    Interactions of the Gasotransmitters Contribute to Microvascular Tone (Dys)regulation in the Preterm Neonate

    Background & Aims Hydrogen sulphide (H2S), nitric oxide (NO), and carbon monoxide (CO) are involved in transitional microvascular tone dysregulation in the preterm infant; however there is conflicting evidence on the interaction of these gasotransmitters, and their overall contribution to the microcirculation in newborns is not known. The aim of this study was to measure the levels of all 3 gasotransmitters, characterise their interrelationships and elucidate their combined effects on microvascular blood flow. Methods 90 preterm neonates were studied at 24h postnatal age. Microvascular studies were performed by laser Doppler. Arterial COHb levels (a measure of CO) were determined through co-oximetry. NO was measured as nitrate and nitrite in urine. H2S was measured as thiosulphate by liquid chromatography. Relationships between levels of the gasotransmitters and microvascular blood flow were assessed through partial correlation controlling for the influence of gestational age. Structural equation modelling was used to examine the combination of these effects on microvascular blood flow and derive a theoretical model of their interactions. Results No relationship was observed between NO and CO (p = 0.18, r = 0.18). A positive relationship between NO and H2S (p = 0.008, r = 0.28) and an inverse relationship between CO and H2S (p = 0.01, r = -0.33) exists. Structural equation modelling was used to examine the combination of these effects on microvascular blood flow. The model with the best fit is presented. Conclusions The relationships between NO and H2S, and CO and H2S may be of importance in the preterm newborn, particularly as NO levels in males are associated with higher H2S levels and higher microvascular blood flow and CO in females appears to convey protection against vascular dysregulation.     Microflare Heating of a Solar Active Region Observed with NuSTAR, Hinode/XRT, and SDO/AIA

    NuSTAR is a highly sensitive focusing hard X-ray (HXR) telescope and has observed several small microflares in its initial solar pointings. In this paper, we present the first joint observation of a microflare with NuSTAR and Hinode/XRT on 2015 April 29 at ~11:29 UT. This microflare shows heating of material to several million Kelvin, observed in Soft X-rays (SXRs) with Hinode/XRT, and was faintly visible in Extreme Ultraviolet (EUV) with SDO/AIA. For three of the four NuSTAR observations of this region (pre-, decay, and post phases) the spectrum is well fitted by a single thermal model of 3.2-3.5 MK, but the spectrum during the impulsive phase shows additional emission up to 10 MK, emission equivalent to A0.1 GOES class. We recover the differential emission measure (DEM) using SDO/AIA, Hinode/XRT, and NuSTAR, giving unprecedented coverage in temperature. We find the pre-flare DEM peaks at ~3 MK and falls off sharply by 5 MK; but during the microflare's impulsive phase the emission above 3 MK is brighter and extends to 10 MK, giving a heating rate of about 2.5×10252.5 \times 10^{25} erg s1^{-1}. As the NuSTAR spectrum is purely thermal we determined upper-limits on the possible non-thermal bremsstrahlung emission. We find that for the accelerated electrons to be the source of the heating requires a power-law spectrum of δ7\delta \ge 7 with a low energy cut-off Ec7E_{c} \lesssim 7 keV.     Identification of a variant in NDP associated with X-linked retinal dysplasia in the English cocker spaniel dog.

    PURPOSE: Three related male English Cocker Spaniels (ECS) were reported to be congenitally blind. Examination of one of these revealed complete retinal detachment. A presumptive diagnosis of retinal dysplasia (RD) was provided and pedigree analysis was suggestive of an X-linked mode of inheritance. We sought to investigate the genetic basis of RD in this family of ECS. METHODS: Following whole genome sequencing (WGS) of the one remaining male RD-affected ECS, two distinct investigative approaches were employed: a candidate gene approach and a whole genome approach. In the candidate gene approach, COL9A2, COL9A3, NHEJ1, RS1 and NDP genes were investigated based on their known associations with RD and retinal detachment in dogs and humans. In the whole genome approach, affected WGS was compared with 814 unaffected canids to identify candidate variants, which were filtered based on appropriate segregation and predicted pathogenic effects followed by subsequent investigation of gene function. Candidate variants were tested for appropriate segregation in the ECS family and association with disease was assessed using samples from a total of 180 ECS. RESULTS: The same variant in NDP (c.653_654insC, p.Met114Hisfs*16) that was predicted to result in 15 aberrant amino acids before a premature stop in norrin protein, was identified independently by both approaches and was shown to segregate appropriately within the ECS family. Association of this variant with X-linked RD was significant (P = 0.0056). CONCLUSIONS: For the first time, we report a variant associated with canine X-linked RD. NDP variants are already known to cause X-linked RD, along with other abnormalities, in human Norrie disease. Thus, the dog may serve as a useful large animal model for research

    Gender Roles and Women’s Health in India

    One of the main determinants of the poor health status among women in India is a women’s disadvantaged gender role. This study aims to identify the factors that affect women’s health, and particularly focuses on women’s ability to make decisions in a family concerning health-related issues and education. The data are from the India Human Development Survey-II (IHDS-II), 2011-12 (ICPSR 36151), which includes nationally representative samples across India. Women who have more decision making power within a family had better self-reported health. Women who are more educated have better self-reported health. Self-reported women's health status was dependent on the rank of castes to which the women belonged. Our results suggest that decision-making power and education play a key role in improving the health outcomes for women in India. Health promotion programs for women in India should include a platform for gender roles and decision-making capacity. Keywords: women’s health; gender roles; decision-making power; Indi

    Eukaryotic Plankton Communities Across Reef Environments in Bocas del Toro Archipelago, Panamá

    Variation in light and temperature can influence the genetic diversity and structure of marine plankton communities. While open-ocean plankton communities receive much scientific attention, little is known about how environmental variation affects plankton communities on tropical coral reefs. Here, we characterize eukaryotic plankton communities on coral reefs across the Bocas del Toro Archipelago, Panama´. Temperature loggers were deployed, and midday light levels were measured to quantify environmental differences across reefs at four inshore and four offshore sites (Inshore = Punta Donato, Smithsonian Tropical Research Institute (STRI) Point, Cristobal, Punta Laurel and Offshore = Drago Mar, Bastimentos North, Bastimentos South, and Cayo de Agua). Triplicate vertical plankton tows were collected midday, and high-throughput 18S ribosomal DNA metabarcoding was leveraged to investigate the relationship between eukaryotic plankton community structure and inshore/offshore reef environments. Plankton communities from STRI Point were additionally characterized in the morning (* 08:00), midday (* 12:00), and late-day (* 16:00) to quantify temporal variation within a single site. We found that inshore reefs experienced higher average seawater temperatures, while offshore sites offered higher light levels, presumably associated with reduced water turbidity on reefs further from shore. These significant environmental differences between inshore and offshore reefs corresponded with overall plankton community differences. We also found that temporal variation played a structuring role within these plankton communities, and conclude that time of community sampling is an important consideration for future studies. Follow-up studies focusing on more intensive sampling efforts across space and time, coupled with techniques that can detect more subtle genetic differences between and within communities will more fully capture plankton dynamics in this region and beyond
