    Ecological characterization of the Florida springs coast: Pithlachascotee to Waccasassa Rivers

    This report covers the upper coast of west-central Florida. This region includes the drainage basins and nearshore waters of the west coast of Florida between, but not including, the Anclote River basin and the Suwannee River basin. The name Springs Coast wash chosen because this area contains a multitude of springs, both named and too small or inaccessible to have been names. Much of the area is karstic limestone. Most recognizable among the springs are the famous Crystal river, Weeki Wachee, and Homosassa. This territory includes large expanses of marsh and wetland and, along its shores, the southern end of the largest area of seagrass beds in the state -- the Florida Big Bend Seagrass Beds preserve. It also possesses numerous spring-fed rivers and streams along the coast, whose constant discharges provide unique, relatively stable estuarine environments. This document is a summary of the available information on the Springs Coast area of Florida, for use by planners, developers, regulatory authorities, and other interested parties. An understanding of the factors affecting their plans and the possibly unexpected impacts of their actions on others will, it is hoped, promote intelligent development in areas capable of supporting it. We have tried to provide a clear, coherent picture of what is currently known about how the physical, chemical, and biological factors of the environment interact. (343 pp.

    Poor metacognitive awareness of belief change

    When people change beliefs as a result of reading a text, are they aware of these changes? This question was examined for beliefs about spanking as an effective means of discipline. In two experiments, subjects reported beliefs about spanking effectiveness during a prescreening session. In a subsequent experimental session, subjects read a one-sided text that advocated a belief consistent or inconsistent position on the topic. After reading, subjects reported their current beliefs and attempted to recollect their initial beliefs. Subjects reading a belief inconsistent text were more likely to change their beliefs than those who read a belief consistent text. Recollections of initial beliefs tended to be biased in the direction of subjects’ current beliefs. In addition, the relationship between the belief consistency of the text read and accuracy of belief recollections was mediated by belief change. This belief memory bias was independent of on-line text processing and comprehension measures, and indicates poor metacognitive awareness of belief change

    Hydrophilic loop 1 of Presenilin-1 and the APP GxxxG transmembrane motif regulate γ-secretase function in generating Alzheimer-causing Aβ peptides

    γ-Secretase is responsible for the proteolysis of amyloid precursor protein (APP) into amyloid-beta (Aβ) peptides, which are centrally implicated in the pathogenesis of Alzheimer’s disease (AD). The biochemical mechanism of how processing by γ-secretase is regulated, especially as regards the interaction between enzyme and substrate, remains largely unknown. Here, mutagenesis reveals that the hydrophilic loop-1 (HL-1) of presenilin-1 (PS1) is critical for both γ-secretase step-wise cleavages (processivity) and its allosteric modulation by heterocyclic γ-modulatory compounds. Systematic mutagenesis of HL-1, including all of its familial AD mutations and additional engineered variants, and quantification of the resultant Aβ products show that HL-1 is necessary for proper sequential γ-secretase processivity. We identify Y106, L113, and Y115 in HL-1 as key targets for heterocyclic γ-secretase modulators (GSMs) to stimulate processing of pathogenic Aβ peptides. Further, we confirm that the GxxxG domain in the APP transmembrane region functions as a critical substrate motif for γ-secretase processivity: a G29A substitution in APP-C99 mimics the beneficial effects of GSMs. Together, these findings provide a molecular basis for the structural regulation of γ-processivity by enzyme and substrate, facilitating the rational design of new GSMs that lower AD-initiating amyloidogenic Aβ peptides
