the amount and spatial distribution of the precipitation in this region. When the ridge was northward, an anomalous anticyclonic circulation will appear over the western Pacific, leading to the weakening of the summer monsoon and the reduction of moisture transport from the Indian Ocean, therefore decreasing precipitation in the Jianghuai region, while the situation is opposite when the ridge was southward. The Niño 3.4 index in March and the India–Burma trough intensity index in June have significant correlations with the July WPSH ridge location, and both can be used as precursors to predict the WPSH ridge location and, therefore, the precipitation in this region

    Long-range prediction of the tropical cyclone frequency landfalling in China using thermocline temperature anomalies at different longitudes

    The landfalls of the tropical cyclone (TC) along the coast of China have caused huge economic damages. There are approximately nine TC landfalls in China every year. It will be beneficial if the landfall frequency can be predicted in advance. Inspired by the study of Sparks and Toumi (Commun Earth Environ, 30-1-2020), six datasets, including four ocean reanalyses and two object analyses from 1993 to 2019, are employed to study the consistency in the relationship between the thermocline temperature anomalies at different longitudes and the frequency of TC landfalls along the coastal areas of China (South China, East China, and the whole of China). The thermocline temperature anomalies at different longitudes are tested in order to confirm our hypothesis that the eastward and westward transports of ocean heat from the warm pool are the causes of the significant correlations. The results show some significant correlations at various longitudes, and the temperature anomalies can predict the TC landfall frequency for several months or longer. Further study also shows the close relationship between the ocean heat transport and the sea surface temperature anomalies at the genesis locations of TC landfalls. The locations of the western Pacific subtropical high (WPSH) during high-frequency TC landfall years also show favorable spatial patterns to the TC landfall in South China and East China, respectively. In years with a high TC frequency in South China, the westward displacement of the WPSH ridge steers TC toward South China, while during high-frequency TC landfall years in East China, WPSH is located further north, and the westward extension of the ridge is in close proximity to the East China Sea
