    A Comparison of Vertical Ground Reaction Forces in Single-leg Jump Take-offs and Double-leg Jump Take-offs in Indoor Collegiate Volleyball Players

    The purpose of this study was to determine if differences in vertical ground reaction forces (vGRFs) exist between single-leg take-off approaches (SL) and double-leg take-off approaches (DL) and if sex or positional differences existed in vGRFs in SL- or DL-style approaches. It was hypothesized that DL jumps would generate greater vGRFs, males would generate greater vGRFs in both styles, and middle blockers would have higher SL vGRFs while pin hitters would have higher DL vGRFs. Nine collegiate volleyball players (4 female; 5 male) (age: 20.44 Β± 1.42 years; height: 186.02 Β± 6.44 cm; body mass: 78.21 Β± 10.63kg; body fat: 11.66 Β±5.62%) were tested. An AMTI force plate collected vGRFs with three recorded trials per approach style per subject with randomization of jump-style order. The level of significance was set at p\u3c 0.05. No significant differences were found between vGRFs and position or SL and DL vGRFs. Males had significantly higher vGRFs overall compared to females (p=0.007). Male SL vGRFs were significantly larger than females (p=0.017). DL, relative SL, and relative DL vGRFs were not significantly different between genders. Height, mass, and body fat percentage had at least moderate correlations to combined vGRFs, and height moderately correlated to DL vGRFs. These results suggest that both takeoffs have similar enough force load to be interchangeable. Strength coaches may use both styles to train their athletes and improve neural integration. Positional jump loads during practice or games may be tracked interchangeably in style regarding the take-off phase

    Laser desorption/ionization coupled to FT-ICR mass spectrometry for studies of natural organic matter

    Laser desorption/ionization (LDI) was investigated as an ionization method for Fourier transform ion cyclotron resonance mass spectrometry (FTICR MS) studies of natural organic matter (NOM). Using International Humic Substances Society standards, Suwannee River fulvic acid (SRFA) and Suwannee River natural organic matter (SRNOM), LDI was found to ionize a very similar set of compounds (>90% of molecular formulas identity) to the matrix assisted laser desorption/ionization (MALDI), while producing higher quality spectra. A comparison of electrospray ionization (ESI) and LDI spectra showed that different types of compounds are ionized by these methods with only 9.9% of molecular formulas common to both. The compounds ionized by LDI/MALDI belong to low oxygen classes (maximum number of species for O7–O9), while ESI compounds belong to higher oxygen classes (maximum number of species for O14–O16). Compounds ionized by LDI can be classified as aliphatic, aromatic, and condensed aromatics in approximately equal measure, while aliphatic compounds dominated the ESI spectra of SRFA. In order to maximize the coverage of molecular species, LDI, as a particularly convenient and readily deployable ionization method, should be used routinely in combination with other ionization methods, such as ESI, for FTICR MS studies of NOM

    Diffuse Optical Tomography Activation in the Somatosensory Cortex: Specific Activation by Painful vs. Non-Painful Thermal Stimuli

    Background: Pain is difficult to assess due to the subjective nature of self-reporting. The lack of objective measures of pain has hampered the development of new treatments as well as the evaluation of current ones. Functional MRI studies of pain have begun to delineate potential brain response signatures that could be used as objective read-outs of pain. Using Diffuse Optical Tomography (DOT), we have shown in the past a distinct DOT signal over the somatosensory cortex to a noxious heat stimulus that could be distinguished from the signal elicited by innocuous mechanical stimuli. Here we further our findings by studying the response to thermal innocuous and noxious stimuli. Methodology/Principal Findings: Innocuous and noxious thermal stimuli were applied to the skin of the face of the first division (ophthalmic) of the trigeminal nerve in healthy volunteers (Nβ€Š=β€Š6). Stimuli temperatures were adjusted for each subject to evoke warm (equivalent to a 3/10) and painful hot (7/10) sensations in a verbal rating scale (0/10β€Š=β€Šno/max pain). A set of 26 stimuli (5 sec each) was applied for each temperature with inter-stimulus intervals varied between 8 and 15 sec using a Peltier thermode. A DOT system was used to capture cortical responses on both sides of the head over the primary somatosensory cortical region (S1). For the innocuous stimuli, group results indicated mainly activation on the contralateral side with a weak ipsilateral response. For the noxious stimuli, bilateral activation was observed with comparable amplitudes on both sides. Furthermore, noxious stimuli produced a temporal biphasic response while innocuous stimuli produced a monophasic response. Conclusions/Significance: These results are in accordance with fMRI and our other DOT studies of innocuous mechanical and noxious heat stimuli. The data indicate the differentiation of DOT cortical responses for pain vs. innocuous stimuli that may be useful in assessing objectively acute pain

    Look Twice: A Computational Model of Return Fixations across Tasks and Species

    Saccadic eye movements allow animals to bring different parts of an image into high-resolution. During free viewing, inhibition of return incentivizes exploration by discouraging previously visited locations. Despite this inhibition, here we show that subjects make frequent return fixations. We systematically studied a total of 44,328 return fixations out of 217,440 fixations across different tasks, in monkeys and humans, and in static images or egocentric videos. The ubiquitous return fixations were consistent across subjects, tended to occur within short offsets, and were characterized by longer duration than non-return fixations. The locations of return fixations corresponded to image areas of higher saliency and higher similarity to the sought target during visual search tasks. We propose a biologically-inspired computational model that capitalizes on a deep convolutional neural network for object recognition to predict a sequence of fixations. Given an input image, the model computes four maps that constrain the location of the next saccade: a saliency map, a target similarity map, a saccade size map, and a memory map. The model exhibits frequent return fixations and approximates the properties of return fixations across tasks and species. The model provides initial steps towards capturing the trade-off between exploitation of informative image locations combined with exploration of novel image locations during scene viewing

    SWI/SNF-like chromatin remodeling factor Fun30 supports point centromere function in S. cerevisiae

    Budding yeast centromeres are sequence-defined point centromeres and are, unlike in many other organisms, not embedded in heterochromatin. Here we show that Fun30, a poorly understood SWI/SNF-like chromatin remodeling factor conserved in humans, promotes point centromere function through the formation of correct chromatin architecture at centromeres. Our determination of the genome-wide binding and nucleosome positioning properties of Fun30 shows that this enzyme is consistently enriched over centromeres and that a majority of CENs show Fun30-dependent changes in flanking nucleosome position and/or CEN core micrococcal nuclease accessibility. Fun30 deletion leads to defects in histone variant Htz1 occupancy genome-wide, including at and around most centromeres. FUN30 genetically interacts with CSE4, coding for the centromere-specific variant of histone H3, and counteracts the detrimental effect of transcription through centromeres on chromosome segregation and suppresses transcriptional noise over centromere CEN3. Previous work has shown a requirement for fission yeast and mammalian homologs of Fun30 in heterochromatin assembly. As centromeres in budding yeast are not embedded in heterochromatin, our findings indicate a direct role of Fun30 in centromere chromatin by promoting correct chromatin architecture
