    Inelastic neutron scattering study of crystal field excitations of Nd<sup>3+</sup> in NdFeAsO

    Inelastic neutron scattering experiments were performed to investigate the crystalline electric field (CEF) excitations of Nd3+ (J = 9/2) in the iron pnictide NdFeAsO. The crystal field level structures for both the high-temperature paramagnetic phase and the low-temperature antiferromagnetic phase of NdFeAsO are constructed. The variation of CEF excitations of Nd3+ reflects not only the change of local symmetry but also the change of magnetic ordered state of the Fe sublattice. By analyzing the crystal field interaction with a crystal field Hamiltonian, the crystal field parameters are obtained. It was found that the sign of the fourth and sixth-order crystal field parameters change upon the magnetic phase transition at 140 K, which may be due to the variation of exchange interactions between the 4f and conduction electrons.Comment: 5 pages, 4 figure

    Continental igneous rock composition: A major control of past global chemical weathering

    The composition of igneous rocks in the continental crust has changed throughout Earth’s history. However, the impact of these compositional variations on chemical weathering, and by extension on seawater and atmosphere evolution, is largely unknown. We use the strontium isotope ratio in seawater [(87Sr/86Sr)seawater] as a proxy for chemical weathering, and we test the sensitivity of (87Sr/86Sr)seawater variations to the strontium isotopic composition (87Sr/86Sr) in igneous rocks generated through time. We demonstrate that the 87Sr/86Sr ratio in igneous rocks is correlated to the epsilon hafnium (εHf) of their hosted zircon grains, and we use the detrital zircon record to reconstruct the evolution of the 87Sr/86Sr ratio in zircon-bearing igneous rocks. The reconstructed 87Sr/86Sr variations in igneous rocks are strongly correlated with the (87Sr/86Sr)seawater variations over the last 1000 million years, suggesting a direct control of the isotopic composition of silicic magmatism on (87Sr/86Sr)seawater variations. The correlation decreases during several time periods, likely reflecting changes in the chemical weathering rate associated with paleogeographic, climatic, or tectonic events. We argue that for most of the last 1000 million years, the (87Sr/86Sr)seawater variations are responding to changes in the isotopic composition of silicic magmatism rather than to changes in the global chemical weathering rate. We conclude that the (87Sr/86Sr)seawater variations are of limited utility to reconstruct changes in the global chemical weathering rate in deep times

    Challenges of Early Estimation of Infrastructure Projects within the UK: an Information Perspective

    This research examines the challenges of early cost estimation of infrastructure projects within the UK with a focus on why cost overruns are such a persistent issue. The research was carried out by an investigation of existing literature surrounding early estimation in construction, characteristics of UK-based infrastructure projects and cost overruns within infrastructure projects. The data were collected through fourteen semi-structured interviews with estimating professionals who work predominantly on infrastructure projects across the UK. The key findings were that the early estimation process consists of an intricate system of hard and soft information exchange from the many involved parties due to the social and political nature of infrastructure projects; this provides many challenges for the estimator. This led to the idea that an estimate is actually a soft input itself and should not be taken as a hard numerical figure but something which requires human interpretation. Moreover, these challenges are heightened by the number of unknowns and uncertainties that are again part of the very nature of large scale infrastructure projects. This is difficult to address as it is the process of converting soft information into hard information and when soft information is hardened it will inevitably lose some of its information or context. Unfortunately, cost estimates have become to be accepted as hard information even when it is known to be soft information that requires interpretation and sense making

    Lessons learnedThe combination of standard dose abiraterone acetate and BEZ235, a pan-class I PI3K and mTORC1/2 inhibitor, was poorly tolerated in men with progressive mCRPC.Although the clinical development of BEZ235 has been discontinued in prostate cancer, agents that more selectively target PI3K-AKT-mTOR signaling may have a more favorable therapeutic index and should continue to be explored.BackgroundAndrogen receptor (AR) and phosphatidylinositol-3 kinase (PI3K) signaling are two commonly perturbed pathways in prostate cancer. Preclinical data have shown that the two pathways compensate for each other when one is inhibited, and combined inhibition of AR and PI3K signaling may be a viable strategy to prevent or overcome castration resistance.MethodsThis phase I study evaluated the safety and tolerability of abiraterone acetate and prednisone combined with BEZ235, a dual PI3K and mTORC1/2 inhibitor, in men with progressive metastatic castration resistant prostate cancer (mCRPC) who have not received prior chemotherapy.ResultsSix patients (n = 6) were treated at the starting dose level of abiraterone acetate 1,000 mg with prednisone 5 mg twice daily and BEZ235 200 mg twice daily in a 3 + 3 dose escalation design. The study was terminated early because three of the six patients (50%) experienced dose-limiting toxicities: grade 3 mucositis, grade 3 hypotension, and grade 4 dyspnea and pneumonitis. All six patients had previously progressed on abiraterone/prednisone. The median treatment duration was 27 days (range: 3-130 days). No prostate-specific antigen (PSA) decline or objective response were observed.ConclusionThe combination of standard-dose abiraterone/prednisone with BEZ235 200 mg twice daily was poorly tolerated in patients with mCRPC. The on-target and off-target effects of dual PI3K and mTORC inhibition likely contributed to the unacceptable toxicity profile. The Oncologist 2017;22:503-e43

    Mid-infrared flare of TDE candidate PS16dtm: dust echo and implications for the spectral evolution

    PS16dtm was classified as a candidate tidal disruption event (TDE) in a dwarf Seyfert 1 galaxy with low-mass black hole (106M\sim10^6M\odot) and has presented various intriguing photometric and spectra characteristics. Using the archival WISE and the newly released NEOWISE data, we found PS16dtm is experiencing a mid-infrared (MIR) flare which started 11\sim11 days before the first optical detection. Interpreting the MIR flare as a dust echo requires close pre-existing dust with a high covering factor, and suggests the optical flare may have brightened slowly for some time before it became bright detectable from the ground. More evidence is given at the later epochs. At the peak of the optical light curve, the new inner radius of the dust torus has grown to much larger size, a factor of 7 of the initial radius due to strong radiation field. At 150\sim150 days after the first optical detection, the dust temperature has dropped well below the sublimation temperature. Other peculiar spectral features shown by PS16dtm are the transient, prominent FeII emission lines and outflows indicated by broad absorption lines detected during the optical flare. Our model explains the enhanced FeII emission from iron newly released from the evaporated dust. The observed broad absorption line outflow could be explained by accelerated gas in the dust torus due to the radiation pressure.Comment: Accepted by ApJ, 5 figure

    Anti-TNF-α antibody allows healing of joint damage in polyarthritic transgenic mice

    Anti-tumor-necrosis-factor-α (TNF-α) monoclonal antibody was used to treat Tg197 transgenic mice, which constitutively produce human TNF-α (hTNF-α) and develop a progressive polyarthritic disease. Treatment of both young (7- or 8-week-old) and aged (27- or 28-week-old) mice commenced when at least two limbs showed signs of moderate to severe arthritis. The therapeutic efficacy of anti-TNF-α antibody was assessed using various pathological indicators of disease progression. The clinical severity of arthritis in Tg197 mice was significantly reduced after anti-TNF-α treatment in comparison with saline-treated mice and in comparison with baseline assessments in both young and aged mice. The treatment with anti-TNF-α prevented loss of body weight. Inflammatory pathways as reflected by elevated circulating hTNF-α and local expression of various proinflammatory mediators were all diminished by anti-TNF-α treatment, confirming a critical role of hTNF-α in this model of progressive polyarthritis. More importantly, the amelioration of the disease was associated with reversal of existing structural damage, including synovitis and periosteal bone erosions evident on histology. Repair of cartilage was age dependent: reversal of cartilage degradation after anti-TNF-α treatment was observed in young mice but not in aged mice

    A Network of Multi-Tasking Proteins at the DNA Replication Fork Preserves Genome Stability

    To elucidate the network that maintains high fidelity genome replication, we have introduced two conditional mutant alleles of DNA2, an essential DNA replication gene, into each of the approximately 4,700 viable yeast deletion mutants and determined the fitness of the double mutants. Fifty-six DNA2-interacting genes were identified. Clustering analysis of genomic synthetic lethality profiles of each of 43 of the DNA2-interacting genes defines a network (consisting of 322 genes and 876 interactions) whose topology provides clues as to how replication proteins coordinate regulation and repair to protect genome integrity. The results also shed new light on the functions of the query gene DNA2, which, despite many years of study, remain controversial, especially its proposed role in Okazaki fragment processing and the nature of its in vivo substrates. Because of the multifunctional nature of virtually all proteins at the replication fork, the meaning of any single genetic interaction is inherently ambiguous. The multiplexing nature of the current studies, however, combined with follow-up supporting experiments, reveals most if not all of the unique pathways requiring Dna2p. These include not only Okazaki fragment processing and DNA repair but also chromatin dynamics

    Low-grade systemic inflammation is often associated with metabolic syndrome, which plays a critical role in the development of the obesity-associated inflammatory diseases, including insulin resistance and atherosclerosis. Here, we investigate how Toll-like receptor-MyD88 signaling in myeloid and endothelial cells coordinately participates in the initiation and progression of high fat diet-induced systemic inflammation and metabolic inflammatory diseases. MyD88 deficiency in myeloid cells inhibits macrophage recruitment to adipose tissue and their switch to an M1-like phenotype. This is accompanied by substantially reduced diet-induced systemic inflammation, insulin resistance, and atherosclerosis. MyD88 deficiency in endothelial cells results in a moderate reduction in diet-induced adipose macrophage infiltration and M1 polarization, selective insulin sensitivity in adipose tissue, and amelioration of spontaneous atherosclerosis. Both in vivo and ex vivo studies suggest that MyD88-dependent GM-CSF production from the endothelial cells might play a critical role in the initiation of obesity-associated inflammation and development of atherosclerosis by priming the monocytes in the adipose and arterial tissues to differentiate into M1-like inflammatory macrophages. Collectively, these results implicate a critical MyD88-dependent interplay between myeloid and endothelial cells in the initiation and progression of obesity-associated inflammatory diseases

    RE: How the Coronavirus Disease-2019 May Improve Care: Rethinking Cervical Cancer Prevention

    Feldman and Haas have written a timely piece on the potential to enhance cancer prevention and cancer care delivery in the COVID-19 era. Using cervical cancer prevention as a use case, the commentary describes clinical care provided via virtual platforms and in nontraditional settings, such as the patient’s home, as areas needing creative approaches to ensure care is provided safely and efficiently. As we consider factors that are relevant to delivering effective cancer prevention and cancer care post-COVID, we suggest that addressing social determinants of health, an often forgotten dimension of lived experience, should be prioritized as a strategy to enhance the equity of care provision. Social determinants of health, including food and housing insecurity have been shown to impact outcomes of patients with cancer, through a number of mechanisms including delays and incomplete care
