    Evaporation of a thin droplet on a thin substrate with a high thermal resistance

    A mathematical model for the quasi-steady evaporation of a thin liquid droplet on a thin substrate that incorporates the dependence of the saturation concentration of vapour at the free surface of the droplet on temperature is used to examine an atypical situation in which the substrate has a high thermal resistance relative to the droplet (i.e. it is highly insulating and/or is thick relative to the droplet). In this situation diffusion of heat through the substrate is the rate-limiting evaporative process and at leading order the local mass flux is spatially uniform, the total evaporation rate is proportional to the surface area of the droplet, and the droplet is uniformly cooled. In particular, the qualitative differences between the predictions of the present model in this situation and those of the widely used 'basic' model in which the saturation concentration is independent of temperature are highlighted

    The verse-epistles of Robert Burns : a critical study

    From the introduction: So vast is the body of published work on Burns that one must justify yet another study of the poet. From 1786 to the present, his life and poetry have always had popular appeal. In his lifetime, he was an object of attention to all classes of society, from Ayrshire peasants to the habitue of Edinburgh drawing-roans, and detractors, idolaters, and disinterested parties have continued to scrutinize his achievements and failings. Popular attention has never wavered. In the nineteenth century especially, many and varied editions of Burns's poetry were published to satisfy this curiosity. Some were lavish, some cheap; some accurate, others, wildly imaginative. Nor has this demand noticeably slackened in the present century. Not a year passes without some book or pamphlet, albeit ephemeral, being published on Burns. To the scholarly mind, "popular", when applied to Burns studies, usually implies superficiality and this assumption all too often proves correct. It can hardly be said that the best minds of each age since Burns's death have considered him worthy of their critical attention in the way that Shakespeare, or Dante, or Milton have engaged scholars, editors, and publishers in succeeding generations. Byron, Coleridge, Hazlitt, Emerson, Carlyle, and Mac=armid have commented on Burns, and in the nineteenth century important and durable editorial work was undertaken. Nevertheless, it remains true that it was not until the twentieth century, and then only in bursts, that there developed a scholarly, academic interest to match the popular enthusiasm for Robert Burns

    PHAGE T4 mobE IS A REGULATED HOMING ENDONUCLEASE GENE THAT IS REQUIRED FOR THE trans HOMING OF THE DEFUNCT HOMING ENDONUCLEASE I- TevIII

    The E. coli bacteriophage T4 has fifteen homing endonuclease gene insertions, five of which are predicted to be free-standing HNH homing endonucleases, named mobA to mobE. The focus of my studies is the mobE homing endonuclease gene found inserted between the conserved aerobic ribonucleotide reductase large (nrdA) and small subunits (nrdB). I showed that mobE is expressed during T4 infection, and a conserved Rho- independent transcriptional terminator regulates mobE expression. I demonstrated that mobE is a functional homing endonuclease gene that introduces a nick on the non-coding strand of the T2 nrdB gene. In T2xT4 mixed infections, mobE is inherited in -91.3% of the progeny and the inheritance was reduced to -72.8% in infections with T4 bearing a mobE internal deletion of the predicted HNH motif. Consequently, MobE activity also causes trans homing of the defunct I-TevIII homing endonuclease gene encoded within an intron interrupting the T2 nrdB gene

    An investigation of thin amorphous carbon-based sputtered coatings for MEMS and micro-engineering applications

    The work presented in this thesis describes an investigation into the production and properties of thin amorphous C films, with and without Cr doping, as a low wear / friction coating applicable to MEMS and other micro- and nano-engineering applications. Firstly, an assessment was made of the available testing techniques. Secondly, the optimised test methods were applied to a series of sputtered films of thickness 10 - 2000 nm in order to: (i) investigate the effect of thickness on the properties of coatingslcoating process (ii) investigate fundamental tribology at the nano-scale and (iii) provide a starting point for nanotribological coating optimisation at ultra low thickness. The use of XPS was investigated for the determination of Sp3/Sp2 carbon bonding. Under C 1s peak analysis, significant errors were identified and this was attributed to the absence of sufficient instrument resolution to guide the component peak structure (even with a high resolution instrument). A simple peak width analysis and correlation work with C KLL D value confirmed the errors. The use of XPS for Sp3/Sp2 was therefore limited to initial tentative estimations. Nanoindentation was shown to provide consistent hardness and reduced modulus results with depth (to < 7nm) when replicate data was suitably statistically processed. No significant pile-up or cracking of the films was identified under nanoindentation. Nanowear experimentation by multiple nanoscratching provided some useful information, however the conditions of test were very different to those expect for MEMS and micro- / nano-engineering systems. A novel 'sample oscillated nanoindentation' system was developed for testing nanowear under more relevant conditions. The films were produced in an industrial production coating line. In order to maximise the available information and to take account of uncontrolled process variation a statistical design of experiment procedure was used to investigate the effect of four key process control parameters. Cr doping was the most significant control parameter at all thicknesses tested and produced a softening effect and thus increased nanowear. Substrate bias voltage was also a significant parameter and produced hardening and a wear reducing effect at all thicknesses tested. The use of a Cr adhesion layer produced beneficial results at 150 nm thickness, but was ineffective at 50 nm. Argon flow to the coating chamber produced a complex effect. All effects reduced significantly with reducing film thickness. Classic fretting wear was produced at low amplitude under nanowear testing. Reciprocating sliding was produced at higher amplitude which generated three body abrasive wear and this was generally consistent with the Archard model. Specific wear rates were very low (typically 10-16 - 10-18 m3N-1m-1). Wear rates reduced exponentially with reduced film thickness and below (approx.) 20 nm, thickness was identified as the most important control of wear

    Sialic Acid Mutarotation Is Catalyzed by the Escherichia coli β-Propeller Protein YjhT

    The acquisition of host-derived sialic acid is an important virulence factor for some bacterial pathogens, but in vivo this sugar acid is sequestered in sialoconjugates as the {alpha}-anomer. In solution, however, sialic acid is present mainly as the β-anomer, formed by a slow spontaneous mutarotation. We studied the Escherichia coli protein YjhT as a member of a family of uncharacterized proteins present in many sialic acid-utilizing pathogens. This protein is able to accelerate the equilibration of the {alpha}- and β-anomers of the sialic acid N-acetylneuraminic acid, thus describing a novel sialic acid mutarotase activity. The structure of this periplasmic protein, solved to 1.5Å resolution, reveals a dimeric 6-bladed unclosed β-propeller, the first of a bacterial Kelch domain protein. Mutagenesis of conserved residues in YjhT demonstrated an important role for Glu-209 and Arg-215 in mutarotase activity. We also present data suggesting that the ability to utilize {alpha}-N-acetylneuraminic acid released from complex sialoconjugates in vivo provides a physiological advantage to bacteria containing YjhT

    Negotiating autobiographical truth: Embodying sensation in the narrative screenplay.

    This article examines how screenwriting adaptations of written material speak of levels of truth-telling within various autobiographical texts. These include literary adaptations by Marguerite Duras: Hiroshima Mon Amour (1959) and The North China Lover (1992), and the autobiographical filmmaking of Maya Deren: Meshes of the Afternoon (1946) and The Very Eye of Night (1958). I argue that descriptions of tactile sensation necessarily remain codified in screenplays, their connotations left hanging even when the filmmaking process often falls short of depicting final truth. What remains is an unresolved problematic perception standing in for an experience. Our own experience of cinema can invariably be one wherein neither words nor images appear, or reappear, as to how they felt for the screenwriter. Is this a wholly negative situation, or merely the continuation of mediation, remediation and the contingent transposition of one medium into another? Drawing on examples from the screenwriting and/or filmmaking of Duras and Deren, I discuss why the screenwriter always writes in personal terms (because the personal is inescapable), and that this is a personal experience of imagination through the writing. Moreover, I test the idea that screenwriting only emerges in a form that we can recognize as truth, through its depictions of tactility and its representations of sensation
