    Serological detection of infection with canine distemper virus, canine parvovirus and canine adenovirus in communal dogs from Zimbabwe

    Domestic dogs are common amongst communities in sub-Saharan Africa and may serve as important reservoirs for infectious agents that may cause diseases in wildlife. Two agents of concern are canine parvovirus (CPV) and canine distemper virus (CDV), which may infect and cause disease in large carnivore species such as African wild dogs and African lions, respectively. The impact of domestic dogs and their diseases on wildlife conservation is increasing in Zimbabwe, necessitating thorough assessment and implementation of control measures. In this study, domestic dogs in north-western Zimbabwe were evaluated for antibodies to CDV, CPV, and canine adenovirus (CAV). These dogs were communal and had no vaccination history. Two hundred and twenty-five blood samples were collected and tested using a commercial enzyme-linked immunosorbent assay (ELISA) for antibodies to CPV, CDV, and CAV. Of these dogs, 75 (34%) had detectable antibodies to CDV, whilst 191 (84%) had antibodies to CPV. Antibodies to canine adenovirus were present in 28 (13%) dogs. Canine parvovirus had high prevalence in all six geographic areas tested. These results indicate that CPV is circulating widely amongst domestic dogs in the region. In addition, CDV is present at high levels. Both pathogens can infect wildlife species. Efforts for conservation of large carnivores in Zimbabwe must address the role of domestic dogs in disease transmission

    Fifteen Years of Chandra Operation: Scientific Highlights and Lessons Learned

    NASA's Chandra X-Ray Observatory, designed for three years of operation with a goal of five years is now entering its 15-th year of operation. Thanks to its superb angular resolution, the Observatory continues to yield new and exciting results, many of which were totally unanticipated prior to launch. We discuss the current technical status, review recent scientific highlights, indicate a few future directions, and present what we feel is the most important lesson learned from our experience of building and operating this great observatory

    A meta-analysis of protein binding of flucloxacillin in healthy volunteers and hospitalized patients

    Objectives: The aim of this study was to develop a mechanistic protein-binding model to predict the unbound flucloxacillin concentrations in different patient populations. Methods: A mechanistic protein-binding model was fitted to the data using non-linear mixed-effects modelling. Data were obtained from four datasets, containing 710 paired total and unbound flucloxacillin concentrations from healthy volunteers, non-critically ill and critically ill patients. A fifth dataset with data from hospitalized patients was used for evaluation of our model. The predictive performance of the mechanistic model was evaluated and compared with the calculation of the unbound concentration with a fixed unbound fraction of 5%. Finally, we performed a fit-for-use evaluation, verifying whether the model-predicted unbound flucloxacillin concentrations would lead to clinically incorrect dose adjustments. Results: The mechanistic protein-binding model predicted the unbound flucloxacillin concentrations more accurately than assuming an unbound fraction of 5%. The mean prediction error varied between -26.2% to 27.8% for the mechanistic model and between -30.8% to 83% for calculation with a fixed factor of 5%. The normalized root mean squared error varied between 36.8% and 69% respectively between 57.1% and 134%. Predicting the unbound concentration with the use of the mechanistic model resulted in 6.1% incorrect dose adjustments versus 19.4% if calculated with a fixed unbound fraction of 5%. Conclusions: Estimating the unbound concentration with a mechanistic protein-binding model outperforms the calculation with the use of a fixed protein binding factor of 5%, but neither demonstrates acceptable performance. When performing dose individualization of flucloxacillin, this should be done based on measured unbound concentrations rather than on estimated unbound concentrations from the measured total concentrations. In the absence of an assay for unbound concentrations, the mechanistic binding model should be preferred over assuming a fixed unbound fraction of 5%

    Dynamics of Broad-Emission Line Region in NGC 5548: Hydromagnetic Wind Model versus Observations

    We analyze the results of long-term observations of broad-line region (BLR) in the Sy1 galaxy NGC 5548 and provide a critical comparison with the predictions of a hydromagnetically-driven outflow model of Emmering, Blandford and Shlosman. This model was used to generate a time series of CIV line profiles that have responded to a time varying continuum. The model includes cloud emission anisotropy, cloud obscuration, a CLOUDY-generated emissivity function and a narrow-line component used to generate the line profiles, and is driven with continuum input based on the monitoring campaigns of NGC 5548. The line strengths, profiles and lags are compared with the observations. It reproduces the basic features of CIV line variability in this AGN without varying the model parameters. The best fit model provides the effective size, the dominant geometry, the emissivity distribution and the 3D velocity field of the CIV BLR and constrains the mass of the central BH. The inner part of the wind appears to be responsible for the anisotropically emitted CIV line, while its outer part remains dusty and molecular, thus having similar spectral characteristics to a molecular torus, although its dynamics is fundamentally different. In addition, our model predicts a differential response across the CIV line profile, producing a red-side-first response in the mid-wings followed by the blue mid-wing and by the line core. Based on the comparison of data and model cross-correlation functions and 1D and 2D transfer functions, we find that the rotating outflow model is compatible with observations of the BLR in NGC 5548.Comment: 50 pages, TeX, 14 figures.     Effect of Myostatin Depletion on Weight Gain, Hyperglycemia, and Hepatic Steatosis during Five Months of High-Fat Feeding in Mice

    The marked hypermuscularity in mice with constitutive myostatin deficiency reduces fat accumulation and hyperglycemia induced by high-fat feeding, but it is unclear whether the smaller increase in muscle mass caused by postdevelopmental loss of myostatin activity has beneficial metabolic effects during high-fat feeding. We therefore examined how postdevelopmental myostatin knockout influenced effects of high-fat feeding. Male mice with ubiquitous expression of tamoxifen-inducible Cre recombinase were fed tamoxifen for 2 weeks at 4 months of age. This depleted myostatin in mice with floxed myostatin genes, but not in control mice with normal myostatin genes. Some mice were fed a high-fat diet (60% of energy) for 22 weeks, starting 2 weeks after cessation of tamoxifen feeding. Myostatin depletion increased skeletal muscle mass āˆ¼30%. Hypermuscular mice had āˆ¼50% less weight gain than control mice over the first 8 weeks of high-fat feeding. During the subsequent 3 months of high-fat feeding, additional weight gain was similar in control and myostatin-deficient mice. After 5 months of high-fat feeding, the mass of epididymal and retroperitoneal fat pads was similar in control and myostatin-deficient mice even though myostatin depletion reduced the weight gain attributable to the high-fat diet (mean weight with high-fat diet minus mean weight with low-fat diet: 19.9 g in control mice, 14.1 g in myostatin-deficient mice). Myostatin depletion did not alter fasting blood glucose levels after 3 or 5 months of high-fat feeding, but reduced glucose levels measured 90 min after intraperitoneal glucose injection. Myostatin depletion also attenuated hepatic steatosis and accumulation of fat in muscle tissue. We conclude that blocking myostatin signaling after maturity can attenuate some of the adverse effects of a high-fat diet

    Macronuclear Genome Sequence of the Ciliate Tetrahymena thermophila, a Model Eukaryote

    The ciliate Tetrahymena thermophila is a model organism for molecular and cellular biology. Like other ciliates, this species has separate germline and soma functions that are embodied by distinct nuclei within a single cell. The germline-like micronucleus (MIC) has its genome held in reserve for sexual reproduction. The soma-like macronucleus (MAC), which possesses a genome processed from that of the MIC, is the center of gene expression and does not directly contribute DNA to sexual progeny. We report here the shotgun sequencing, assembly, and analysis of the MAC genome of T. thermophila, which is approximately 104 Mb in length and composed of approximately 225 chromosomes. Overall, the gene set is robust, with more than 27,000 predicted protein-coding genes, 15,000 of which have strong matches to genes in other organisms. The functional diversity encoded by these genes is substantial and reflects the complexity of processes required for a free-living, predatory, single-celled organism. This is highlighted by the abundance of lineage-specific duplications of genes with predicted roles in sensing and responding to environmental conditions (e.g., kinases), using diverse resources (e.g., proteases and transporters), and generating structural complexity (e.g., kinesins and dyneins). In contrast to the other lineages of alveolates (apicomplexans and dinoflagellates), no compelling evidence could be found for plastid-derived genes in the genome. UGA, the only T. thermophila stop codon, is used in some genes to encode selenocysteine, thus making this organism the first known with the potential to translate all 64 codons in nuclear genes into amino acids. We present genomic evidence supporting the hypothesis that the excision of DNA from the MIC to generate the MAC specifically targets foreign DNA as a form of genome self-defense. The combination of the genome sequence, the functional diversity encoded therein, and the presence of some pathways missing from other model organisms makes T. thermophila an ideal model for functional genomic studies to address biological, biomedical, and biotechnological questions of fundamental importance

Recent studies have shown that disturbed sleep at altitude, one of the five symptoms scored for AMS, is more likely due to altitude hypoxia per se, and is not closely related to AMS. To address this issue, and also to evaluate the Lake Louise AMS score in light of decades of experience, experts in high altitude research undertook to revise the score. We here present an international consensus statement resulting from online discussions and meetings at the International Society of Mountain Medicine World Congress in Bolzano, Italy, in May 2014 and at the International Hypoxia Symposium in Lake Louise, Canada, in February 2015. The consensus group has revised the score to eliminate disturbed sleep as a questionnaire item, and has updated instructions for use of the score

    B.FLT.1005: Survey of Australian feedlot drinking water quality

    High concentrations of Total Dissolved Solids (TDS) and salts in drinking water, often exacerbated by drought conditions, have been reported to have a detrimental impact on cattle health and performance. This research investigated water quality parameters including TDS and salts (sodium, calcium, magnesium, potassium, aluminium, zinc, iron, manganese, carbonate, bicarbonate, sulphate, nitrate and nitrite). The project summarised the range of TDS and salts in feedlot drinking water across Australia, thus determining potential impacts on cattle health and production. Additionally, feedlots using surface water sources for drinking water had the cyanobacteria species present identified and quantified. Such a study has not previously occurred in Australia. The study was split into four parts with this report including over 100 historical water samples taken prior to 2018; 68 feedlot managers/owners completed the survey regarding water use delivered in paper and online format; 82 water samples from April to June 2019 from 58 feedlots were analysed for a suite of parameters. Where multiple water sources were used, the source water was analysed, this occurred for 24 samples. Additionally, water samples that included surface water were analysed for cyanobacteria (27 samples). Feedlots participating in the project were equivalent to over 50% of the licenced cattle feedlots. Of the 68 feedlots completing the paper based and online survey, the majority (64%) source their cattle drinking water from groundwater. On a per-head basis, the use of groundwater as a drinking water source became even more important, covering two thirds of surveyed feedlot cattle. Surface water from dams and rivers were also a common source of water, and less common sources included reverse osmosis treated water from coal seam gas operations, tank roof water, and irrigation water. The majority of feedlots (75%) were not aware of any issues with their drinking water quality. Of those that indicated that they had concerns about their water quality, seven feedlots identified cyanobacteria (Blue green algae) and Escherichia coli as an issue; four identified turbidity and scale (likely from calcium build up) clogging floats; and four feedlots reported that they now treat water for use in their boiler, but do not treat for cattle, while one feedlot identified high iron as an issue for boiler water. Among trough water samples analysed for TDS, the majority (86 %) were considered satisfactory for cattle consumption and would not be expected to limit animal performance (ā‰¤ 3,000 mg/L). There were, however, cases of poor water quality identified. The highest TDS reported was 11,600 mg/L in groundwater. This water was shandied with surface water and was the maximum in the mixed trough water (5,400 mg/L), which would be expected to limit cattle performance (NASEM, 2016). Chloride was present in the highest concentration of all anions analysed. Nitrate concentrations were highest in the groundwater samples with only one trough sample exceeding the nitrate concentration threshold of 20 mg/L (NASEM, 2016). Sulphate ranged from undetected ( Several water treatment scenarios were investigated with distributors and installers in Australia with reverse osmosis being the most suitable treatment option. As there are no Australian references for the effect of water quality on the performance of the cattle, Patterson et al. (2004), a publication from the USA, was used for the analysis of benefit and cost of water treatment with reverse osmosis. Treatment with reverse osmosis lead to increased, and more cost-effective, cattle productivity. However, the water used in the article by Patterson et al. (2004) had high sulphate concentrations, so the same responses are unlikely with Australian water. Future research testing water quality in the range of variation experienced by Australian feedlots, in a controlled manner, would allow the industry to determine the most relevant animal production gains and thus the benefits of reverse osmosis. In conclusion, water quality was determined to be of suitable quality for the majority of feedlots surveyed. Isolated cases of poor water quality were identified. This project is beneficial to the industry as it has yielded a comprehensive understanding of the current sources and quality of feedlot drinking water for a single point in time. Overall, this project will improve feedlot decision-making regarding the conditions when water quality parameters may impact animal health and production in beef cattle feedlots
