    The BTB/POZ zinc finger protein Broad-Z3 promotes dendritic outgrowth during metamorphic remodeling of the peripheral stretch receptor dbd

    <p>Abstract</p> <p>Background</p> <p>Various members of the family of BTB/POZ zinc-finger transcription factors influence patterns of dendritic branching. One such member, <it>Broad</it>, is notable because its BrZ3 isoform is widely expressed in <it>Drosophila </it>in immature neurons around the time of arbor outgrowth. We used the metamorphic remodeling of an identified sensory neuron, the dorsal bipolar dendrite sensory neuron (dbd), to examine the effects of BrZ3 expression on the extent and pattern of dendrite growth during metamorphosis.</p> <p>Results</p> <p>Using live imaging of dbd in <it>Drosophila </it>pupae, we followed its normal development during metamorphosis and the effect of ectopic expression of BrZ3 on this development. After migration of its cell body, dbd extends a growth-cone that grows between two muscle bands followed by branching and turning back on itself to form a compact dendritic bundle. The ectopic expression of the BrZ3 isoform, using the GAL4/UAS system, caused dbd's dendritic tree to transform from its normal, compact, fasciculated form into a comb-like arbor that spread over on the body wall. Time-lapse analysis revealed that the expression of BrZ3 caused the premature extension of the primary dendrite onto immature myoblasts, ectopic growth past the muscle target region, and subsequent elaboration onto the epidermis. To control the timing of expression of BrZ3, we used a temperature-sensitive GAL80 mutant. When BrZ3 expression was delayed until after the extension of the primary dendrite, then a normal arbor was formed. By contrast, when BrZ3 expression was confined to only the early outgrowth phase, then ectopic arbors were subsequently formed and maintained on the epidermis despite the subsequent absence of BrZ3.</p> <p>Conclusions</p> <p>The adult arbor of dbd is a highly branched arbor whose branches self-fasciculate to form a compact dendritic bundle. The ectopic expression of BrZ3 in this cell causes a premature extension of its growth-cone, resulting in dendrites that extend beyond their normal muscle substrate and onto the epidermis, where they form a comb-shaped, ectopic arbor. Our quantitative data suggest that new ectopic arbor represents an 'unpacking' of the normally fasciculated arbor onto the epidermis. These data suggest that the nature of their local environment can change dendrite behavior from self-adhesion to self-avoidance.</p

    An ESCRT module is required for neuron pruning

    Neural circuits are refined by both functional and structural changes. Structural remodeling by large-scale pruning occurs where relatively long neuronal branches are cut away from their parent neuron and removed by local degeneration. Until now, the molecular mechanisms executing such branch severing events have remained poorly understood. Here, we reveal a role for the Endosomal Sorting Complex Required for Transport (ESCRT) machinery during neuronal remodeling. Our data show that a specific ESCRT pruning module, including members of the ESCRT-I and ESCRT-III complexes, but not ESCRT-0 or ESCRT-II, are required for the neurite scission event during pruning. Furthermore we show that this ESCRT module requires a direct, in vivo, interaction between Shrub/CHMP4B and the accessory protein Myopic/HD-PTP

    Developmental organization of central neurons in the adult Drosophila ventral nervous system

    We have used MARCM to reveal the adult morphology of the post embryonically produced neurons in the thoracic neuromeres of the Drosophila VNS. The work builds on previous studies of the origins of the adult VNS neurons to describe the clonal organization of the adult VNS. We present data for 58 of 66 postembryonic thoracic lineages, excluding the motor neuron producing lineages (15 and 24) which have been described elsewhere. MARCM labels entire lineages but where both A and B hemilineages survive (e.g., lineages 19, 12, 13, 6, 1, 3, 8, and 11), the two hemilineages can be discriminated and we have described each hemilineage separately. Hemilineage morphology is described in relation to the known functional domains of the VNS neuropil and based on the anatomy we are able to assign broad functional roles for each hemilineage. The data show that in a thoracic hemineuromere, 16 hemilineages are primarily involved in controlling leg movements and walking, 9 are involved in the control of wing movements, and 10 interface between both leg and wing control. The data provide a baseline of understanding of the functional organization of the adult Drosophila VNS. By understanding the morphological organization of these neurons, we can begin to define and test the rules by which neuronal circuits are assembled during development and understand the functional logic and evolution of neuronal networks.</p

    Fault diagnosis of a rotor-bearing system under variable rotating speeds using two-stage parameter transfer and infrared thermal images

    Current fault diagnosis methods for rotor-bearing system are mostly based on analyzing the vibration signals collected at steady rotating speeds. In those methods, the data collected under one operating condition cannot be accurately used for diagnosis under a different condition. Moreover, in vibration monitoring, installing the necessary sensors will affect the equipment structure and hence the vibration response itself. The present paper proposes a new method based on two-stage parameter transfer and infrared thermal images for fault diagnosis of rotor-bearing system under variable rotating speeds. The method of parameter transfer enables the use of data (or parameters) acquired under one operating condition (called the source domain) to be extended for use in a different operating condition (called the target domain). First, scaled exponential linear unit (SELU) and modified stochastic gradient descent (MSGD) are used to construct an enhanced convolutional neural network (ECNN). Second, a stacked convolutional auto-encoder (CAE) trained based on unlabeled source-domain thermal images is employed to initialize a source-domain ECNN. Third, model parameters from the pre-trained source-domain ECNN are transferred to the target-domain ECNN to adapt to the characteristics of the target domain. The collected thermal images for a rotor-bearing system under variable speeds are used to test the transfer diagnosis performance of the proposed method. The experimental results demonstrate the performance improvement and the advantages of the proposed method

    Utilising light-emitting diodes of specific narrow wavelengths for the optimization and co-production of multiple high-value compounds in Porphyridium purpureum

    The effect of specific narrow light-emitting diode (LED) wavelengths (red, green, blue) and a combination of LED wavelengths (red, green and blue - RGB) on biomass composition produced by Porphyridium purpureum is studied. Phycobiliprotein, fatty acids, exopolysaccharides, pigment content, and the main macromolecules composition were analysed to determine the effect of wavelength on multiple compounds of commercial interest. The results demonstrate that green light plays a significant role in the growth of rhodophyta, due to phycobiliproteins being able to harvest green wavelengths where chlorophyll pigments absorb poorly. However, under multi-chromatic LED wavelengths, P. purpureum biomass accumulated the highest yield of valuable products such as eicosapentaenoic acid (~2.9 %DW), zeaxanthin (~586 μg g− 1 DW), β-carotene (397 μg g− 1 DW), exopolysaccharides (2.05 g/L-1), and phycobiliproteins (~ 4.8 % DW). This increased accumulation is likely to be the combination of both photo-adaption and photo-protection, under the combined specific wavelengths employed

    Femoroacetabular impingement and classification of the cam deformity: the reference interval in normal hips

    BACKGROUND AND PURPOSE: Most patients with femoroacetabular impingement (FAI) have a cam deformity, which may be quantified by measuring the alpha angle and anterior offset ratio (AOR). Knowledge of what constitutes a "normal" alpha angle and AOR is limited. We defined the reference intervals of these measurements from normal hips in the general population. PATIENTS AND METHODS: 157 individuals from the general population were reviewed clinically and radiographically. 74 individuals with clinical evidence of hip disease or radiographic evidence of osteoarthritis (OA) were excluded, leaving a study group of 83 individuals (mean age 46 (22-69) years, 44 females) with normal hips. The alpha angles and AORs were measured from cross-table lateral radiographs taken in 15 degrees internal rotation. A validation study consisting of a cadaver study and a measurement reliability study was also performed. RESULTS: The mean alpha angle was 48 degrees in men and 47 degrees in women. The mean AOR was 0.19, the same in men and women. Thus, sexes were combined to derive 95% confidence intervals for the population mean alpha angle (46-49 degrees ) and AOR (0.18-0.20). The 95% reference interval for the alpha angle was 32-62 degrees degrees, and for the AOR it was 0.14-0.24. The validation study confirmed that these measurements were resistant to a reasonable degree of variation in positioning and that the repeatability and reproducibility of the measurements was good. INTERPRETATION: These reference intervals indicate that clinically and radiographically normal hips may have alpha angles and AORs that have previously been considered "abnormal". The thresholds provided by this study will aid classification of individuals involved in longitudinal studies of FAI and OA, and may be of use to the practicing clinician in evaluating the young adult with hip pain
