    Influencing Factors Study of the Variable Speed Scroll Compressor with EVI Technology

    Heating capacity of general air-condition system at low ambient temperature has been heavily attenuated. To solve the problem stated, the variable speed Scroll compressor with enhanced vapor injection (EVI) becomes the focus of the study in recent years. Influencing factors of the capacity and energy efficiency of the scroll compressor with EVI technology have been introduced in paper, including the area of Supplementary channel, the parameters of orbiting and fixed scroll profile, the position and number of Supplementary inlet. The different scheme of prototype is tested and the effect of the scheme is compared and analyzed. With the vapor injected into compress chambers, the Gas forces of working chambers change, and the characteristics of gas force about the scroll compressor with EVI Technology have also been analyzed

    Dual-Stream Pyramid Registration Network

    We propose a Dual-Stream Pyramid Registration Network (referred as Dual-PRNet) for unsupervised 3D medical image registration. Unlike recent CNN-based registration approaches, such as VoxelMorph, which explores a single-stream encoder-decoder network to compute a registration fields from a pair of 3D volumes, we design a two-stream architecture able to compute multi-scale registration fields from convolutional feature pyramids. Our contributions are two-fold: (i) we design a two-stream 3D encoder-decoder network which computes two convolutional feature pyramids separately for a pair of input volumes, resulting in strong deep representations that are meaningful for deformation estimation; (ii) we propose a pyramid registration module able to predict multi-scale registration fields directly from the decoding feature pyramids. This allows it to refine the registration fields gradually in a coarse-to-fine manner via sequential warping, and enable the model with the capability for handling significant deformations between two volumes, such as large displacements in spatial domain or slice space. The proposed Dual-PRNet is evaluated on two standard benchmarks for brain MRI registration, where it outperforms the state-of-the-art approaches by a large margin, e.g., having improvements over recent VoxelMorph [2] with 0.683->0.778 on the LPBA40, and 0.511->0.631 on the Mindboggle101, in term of average Dice score.Comment: To appear in MICCAI 2019 (Oral

    KMT2A promotes melanoma cell growth by targeting hTERT signaling pathway.

    Melanoma is an aggressive cutaneous malignancy, illuminating the exact mechanisms and finding novel therapeutic targets are urgently needed. In this study, we identified KMT2A as a potential target, which promoted the growth of human melanoma cells. KMT2A knockdown significantly inhibited cell viability and cell migration and induced apoptosis, whereas KMT2A overexpression effectively promoted cell proliferation in various melanoma cell lines. Further study showed that KMT2A regulated melanoma cell growth by targeting the hTERT-dependent signal pathway. Knockdown of KMT2A markedly inhibited the promoter activity and expression of hTERT, and hTERT overexpression rescued the viability inhibition caused by KMT2A knockdown. Moreover, KMT2A knockdown suppressed tumorsphere formation and the expression of cancer stem cell markers, which was also reversed by hTERT overexpression. In addition, the results from a xenograft mouse model confirmed that KMT2A promoted melanoma growth via hTERT signaling. Finally, analyses of clinical samples demonstrated that the expression of KMT2A and hTERT were positively correlated in melanoma tumor tissues, and KMT2A high expression predicted poor prognosis in melanoma patients. Collectively, our results indicate that KMT2A promotes melanoma growth by activating the hTERT signaling, suggesting that the KMT2A/hTERT signaling pathway may be a potential therapeutic target for melanoma

    Citizen Ai: warrior, jester, and middleman

    Ai Weiwei is famous for crossing boundaries, especially the boundary between art and politics. To appreciate the often contradictory nature of Ai’s work, this essay employs multiple narratives: “Ai the Heroic Warrior” who criticizes the Chinese government; “Ai the Court Jester” who plays with the Chinese state and Western media; and “Ai the Middleman” who acts as a broker between China and the West, between young and old people, and between civil society and the state in the PRC. The essay concludes that a fourth narrative can bring together these three stories in a multicoded understanding of Ai’s work: “Ai the Citizen Intellectual” who sometimes works with the state, and at other times against it—but always for the good of China. By comparing Ai’s work with that of other public intellectuals and placing it in the context of debates about civil society, the conclusion argues that “citizen intellectual” also tells us about a broader movement of activists and public intellectuals who are creating a new form of political space in postsocialist China

    Transcriptome Profile Analysis Reveals an Estrogen Induced LncRNA Associated with Lipid Metabolism and Carcass Traits in Chickens (Gallus Gallus)

    Background/Aims: Accumulating evidences have demonstrated that long noncoding RNAs (lncRNA) play important roles in hepatic lipid metabolism in mammals. However, no systematic screening of the potential lncRNAs in the livers of laying hens has been performed, and few studies have been reported concerning the effects of the lncRNAs on lipid metabolism in the livers of chickens during egg-laying period. The purpose of this study was to compare the difference in lncRNA expression in the livers of pre-laying and peak-laying hens at the age of 20 and 30 weeks old by transcriptome sequencing and to investigate the interaction networks among lncRNAs, mRNAs and miRNAs. Moreover, the regulatory mechanism and biological function of lncLTR, a significantly differentially expressed lncRNA in the liver between pre- and peak-laying hens, was explored in vitro and in vivo. Methods: Bioinformatics analyses were conducted to identify the differentially expressed (DE) lncRNAs between the two groups of hens. The target genes of the DE lncRNA were predicated for further functional enrichment. An integrated analysis was performed among the DE lncRNA datasets, DE mRNAs and DE miRNA datasets obtained from the same samples to predict the interaction relationship. In addition, in vivo and in vitro trials were carried out to determine the expression regulation of lncLTR, and polymorphism association analysis was conducted to detect the biological role of ncLTR. Results: A total of 124 DE lncRNAs with a P-value ≤ 0.05 were identified. Among them, 44 lncRNAs including 30 known and 14 novel lncRNAs were significant differentially expressed (SDE) with FDR ≤ 0.05. Thirty-two lncRNAs were upregulated and 12 were downregulated in peak-laying group compared with pre-laying group. The functional enrichment results revealed that target genes of some lncRNAs are involved in the lipid metabolism process. Integrated analysis suggested that some of the genes involved in lipid metabolism might be regulated by both the lncRNA and the miRNA. In addition, an upregulated lncRNA, designated lncLTR, was demonstrated to be induced by estrogen via ERβ signaling. The c242. G>A SNP in lncLTR was significantly associated with chicken carcass weight, evisceration weight, semi-evisceration weight, head weight, double-wing weight, claw weight traits, and blood biochemical index, especially for the blood triglyceride content. Conclusion: A series of lncRNAs associated with lipid metabolism in the livers of chickens were identified by transcriptome sequencing and functional analysis, providing a valuable data resource for further studies on chicken hepatic metabolism activities.     Construction and Characterization of a Bacterial Artificial Chromosome Library for the Hexaploid Wheat Line 92R137

    For map-based cloning of genes conferring important traits in the hexaploid wheat line 92R137, a bacterial artificial chromosome (BAC) library, including two sublibraries, was constructed using the genomic DNA of 92R137 digested with restriction enzymes HindIII and BamHI. The BAC library was composed of total 765,696 clones, of which 390,144 were from the HindIII digestion and 375,552 from the BamHI digestion. Through pulsed-field gel electrophoresis (PFGE) analysis of 453 clones randomly selected from the HindIII sublibrary and 573 clones from the BamHI sublibrary, the average insert sizes were estimated as 129 and 113 kb, respectively. Thus, the HindIII sublibrary was estimated to have a 3.01-fold coverage and the BamHI sublibrary a 2.53-fold coverage based on the estimated hexaploid wheat genome size of 16,700 Mb. The 765,696 clones were arrayed in 1,994 384-well plates. All clones were also arranged into plate pools and further arranged into 5-dimensional (5D) pools. The probability of identifying a clone corresponding to any wheat DNA sequence (such as gene Yr26 for stripe rust resistance) from the library was estimated to be more than 99.6%. Through polymerase chain reaction screening the 5D pools with Xwe173, a marker tightly linked to Yr26, six BAC clones were successfully obtained. These results demonstrate that the BAC library is a valuable genomic resource for positional cloning of Yr26 and other genes of interest

    Development of New Microsatellite DNA Markers from Apostichopus japonicus and Their Cross-Species Application in Parastichopus parvimensis and Pathallus mollis

    Twenty microsatellite DNA markers were developed for sea cucumber and used to investigate polymorphisms of 60 wild Apostichopus japonicus individuals collected from China. It revealed that all the markers were polymorphic. A total of 164 alleles were detected at 20 loci. The number of alleles per locus varied from 3 to 17 with an average of 8.2, and the expected heterozygosities of each locus ranged from 0.03 to 0.89 with an average of 0.64. Cross-species amplification was also conducted in Parastichopus parvimensis collected from the United States and Pathallus mollis collected from Peru. The result showed that 17 loci amplified Parastichopus parvimensis DNAs while only 4 loci could amplify Pathallus mollis DNAs. All of the polymorphic markers would be useful for future genetic breeding and the assessment of genetic variation within sea cucumbers
