    Amino acid racemization dating of marine shells: a mound of possibilities

    Shell middens are one of the most important and widespread indicators for human exploitation of marine resources and occupation of coastal environments. Establishing an accurate and reliable chronology for these deposits has fundamental implications for understanding the patterns of human evolution and dispersal. This paper explores the potential application of a new methodology of amino acid racemization(AAR) dating of shell middens and describes a simple protocol to test the suitability of different molluscanspecies. This protocol provides a preliminary test for the presence of an intracrystalline fraction of proteins(by bleaching experiments and subsequent heating at high temperature), checking the closed system behaviour of this fraction during diagenesis. Only species which pass both tests can be considered suitable for further studies to obtain reliable age information. This amino acid geochronological technique is also applied to midden deposits at two latitudinal extremes: Northern Scotland and the Southern Red Sea.Results obtained in this study indicate that the application of this new method of AAR dating of shells has the potential to aid the geochronological investigation of shell mounds in different areas of the world

    Knockdown of the schizophrenia susceptibility gene TCF4 alters gene expression and proliferation of progenitor cells from the developing human neocortex

    BACKGROUND: Common variants in the TCF4 gene are among the most robustly supported genetic risk factors for schizophrenia. Rare TCF4 deletions and loss-of-function point mutations cause Pitt-Hopkins syndrome, a developmental disorder associated with severe intellectual disability. METHODS: To explore molecular and cellular mechanisms by which TCF4 perturbation could interfere with human cortical development, we experimentally reduced the endogenous expression of TCF4 in a neural progenitor cell line derived from the developing human cerebral cortex using RNA interference. Effects on genome-wide gene expression were assessed by microarray, followed by Gene Ontology and pathway analysis of differentially expressed genes. We tested for genetic association between the set of differentially expressed genes and schizophrenia using genome-wide association study data from the Psychiatric Genomics Consortium and competitive gene set analysis (MAGMA). Effects on cell proliferation were assessed using high content imaging. RESULTS: Genes that were differentially expressed following TCF4 knockdown were highly enriched for involvement in the cell cycle. There was a nonsignificant trend for genetic association between the differentially expressed gene set and schizophrenia. Consistent with the gene expression data, TCF4 knockdown was associated with reduced proliferation of cortical progenitor cells in vitro. LIMITATIONS: A detailed mechanistic explanation of how TCF4 knockdown alters human neural progenitor cell proliferation is not provided by this study. CONCLUSION: Our data indicate effects of TCF4 perturbation on human cortical progenitor cell proliferation, a process that could contribute to cognitive deficits in individuals with Pitt-Hopkins Syndrome and risk for schizophrenia

    Experiences of minority stress among gay, bisexual, and other men who have sex with men (GBMSM) in Nigeria, Africa: The intersection of mental health, substance use, and HIV sexual risk behavior

    Nigerian gay, bisexual, and other men who have sex with men (GBMSM) experience social marginalisation, discrimination and violence due to their sexual orientation and same-sex attraction, which may affect mental health, substance use, and HIV sexual risk behavior. The goal of the current study was to conduct formative qualitative research to gain better understanding of these issues among GBMSM in Lagos, Nigeria. Face-to-face, semi-structured, in-depth interviews were conducted with 30 GBMSM in Lagos, Nigeria. Data were analysed using a deductive content analysis approach. We found that Nigerian GBMSM experienced both general life stressors as well as proximal and distal sexual minority identity stressors, including rejection by family members, harassment, and physical violence perpetrated by the general public and police officers. Participants described dealing with mental health problems within the context of family rejection, experienced stigma due to sexual orientation, and feelings of social isolation. Substance use was described as occurring within the context of social settings. Lastly, some participants mentioned that they engaged in risky sexual behaviour while under the influence of alcohol and drugs. These findings call for comprehensive and innovative, GBMSM-affirming behavioural healthcare, substance cessation services, and innovative HIV prevention interventions specifically designed and tailored for Nigerian GBMSM

    The NOvA Technical Design Report

    This Technical Design Report (TDR) describes the preliminary design of the NOÎœA accelerator upgrades, NOÎœA detectors, detector halls and detector sites. Compared to the March 2006 and November 2006 NOÎœA Conceptual Design Reports (CDR), critical value engineering studies have been completed and the alternatives still active in the CDR have been narrowed to achieve a preliminary technical design ready for a Critical Decision 2 review. Many aspects of NOÎœA described this TDR are complete to a level far beyond a preliminary design. In particular, the access road to the NOvA Far Detector site in Minnesota has an advanced technical design at a level appropriate for a Critical Decision 3a review. Several components of the accelerator upgrade and new neutrino detectors also have advanced technical designs appropriate for a Critical Decision 3a review

    Observation of Ξb0→Ξc+Ds−\Xi_b^0 \rightarrow \Xi_c^+ D_s^- and Ξb−→Ξc0Ds−\Xi_b^- \rightarrow \Xi_c^0 D_s^- decays

    International audienceThe Ξb0→Ξc+Ds−\Xi_b^0 \rightarrow \Xi_c^+ D_s^- and Ξb−→Ξc0Ds−\Xi_b^- \rightarrow \Xi_c^0 D_s^- decays are observed for the first time using proton-proton collision data collected by the LHCb experiment at a centre-of-mass energy of s=13TeV\sqrt{s}=13\mathrm{TeV}, corresponding to an integrated luminosity of 5.1fb−15.1\mathrm{fb}^{-1}. The relative branching fractions times the beauty-baryon production cross-sections are measured to be \begin{align*} \mathcal{R}\left(\frac{\Xi_b^0}{\Lambda_b^0}\right) \equiv \frac{\sigma\left(\Xi_b^0\right)}{\sigma\left(\Lambda_b^0\right)} \times \frac{\mathcal{B}\left(\Xi_b^0 \rightarrow \Xi_c^+ D_s^-\right)}{\mathcal{B}\left(\Lambda_b^0 \rightarrow \Lambda_c^0 D_s^-\right)} =(15.8\pm1.1\pm0.6\pm7.7)\%, \mathcal{R}\left(\frac{\Xi_b^-}{\Lambda_b^0}\right) \equiv \frac{\sigma\left(\Xi_b^-\right)}{\sigma\left(\Lambda_b^0\right)} \times \frac{\mathcal{B}\left(\Xi_b^- \rightarrow \Xi_c^0 D_s^-\right)}{\mathcal{B}\left(\Lambda_b^0 \rightarrow \Lambda_c^0 D_s^-\right)} =(16.9\pm1.3\pm0.9\pm4.3)\%, \end{align*} where the first uncertainties are statistical, the second systematic, and the third due to the uncertainties on the branching fractions of relevant charm-baryon decays. The masses of Ξb0\Xi_b^0 and Ξb−\Xi_b^- baryons are measured to be mΞb0=5791.12±0.60±0.45±0.24MeV/c2m_{\Xi_b^0}=5791.12\pm0.60\pm0.45\pm0.24\mathrm{MeV}/c^2 and mΞb−=5797.02±0.63±0.49±0.29MeV/c2m_{\Xi_b^-}=5797.02\pm0.63\pm0.49\pm0.29\mathrm{MeV}/c^2, where the uncertainties are statistical, systematic, and those due to charm-hadron masses, respectively

    Digital Interventions for Mental Disorders:Key Features, Efficacy, and Potential for Artificial Intelligence Applications

    Mental disorders are highly prevalent and often remain untreated. Many limitations of conventional face-to-face psychological interventions could potentially be overcome through Internet-based and mobile-based interventions (IMIs). This chapter introduces core features of IMIs, describes areas of application, presents evidence on the efficacy of IMIs as well as potential effect mechanisms, and delineates how Artificial Intelligence combined with IMIs may improve current practices in the prevention and treatment of mental disorders in adults. Meta-analyses of randomized controlled trials clearly show that therapist-guided IMIs can be highly effective for a broad range of mental health problems. Whether the effects of unguided IMIs are also clinically relevant, particularly under routine care conditions, is less clear. First studies on IMIs for the prevention of mental disorders have shown promising results. Despite limitations and challenges, IMIs are increasingly implemented into routine care worldwide. IMIs are also well suited for applications of Artificial Intelligence and Machine Learning, which provides ample opportunities to improve the identification and treatment of mental disorders. Together with methodological innovations, these approaches may also deepen our understanding of how psychological interventions work, and why. Ethical and professional restraints as well as potential contraindications of IMIs, however, should also be considered. In sum, IMIs have a high potential for improving the prevention and treatment of mental health disorders across various indications, settings, and populations. Therefore, implementing IMIs into routine care as both adjunct and alternative to face-to-face treatment is highly desirable. Technological advancements may further enhance the variability and flexibility of IMIs, and thus even further increase their impact in people’s lives in the future

    Study of Bc+→χcπ+B_c^+ \rightarrow \chi_c \pi^+ decays

    International audienceA study of Bc+→χcπ+B_c^+ \rightarrow \chi_c \pi^+ decays is reported using proton-proton collision data, collected with the LHCb detector at centre-of-mass energies of 7, 8, and 13 TeV, corresponding to an integrated luminosity of 9fb−1^{-1}. The decay Bc+→χc2π+B_c^+ \rightarrow \chi_{c2} \pi^+ is observed for the first time, with a significance exceeding seven standard deviations. The relative branching fraction with respect to the Bc+→J/ψπ+B_c^+ \rightarrow J/\psi \pi^+ decay is measured to be BBc+→χc2π+BBc+→J/ψπ+=0.37±0.06±0.02±0.01, \frac{\mathcal{B}_{B_c^+ \rightarrow \chi_{c2} \pi^+}} {\mathcal{B}_{B_c^+ \rightarrow J/\psi \pi^+}} = 0.37 \pm 0.06 \pm 0.02 \pm 0.01 , where the first uncertainty is statistical, the second is systematic, and the third is due to the knowledge of the χc→J/ÏˆÎł\chi_c \rightarrow J/\psi \gamma branching fraction. No significant Bc+→χc1π+B_c^+ \rightarrow \chi_{c1} \pi^+ signal is observed and an upper limit for the relative branching fraction for the Bc+→χc1π+B_c^+ \rightarrow \chi_{c1} \pi^+ and Bc+→χc2π+B_c^+ \rightarrow \chi_{c2} \pi^+ decays of BBc+→χc1π+BBc+→χc2π+<0.49 \frac{\mathcal{B}_{B_c^+ \rightarrow \chi_{c1} \pi^+}} {\mathcal{B}_{B_c^+ \rightarrow \chi_{c2} \pi^+}} < 0.49 is set at the 90% confidence level
